Impact of the clonal architecture and probable biallelic inactivation of TET2 mutations in myelodysplastic neoplasms
To the Editor: TET2 mutations are among the most frequent genetic alterations in myelodysplastic neoplasms (MDS), but their prognostic significance remains controversial [1,2,3]. Previous studies suggest TET2 acquisition timing may shape clinical phenotypes and clonal evolution [4, 5], whereas biallelic TET2 inactivation has been associated with monocytosis, chronic myelomonocytic leukemia-like biology [6, 7], and possibly a distinct MDS subgroup enriched for splicing factor co-mutations [8].

Ectopic expression of the transcription factor ONECUT3 drives a complex karyotype in myelodysplastic syndromes
Abstract Chromosomal instability is a prominent biological feature of myelodysplastic syndromes (MDS), with over 50% of patients with MDS harboring chromosomal abnormalities or a complex karyotype (CK). Despite this observation, the mechanisms underlying mitotic and chromosomal defects in MDS remain elusive. In this study, we identified ectopic expression of the transcription factor ONECUT3, which is associated with CKs and poorer survival outcomes in MDS.
