Pressure-Triggered Dystonia in a 35-Year-Old Woman
A 35-year-old woman presented with a 23-year history of paroxysmal dystonic episodes reproducibly triggered by gentle pressure on the right upper limb. Episodes began at age 12 years and gradually worsened, interfering with daily activities. Attacks occurred 2 to 3 seconds after stimulation, with increased muscle tone and abnormal posturing of the right face and limb, including facial stiffening, arm flexion, and leg extension, lasting about 10 to 15 seconds (Video, segment 1).

Repression of EGFR by new biguanide 4C potentiated ovarian cancer to PARP inhibitors through down-regulation of BRCA2 and Rad51
Abstract EGFR, one of the most successful therapeutic targets, has recently been found to exert a novel function for regulating homologous recombination (HR). Activation of HR is the critical event of treatment failure of PARPi in BRCA1/2 wild-type ovarian cancer (OC). Besides, the antitumor effects of biguanides have also been a focus of attention. Here, we discovered that the new biguanide 4C inhibited HR and sensitized BRCA1/2 wild-type OC cells to PARPi by targeting EGFR.

Frontiers | Vericiguat attenuates cyclosporine A-induced nephropathy by targeting the NF-κB/TGF-β1 axis: an integrated network pharmacology, Mendelian randomization, and experimental study
1 Introduction Chronic kidney disease (CKD) represents a pressing global health burden, affecting over 800 million individuals and contributing substantially to worldwide morbidity and mortality (1). Etiologies are varied, encompassing diabetes, hypertension, and, importantly, drug-induced nephrotoxicity (2–5). Cyclosporine A (CsA), a calcineurin inhibitor fundamental to organ transplantation and autoimmune disease management, epitomizes this iatrogenic concern (6).

Peri‐ictal yawning: A potential lateralizing sign in temporal lobe epilepsy
Peri-ictal yawning was an infrequent phenomenon in temporal lobe epilepsy (TLE). It was more commonly observed in patients with non-dominant TLE rather than those with dominant TLE. PY was predominantly associated with the late ictal and postictal phases. It may serve as one of the uncommon vegetative signs in patients with TLE, potentially indicating lateralization to the non-dominant hemisphere.

Targeting TRIP13 in favorable histology Wilms tumor with nuclear export inhibitors synergizes with doxorubicin - Communications Biology
|
Abstract Wilms tumor (WT) is the most common renal malignancy of childhood. Despite improvements in the overall survival, relapse occurs in ~15% of patients with favorable histology WT (FHWT). Half of these patients will succumb to their disease. Identifying novel targeted therapies remains challenging in part due to the lack of faithful preclinical in vitro models.

Comprehensive mutational scanning of EGFR reveals TKI sensitivities of extracellular domain mutants - Nature Communications
Abstract The epidermal growth factor receptor, EGFR, is frequently activated in lung cancer and glioblastoma by genomic alterations including missense mutations. The different mutation spectra in these diseases are reflected in divergent responses to EGFR inhibition: significant patient benefit in lung cancer, but limited in glioblastoma. Here, we report a comprehensive mutational analysis of EGFR function.

Potato Malformation Identification and Classification Based on Improved YOLOv3 Algorithm
1. Introduction Potato is the staple food for most developing countries worldwide and thus has great significance for food security, poverty reduction, and wealth accumulation [1]. However, the market value of the malformed potato is nearly nonexistent, and mixed sales seriously affect overall income. Therefore, the removal of malformed potatoes has become one of the core processes of post-harvest and pre-sales treatment.

East Gobi megalake systems reveal East Asian Monsoon dynamics over the last interglacial-glacial cycle - Nature Communications
Abstract Intense debate persists about the timing and magnitude of the wet phases in the East Asia deserts since the late Pleistocene. Here we show reconstructions of the paleohydrology of the East Gobi Desert since the last interglacial using satellite images and digital elevation models (DEM) combined with detailed section analyses. Paleolakes with a total area of 15,500 km2 during Marine Isotope Stage 5 (MIS 5) were identified.

Structure–function analysis of the SHOC2–MRAS–PP1C holophosphatase complex - Nature
Abstract Receptor tyrosine kinase (RTK)–RAS signalling through the downstream mitogen-activated protein kinase (MAPK) cascade regulates cell proliferation and survival. The SHOC2–MRAS–PP1C holophosphatase complex functions as a key regulator of RTK–RAS signalling by removing an inhibitory phosphorylation event on the RAF family of proteins to potentiate MAPK signalling1.
Allosteric inhibition of PPM1D serine/threonine phosphatase via an altered conformational state - Nature Communications
Abstract PPM1D encodes a serine/threonine phosphatase that regulates numerous pathways including the DNA damage response and p53. Activating mutations and amplification of PPM1D are found across numerous cancer types. GSK2830371 is a potent and selective allosteric inhibitor of PPM1D, but its mechanism of binding and inhibition of catalytic activity are unknown. Here we use computational, biochemical and functional genetic studies to elucidate the molecular basis of GSK2830371 activity.

Fast evolution of SARS-CoV-2 driven by deamination systems in hosts
Cytidine-to-uridine deamination is overrepresented in SARS-CoV-2 compared with other organisms Viruses are exposed to the cellular environment after infecting the cells. As many recent literatures revealed, SARS-CoV-2 is subjected to the hosts’ deamination systems, which include the cytidine-to-uridine (C-to-U) deamination by APOBECs [1,2] and the adenosine-to-inosine (A-to-I) deamination by ADARs [3]. As I is equivalent to G, both C-to-U and A-to-I deamination would cause nucleotide substitution.
