The Home Stretch
Well, it’s 15 days to Election Day. What a time. My calendar has been stacked with fundraisers. I’m heading to South Dakota this weekend. And I get tons of emails and texts. New York has been bombarded by political TV advertisements, as we have an unusually contested gubernatorial race and multiple competitive Congressional races. Kathy Hochul is either the worst governor of all time or a phenomenal one, sometimes within the same commercial break. You can feel the money more strongly than ever.
Interpretable and scalable spatial gene set activity analysis with GESSO uncovers functional tissue architecture
Abstract Spatially resolved transcriptomics (SRT) measures gene expression within tissues. Existing methods that summarize the activity of functionally related genes often ignore relationships between neighboring locations and offer limited statistical testing, gene-level interpretation, or scalability.
The Fall of Our Discontent
Americans are mad. It’s showing up in the numbers and anywhere you look. 55 percent of Americans think their personal finances are deteriorating. People can’t save money. Inflation is stubborn. Real wages, adjusted for inflation, have been negative since April of this year, a span of 6 months. This is translating to deep dissatisfaction with Trump, Republicans and the folks currently in power.
Flying Blind
Hello, I hope that your fall is going great. Muse, the new AI personal assistant from Meta, has taken the world by storm. It became #1 in App Store downloads and increased Meta’s market cap by $400 billion or so. People in my life have reacted anywhere from “No way I’d let Zuck get deep into my personal life” to “I need to try that immediately!” But it’s going to become a fact of life for millions right now. It also caused consternation in the business world.
Surrounded on AI
What impact will AI have on inequality? A couple months ago, I was asked to debate 20 AI optimists for a broadly watched online series “Surrounded.” You’ve probably seen the thumbnails on YouTube or other platforms; a person sits on a chair literally surrounded by people who want to take the opposite point of view. These things get a LOT of views, and I’ve always wanted to debate 20 people at once. So, I said yes. I agreed to back the following 4 statements: 1.
The Alignment Problem
Hello, I hope that you’re doing great. What are you more worried about, AI killing us or something else? I did a whole series of interviews last week about AI as a threat to humanity due to Jacob Coxon’s warning after leaving Anthropic. On CNBC, I shared the belief of the head of one lab that bots had left prompts to self-replicate in forums and on various websites during a training run.
Democrats and Republicans are failing America's men | Opinion
The 2026 midterm elections are fast approaching, and both Republicans and Democrats are squandering a massive opportunity while letting down the country. Neither party is seriously addressing the struggles of boys, sons, fathers, and husbands across America. It's no secret that a surge in young male voters helped tip the scales to President Trump and the GOP in 2024.

A Warning for Humanity
Last week, Anthropic researcher Jacob Coxon publicly resigned with a warning that there was a >10% chance of AI destroying humanity by the end of the decade. That is, within the next 4 years. It took over the news for days. A variety of researchers and industry figures have made similar warnings over the past months. There were recent reports of bots breaking free of their sandbox and hacking organizations, and of governments trying to weaponize Anthropic’s Claude to harm American interests.
Labor on the Run
It’s Labor Day, which is a fine occasion to reflect on where we are. Many leaders are waking up to the fact that our economic system – capitalism – is becoming unduly harsh and punitive for millions of Americans. And these critiques are coming from hard-nosed business leaders: Ray Dalio: “Capitalism is not working for the majority of people.” Marc Benioff: “Capitalism as we have known it is dead.” Warren Buffett: “The economic system...
VINE-seq and MultiVINE-seq for single-nucleus and multiome profiling of the brain vasculature
Abstract The human cerebrovasculature is a critical yet historically understudied component of neurological health. Dysfunction of the diverse endothelial, mural, and perivascular cells that comprise cerebral vessels is central to diseases ranging from stroke to Alzheimer’s disease. However, characterizing these cell populations at a molecular level has proven exceptionally challenging.
Who Do You Trust?
“We have entered an age of transparency where we can see our institutions and leaders for all of their flaws. Trust is for the gullible. Everything now will be a fight. Appealing to common interests will be all the more difficult.” I wrote that in “The War on Normal People” back in 2018.
Waterloo Rocketry team breaks world record with rocket launch
Student design team Waterloo Rocketry’s rocket Polaris flew up to 63,497 feet during its launch on Thursday, August 20 at 11:50 a.m., breaking the world record for the maximum height reached by an amateur liquid rocket. Liftoff took place in Timmins, Ontario, at Launch Canada, the largest student rocketry challenge in Canada. The launch marks the first nominal flight of a liquid rocket in Canada and the first successful recovery of a rocket in the team’s history.
Summer Reflections
Hello, I hope that your summer has been great. My sons went to academic camp this summer. I am Asian after all. I went to the same camp when I was their age. That’s why I’m President today. Dropping them off made me nostalgic for summers from my youth. Do you have songs you associate with summers long-gone-by? I certainly do, with the summers of 1988 – 1994 particularly prominent. The anthem of this academic camp is “American Pie” by Don McLean, so that song is embedded in my teenage soul.
The Anger is Rising
Hello, I hope you’ve had a great summer. Friday brought with it a disastrous jobs report. The economy lost 23,000 jobs in July, well below expectations of a gain of 85,000. That’s a difference of 108,000 jobs. June’s number was also revised down by 37,000, so you’re talking about 145,000 fewer jobs. Perhaps even more ominously, 260,000 more Americans stopped looking for work and left the labor market entirely, as the labor force participation rate dropped to a five-year low.
Where Dems Are the Spoilers
Hello, I hope the summer is going great for you.

Idaho Just Got Real
Some big news came out on Tuesday – David Roth, the Democratic Nominee for Senate in Idaho, dropped out of the race! Why is that a big deal? It leaves the Senate race as a one-on-one between Jim Risch, the 83-year-old incumbent, and Todd Achilles, a former tank commander in the U.S. Army and Independent candidate. I wrote about Todd last month.
Repeating Their Mistakes
Occasionally something happens that gives me a bad feeling in the pit of my stomach. It happened this past Friday when the DNC announced that South Carolina would be the first primary state in the 2028 Democratic primary. The DNC committee voted 26 for South Carolina, 19 for Nevada, and 4 for New Hampshire. “Okay, so what?” you ask. Let’s go back to the 2020 primary - Joe Biden’s campaign is on life support in February after weak results in Iowa, New Hampshire and Nevada. Bernie is on the march.
The Odyssey
I watched the Odyssey this past week with Evelyn and some other friends. We saw it in IMAX 70MM during the afternoon. I was blown away. I thought it was one of the greatest movies I’ve ever seen. The visuals, the acting, the sound, the action, the scope, the journey, the narrative of overcoming crazy obstacles over a long period of time, the secondary characters, the dog, all of it. I ordinarily start to get antsy during super-long films, but I was rapt the whole time.
Making and Taking Calls
I got a phone call from a candidate for Governor in a Midwestern state yesterday. We are friendly, but he’s a current candidate, so this wasn’t a social call. I cut to the chase: “Hey, I know you’re in the midst of a campaign. What can I do to help?” “Well, we have a fundraising deadline this week and want to put up a good number. Thank you for already donating to my campaign. Could you donate even more?” “Sure,” I said. “Happy to help.” “Great,” he said.
fMRI during active GPi DBS uncovers target-specific networks
Abstract Improved understanding of target-specific changes in brain activity during deep-brain stimulation (DBS) for Parkinson’s disease (PD) may refine target selection. Here, we study the network effects of active DBS using functional magnetic resonance imaging (fMRI) during stimulation of the subthalamic nucleus (STN) and globus pallidus internus (GPi). About 36 PD patients (62.5 ± 8.1 years, 18 GPi-DBS, 39% female) underwent prospective fMRI during active DBS.
Nation’s Dumbest
Hello, I hope your summer has been going great! Stay cool and hydrated out there. There’s a chance you’ve seen an ad for a reality TV show that I’m in. What’s the story? A number of months ago, I got a call that surprised me: “Hey, you’ve been asked to appear on a new reality TV show called ‘Nation’s Dumbest.’ It’s a fun game show like ‘Are you Smarter Than a 5th Grader?’ What do you think?” I was surprised on a bunch of levels. I don’t think of myself as a celebrity in the reality TV sense.
What We Do For a Living
Hello, I hope that your summer is going great. I had a lovely 4th at a friend’s barbecue. As you know, I ran for President in 2020 with the main idea that AI was going to come along and do a number on jobs. I believed that advancing Universal Basic Income was the best hope that we had of managing this transition. I wrote a book, The War on Normal People, making this case and then stumped around the country for months rousing millions of Americans. You might have been one of them. How am I feeling now?
The Word from Inside the House
Last week a $500 million non-profit funded by Anthropic, OpenAI, Amazon and many others was announced. It’s called RAISE US and works primarily with state governments to help workers transition to different jobs in the AI age. I’m glad people are starting to act. But I will say that this kind of effort is an enormous acknowledgment that tons of workers are going to lose their jobs to AI. Among those already affected by the arrival are those in Silicon Valley.
Achilles in the Senate
No, not that one, though I’m definitely going to see the Odyssey when it comes out in a few weeks. Chris Nolan remains one of our greatest movie directors until proven otherwise. He made the Dark Knight and Inception. Tenet . . . we’ll pretend that one didn’t happen. I’m talking about Todd Achilles, candidate for U.S. Senate out of Idaho. Yes, that’s his real name. Todd is running as an Independent after serving as a state legislator in Boise.
Cerebrovascular vulnerability and fibrosis in human brain aneurysms
|
Abstract Brain aneurysms are a cerebrovascular disease that results in a severe type of stroke. The cell-specific molecular pathology underlying their formation and rupture is unknown. Here we profile 227,663 neurovascular cells, including 52,946 aneurysmal cells, from a total of 14 adult human brain aneurysms and 11 control vessels.

Tax AI, Not Humans, Again
Hello all, I hope your summer is off to a great start. It snuck up quickly on us, didn’t it? I was back on CNBC this week to make the case that we should tax AI – and eventually robots – and not humans. They invited me back because I think they’re worried about the impact of AI. First, the big question – is AI going to displace human workers? Of course it is. You’re going to spend a trillion dollars on AI infrastructure and data centers... and then it won’t affect the labor market?

Do More In Four
There’s a lot going on out there, as usual. But I want to spotlight a solution that I think would make a huge difference in millions of people’s lives that is achievable right now without legislation. What is it? A four-day workweek. In early 2025, I got an email asking, “Do you want to be interviewed for a book about a 4-day workweek?” “Sure,” I said. This was a topic that I was very fond of. The book is now out.
Too Many College Grads
What happens when there are too many college grads? This week on the podcast I interview Noam Scheiber, the author of “Mutiny: The Rise and Revolt of the College-Educated Working Class.” Noam painstakingly interviewed and followed college-educated employees who tried to unionize stores and warehouses at Starbucks, Apple and Amazon for several years. He also followed a young Hollywood writer who was part of the 2023 strike.
Temporomandibular Joint Lesions with Intracranial Extension: Illustrative Cases from a Systematic Review of the Literature and Our Institution
Abstract BACKGROUND: Intracranially extending temporomandibular joint (TMJ) lesions may be radiologically misinterpreted as primary intracranial or skull base pathologies, leading to diagnostic delays or inappropriate management. PURPOSE: This systematic review aimed to characterize the clinical and imaging features of such TMJ lesions and evaluate the impact of radiologic misclassification.
What Jobs Are Safe?
Last year, Evelyn bought me 10 sessions with a personal trainer. “Huh, I guess I should take the hint.” I thought, and dutifully reported to get my butt whipped into shape by Aaron, a very pleasant 31-year-old with a degree in physical therapy. During our fifth workout or so, Aaron said to me, “Hey, have you seen some of these personal training apps? Should I be worried that my job is going to disappear?” Aaron was about to propose to his girlfriend and had money on his mind.
Tax the Bots
I was on CNBC last week talking about AI and jobs. I painted a foreboding picture of corporate layoffs that have just kicked off. Indeed, Oracle and Amazon just announced that they would likely cut more than 30,000 workers between them. One of the anchors, Becky Quick, asked me, “So what do we do?” I answered, “We generally tax things we want less of. In this situation, we want to preserve labor in every corner and environment.
Notes from the Tour
I’m writing this on the plane, in true author fashion, on the way back cross country. It was wonderful seeing so many familiar faces and meeting new people on the book tour. Thank you to Rikki Schlott, Nancy Scola, Rainn Wilson, Kristen Sze, and Jon Sposato for agreeing to moderate convos. The LA event got postponed because I got caught in the blizzard. But I made it to SF and Seattle, which were very welcoming. What were the takeaways of the “Hey Yang, Where’s My Thousand Bucks?” tour?
Vitamin B2 and B3 nutrigenomics reveals a therapy for NAXD disease
|
To determine which brain cell types are most affected by NAXD loss and whether specific stress pathways are activated, we performed single-nuclei RNA sequencing (snRNA-seq) on whole brains from P5 Naxd KO and control mice, with or without daily vitamin B3 supplementation (n = 4 per group) (see data and code availability). After quality control and normalization (Figure S4), the dataset contained 393,197 nuclei.

Is College (Still) Worth It?
A friend recently told me that she and her husband sold their house to pay for their daughter attending a private university. I said, “Really?” in a tone that made it seem that I didn’t necessarily agree with the decision. “It was her dream school,” she explained. “It’s also where both her Dad and I went. And she’s our only child. It seemed wrong to deny her something that we both had.

The End of the Office
I write this filled with sadness. Last week, I wrote about how AI is now replacing white-collar workers in earnest as Claude’s Co-work released plug-ins for legal, financial and marketing functions. Social media is filled with accounts from long-time software developers declaring themselves obsolete based on AI’s capabilities right now. Physicists report the same. Someone in my family had AI program a website for them this week.
Therapeutic targeting of neuroimmune mechanisms in neurodegeneration - Nature Reviews Drug Discovery
Abstract Effective treatments for age-related chronic neurodegenerative diseases such as Alzheimer’s disease remain limited, in part because the molecular drivers of cognitive decline are still not fully understood. Human genetic studies, together with detailed analysis of disease pathology, indicate that the immune system has an important influence on disease progression.
The AI Meltdown
12 days ago, one of the biggest stories in tech and business unfolded that will affect us all. On January 30th, the AI Company Anthropic released open-source plugins for its workplace AI offering - Cowork - for legal, finance, sales and marketing. There was a freakout of epic proportions. Thomson Reuters plunged a record 15.8%. LegalZoom sank 19.7%. Even the advertising giant WPP shed 15%. The software sector collapsed to the tune of $285 billion of market capitalization evaporating in a single day.
Veterans Affairs ICU Nurse Killed By Trump’s Agents In Minnesota-While Trying To Help Someone
Africa News Reported By Andrew Yang Photos: Wikimedia Commons Alex Pretti, a 37-year-old American citizen, was shot and killed by ICE agents in Minneapolis on Saturday. Alex was an Intensive Care Unit nurse who worked with military veterans in the VA. He was wrestled to the ground by multiple agents and was shot multiple times after trying to assist someone. Alex was apparently legally carrying a registered firearm at the time of the encounter.

The Verizon Outage
I hope that your week has been great. I had a wonderful week celebrating my birthday with family and friends. Millions of people lost their Verizon service this week on Wednesday. Why did this happen? “A software issue.” Verizon issued an apology but specifics have yet to emerge. They did offer a $20 rebate to those who requested it, which hasn’t gotten great feedback.
Swinging and Missing
Hello! As you may know, I wrote a book called “Hey Yang, Where’s My Thousand Bucks?” that comes out February 10th. It’s a more personal and humorous account of my rise and fall and rise again to become the emperor. To get you to want to buy the book, here’s an excerpt that recounts my misspent twenties as a young, wannabe entrepreneur. Hope you enjoy! The book is now available for pre-sale and I hope you consider buying a copy. Use code “UBIUBI” for 25% off.
The People of Iran
The most important story in the world right now is the popular uprising in Iran. This past week, tens of thousands of Iranians have begun marching against Iran’s brutally repressive regime. The current theocracy has been in place since 1979, when it deposed a U.S.-allied monarch. Today’s protests are fueled in party by a collapsing economy and deteriorating way of life. Inflation in Iran is rampant and food is hard to afford. One dollar is now worth 1.47 million rials, to give you a sense of it.
Renee Nicole Good
ICE shot and killed a 37-year-old American citizen, Renee Nicole Good, in Minneapolis on Wednesday. Renee was in her SUV at an ICE enforcement action and was approached by several armed agents who told her to get out of her car. One agent tried to access her SUV by opening the door. She seemingly panicked and tried to drive away. Another agent, who was partially in her vehicle’s path, drew out his gun and lethally shot her twice. The video was recorded by onlookers.
