Increased potency of human placental mesenchymal stromal cell-derived extracellular vesicles from scaled-up hollow fiber bioreactor production
ABSTRACT Mesenchymal stromal cells (MSCs), and more recently MSC-derived extracellular vesicles (MSC-EVs), have demonstrated translational potential, with the first MSC therapy approved by the FDA in 2024 and multiple clinical trials evaluating MSC-EVs. However, clinical translation of MSC-EVs remains limited by batch-to-batch variability in EV properties.
Charge-switching ionizable lipids lower the toxicity of lipid nanoparticles
Abstract Lipid nanoparticles (LNPs) have great potential as nucleic acid delivery vehicles; however, they trigger the production of inflammatory cytokines, which limits their medical applications. Developing non-inflammatory LNPs is challenging because the LNP’s ionizable lipid and the process of endosomal disruption are the major sources of LNP toxicity but are also essential for delivering nucleic acids.

Hypoxanthine—early biomarker of outcomes in an ovine model of neonatal hypoxic ischemic encephalopathy
Abstract Hypoxic-ischemic encephalopathy (HIE) is the leading cause of neonatal morbidity and mortality globally. An early tool to diagnose and prognosticate disease could aid in timely intervention and reduction in disease burden. We aimed to identify biomarkers for HIE and define whether these biomarkers are associated with neurologic outcome severity in our ovine model. Study Cohort 1 (n = 46) included lambs with HIE induced via umbilical cord occlusion (UCO) and healthy controls (n = 19).

Designer artificial environments for membrane protein synthesis - Nature Communications
Abstract Protein synthesis in natural cells involves intricate interactions between chemical environments, protein-protein interactions, and protein machinery. Replicating such interactions in artificial and cell-free environments can control the precision of protein synthesis, elucidate complex cellular mechanisms, create synthetic cells, and discover new therapeutics.
Human skeletal development and regeneration are shaped by functional diversity of stem cells across skeletal sites
|
Results To investigate stem cell diversity in the human skeleton, we isolated hSSCs (CD45−CD235a−TIE2−CD31-CD146−PDPN+CD73+CD164+) from ten skeletal sites of the developing human fetus at the gestational age of 17–20 weeks and conducted functional and single-cell transcriptional analysis (Figures 1A, S1A, and S1B; Table S1).13,14 Using established in vitro assays, we confirmed that hSSCs of different bones and skeletal regions displayed the potential to clonally expand and to differentiate...

Harnessing extracellular vesicle heterogeneity for diagnostic and therapeutic applications - Nature Nanotechnology
Abstract Extracellular vesicles (EVs) are diverse nanoparticles with large heterogeneity in size and molecular composition. Although this heterogeneity provides high diagnostic value for liquid biopsy and confers many exploitable functions for therapeutic applications in cancer detection, wound healing and neurodegenerative and cardiovascular diseases, it has also impeded their clinical translation—hence heterogeneity acts as a double-edged sword.

Acid-degradable lipid nanoparticles enhance the delivery of mRNA - Nature Nanotechnology
Abstract Lipid nanoparticle (LNP)–mRNA complexes are transforming medicine. However, the medical applications of LNPs are limited by their low endosomal disruption rates, high toxicity and long tissue persistence times. LNPs that rapidly hydrolyse in endosomes (RD-LNPs) could solve the problems limiting LNP-based therapeutics and dramatically expand their applications but have been challenging to synthesize.

EOG and the En-Face Inner Segment/Outer Segment-Ellipsoid Complex Imag
Introduction Multiple evanescent white dot syndrome (MEWDS) is a unilateral inflammatory disorder characterized by the presence of multiple subretinal white dots extending from the posterior pole to the mid-periphery, which spontaneously resolve without leaving any scarring.1 This study reports a rare case of MEWDS with aberrant supernormal response of electrooculogram (EOG) and abnormal en-face images of Minimum intensity projection (Min-IP) and the en-face inner segment/outer...
A shift of brain network hub after spinal cord injury
Introduction Spinal cord injury (SCI) causes dysfunction of the spinal cord at and below the level of injury, followed by impairments such as motor and sensory paralysis. Depending on the extent of the injury, spontaneous recovery is limited and often results in serious sequelae. Worldwide, one million people are affected by SCI each year, yielding a significant social loss (GBD 2016 Traumatic Brain Injury and Spinal Cord Injury Collaborators, 2019).
The complete and fully-phased diploid genome of a male Han Chinese - Cell Research
Abstract Since the release of the complete human genome, the priority of human genomic study has now been shifting towards closing gaps in ethnic diversity. Here, we present a fully phased and well-annotated diploid human genome from a Han Chinese male individual (CN1), in which the assemblies of both haploids achieve the telomere-to-telomere (T2T) level. Comparison of this diploid genome with the CHM13 haploid T2T genome revealed significant variations in the centromere.
Semantic Bimodal Presentation Differentially Slows Working Memory Retrieval
1. Introduction Working memory (WM) is typically considered a system with limited capacity that can process, store, and monitor information in a short period [1], which is the ability and/or resources to concurrently process and store task-relevant information or coordinate the processing of multiple competing inputs [2].
Water | Free Full-Text | Dynamic Changes in Landscape Pattern of Mangrove Wetland in Estuary Area Driven by Rapid Urbanization and Ecological Restoration: A Case Study of Luoyangjiang River Estuary in Fujian Province, China
Abstract : Coastal wetlands are natural complexes situated between terrestrial and marine ecosystems and are one of the most productive ecosystems in terms of global biomass production. However, under the influence of intensive human activity, global coastal wetlands have undergone rapid degradation.
Characteristics of Sedimentary Organic Matter in Tidal Estuaries: A Case Study from the Minjiang River Estuary
Abstract : As one of the main interfaces of the Earth system, estuaries show the strongest land–sea interaction in the carbon cycle, which links terrestrial ecosystems to the marginal sea. Furthermore, estuaries are considered as one of the most active intermediate reservoirs for both terrestrial and marine matter due to complex hydrodynamic processes regulated by the river runoff, wave and tide.

Dual blockade of CD47 and HER2 eliminates radioresistant breast cancer cells
Abstract Although the efficacy of cancer radiotherapy (RT) can be enhanced by targeted immunotherapy, the immunosuppressive factors induced by radiation on tumor cells remain to be identified. Here, we report that CD47-mediated anti-phagocytosis is concurrently upregulated with HER2 in radioresistant breast cancer (BC) cells and RT-treated mouse syngeneic BC. Co-expression of both receptors is more frequently detected in recurrent BC patients with poor prognosis.
The pathogenic mechanisms of Tilletia horrida as revealed by comparative and functional genomics
The genome of the T. horrida strain JY-521 was sequenced using a PacBio RS II sequencing strategy and three single-molecule real-time (SMRT) sequence cells were obtained (Supplementary Table S1). The three SMRT cells were assembled into scaffolds using MinHasf Alignment Process (MHAP) assemble, resulting in longer sequences with a total assembly size of 23.2 Mbp; 84 contigs and 84 scaffolds (no gaps within them) were assembled. The N50 size of the scaffold and contigs was 538,348 bp.
