RACK1 maintains mouse hematopoietic stem cells by directly binding to and stabilizing LDB1
Abstract A core complex with transcription factors (TFs) TAL1/TCF3/GATA2 and adaptors LMO2/LDB1 lies at the top of the hematopoietic transcriptional hierarchy. The mechanism(s) underlying the expression of these components remain elusive. Adaptor RACK1 interacts with multiple TFs and modulates their activation and/or stability. However, a role of RACK1 in the transcriptional control of hematopoietic stem cell (HSC) fates hasn’t been disclosed.

Epiblast diversification and blood formation in a human pregastrula
Abstract The incipient stage of gastrulation in human, when the primitive streak is about to emerge, represents a critical yet underexplored period. Here we present the high-resolution spatial transcriptomic landscape of a human embryo at Carnegie stage 6 (approximately 13–14 days post-conception), a stage at which primitive streak remains invisible and gastrulation-derived mesodermal/endodermal progenitors are not yet transcriptomically detected.
Targeting MEF2A suppresses microglial hyperactivation and synaptic phagocytosis to attenuate epilepsy pathogenesis
Abstract Microglia’s role in epilepsy through neuroimmune communication is poorly understood. Mechanisms by which neurons activate microglia and how microglia affect neuronal activity to drive seizure-related inflammation remain unclear. Here, we elucidated a crucial axis connecting pathological adenosine triphosphate (ATP) release induced by epileptiform neuronal activity to microglial MEF2A-dependent hyperactivation, which exacerbates epilepsy pathology.

Asymmetric synthesis of enantioenriched lactams from cyclic ketones via Beckmann rearrangement - Nature Chemistry
Abstract The direct incorporation of a single nitrogen atom into ketones or aldehydes represents one of the most valuable transformations for constructing amides and lactams, motifs that are ubiquitous in pharmaceutical manufacturing and industrial synthesis. Despite its importance, achieving precise stereochemical control in this process remains a formidable and long-standing challenge, primarily owing to the demanding reaction conditions typically required for such transformations.
Photoinduced polarity-mismatched transformations of isoquinolines into naphthalenes - Nature Communications
Abstract Isosteric replacement is an important strategy for lead optimization in drug-development paradigms. However, it is usually restricted to cost- and labor-intensive de novo syntheses. We here report a photochemical photosensitizer-free skeletal-editing strategy to directly transform easily accessible isoquinolines into synthetically challenging naphthalene derivatives in a single operation.
Evolution of manganese low-energy photoredox catalysis from high-energy visible light photocatalysis - Nature Communications
Abstract Reducing the energy input of light in photocatalytic reactions is desirable yet challenging, as modifying the light-absorbing or catalytic properties of a photocatalyst typically requires tedious preparation, resulting in a lengthy development process. Here, we overcome this limitation by achieving manganese-based low-energy photoredox catalysis through the in-situ assembly of simple Mn salts with inexpensive coordinating chemicals, thereby bypassing the need for complex pre-preparation.

Abstract The Mannich reaction represents one of the most fundamental reactions for the stereoselective synthesis of β-amino-carbonyl compounds and has been broadly applied in organic synthesis. In contrast, the homologue of the Mannich reaction, the so-called the homo-Mannich reaction, which generates the homologated γ-amino-carbonyl products, remains largely underexplored despite its potential for diverse applications.
Divergent access to E- or Z-trisubstituted medium-sized cycloalkenes by Pd-catalysed cycloaddition - Nature Chemistry
Abstract Despite their significance in nature, medium-sized rings (particularly 9–11 membered ones) are non-existent in commercial small-molecule drugs largely due to the lack of efficient syntheses. Even with the widespread presence of macrocyclic alkenes in therapeutic agents, stereoselective access to either E- or Z-alkenes remains a formidable challenge.

"Semiparametric Copula-Based Regression Modeling of Semi-competing Risk" by Hong Zhu, Yu Lan et al.
Abstract Semi-competing risks data often arise in medical studies where the terminal event (e.g., death) censors the non-terminal event (e.g., cancer recurrence), but the non-terminal event does not prevent the subsequent occurrence of the terminal event. This article considers regression modeling of semi-competing risks data to assess the covariate effects on the respective non-terminal and terminal event times.
