MMSDH facilitates ACSL4 propionylation to counteract ferroptosis upon hypoxia and impairs PDAC chemotherapy efficacy
Abstract Limited efficacy of neoadjuvant chemotherapy (NAC) in pancreatic ductal adenocarcinoma (PDAC) underscores the need for novel combination strategies and a deeper understanding of metabolic determinants of chemoresistance. Here we identify methylmalonate semialdehyde dehydrogenase (MMSDH), a valine catabolism enzyme, as a driver of gemcitabine (GC) resistance. Hypoxia induces GCN5-mediated lactylation of MMSDH at K113.
A targetable ALDH3A2-NRF2 axis mediates KEAP1-mutant tumour development via ferroptosis inhibition
Abstract The transcription factor nuclear factor erythroid 2-related factor 2 (NRF2) is a master regulator of the cellular antioxidant response that defends against ferroptosis and facilitates tumour development. Here, by performing an aldehyde dehydrogenase (ALDH) family shRNA screen, our study identifies aldehyde dehydrogenase 3 family member A2 (ALDH3A2) as a potent NRF2 activator that inhibits ferroptosis.
Stress granules restrain ferroptosis by sequestering ferritin
|
Abstract Glioblastoma stem cells (GSCs) are refractory to first-line treatment in the clinic, which includes irradiation (IR) and temozolomide (TMZ). Here we find that disrupting stress granules (SGs) sensitizes GSCs to IR/TMZ through ferroptosis. The profiling of SG proteins reveals the recruitment of iron-related proteins including ferritin.

Sensing steroid hormone 17α-hydroxypregnenolone by GPR56 enables protection from ferroptosis-induced liver injury

Palmitoylation-dependent regulation of GPX4 suppresses ferroptosis - Nature Communications
Abstract S-palmitoylation is a reversible and widespread post-translational modification, but its role in the regulation of ferroptosis has been poorly understood. Here, we elucidate that GPX4, an essential regulator of ferroptosis, is reversibly palmitoylated on cysteine 66. The acyltransferase ZDHHC20 palmitoylates GPX4 and increases its protein stability. ZDHHC20 depletion or inhibition of protein palmitoylation by 2-BP sensitizes cancer cells to ferroptosis.

Jag1/2 maintain esophageal homeostasis and suppress foregut tumorigenesis by restricting the basal progenitor cell pool - Nature Communications
Abstract Basal progenitor cells are crucial for maintaining foregut (the esophagus and forestomach) homeostasis. When their function is dysregulated, it can promote inflammation and tumorigenesis. However, the mechanisms underlying these processes remain largely unclear. Here, we employ genetic mouse models to reveal that Jag1/2 regulate esophageal homeostasis and foregut tumorigenesis by modulating the function of basal progenitor cells.

Cholesterol mediated ferroptosis suppression reveals essential roles of Coenzyme Q and squalene - Communications Biology
Abstract Recent findings have shown that fatty acid metabolism is profoundly involved in ferroptosis. However, the role of cholesterol in this process remains incompletely understood. In this work, we show that modulating cholesterol levels changes vulnerability of cells to ferroptosis. Cholesterol alters metabolic flux of the mevalonate pathway by promoting Squalene Epoxidase (SQLE) degradation, a rate limiting step in cholesterol biosynthesis, thereby increasing both CoQ10 and squalene levels.
Structural insights into FSP1 catalysis and ferroptosis inhibition - Nature Communications
Abstract Ferroptosis suppressor protein 1 (FSP1, also known as AIMF2, AMID or PRG3) is a recently identified glutathione-independent ferroptosis suppressor1,2,3, but its underlying structural mechanism remains unknown. Here we report the crystal structures of Gallus gallus FSP1 in its substrate-free and ubiquinone-bound forms.
Inhibition of USP7 induces p53-independent tumor growth suppression in triple-negative breast cancers by destabilizing FOXM1 - Cell Death & Differentiation
Abstract Although numerous studies indicate that inhibition of USP7 suppresses tumor growth by activating p53, the precise mechanism by which USP7 contributes to tumor growth through the p53-independent manner is not well understood. p53 is frequently mutated in most triple-negative breast cancers (TNBC), characterized as the very aggressive form of breast cancers with limited treatment options and poor patient outcomes.
Identification and screening of fungicides against Piper nigrum basal Fusarium wilt disease in Hainan, China
Fusarium wilt has occurred in the main Piper nigrum cultivation regions, which seriously affects the yield and quality of P. nigrum. To identify the pathogen of this disease, the diseased roots were collected from a demonstration base in Hainan Province. The pathogen was obtained by tissue isolation method and confirmed by pathogenicity test. Based on the morphological observation, sequence analyses of TEF1-α nuclear gene, Fusarium solani was identified as the pathogen causing P.
NIR Clusteroluminescence of Non‐conjugated Phenolic Resins Enabled by Through‐Space Interactions
Graphical Abstract Nonconjugated luminescent materials derived from well-developed engineering plastic of phenolic resins could fluoresce from deep blue to the NIR region. This remarkable luminescence is attributed to through-space interactions (TSIs) and can be manipulated using two molecule-engineering strategies: increasing the number of TSIs units and introducing D/A groups.
Molecular subtyping of acute myeloid leukemia through ferroptosis signatures predicts prognosis and deciphers the immune microenvironment
Introduction Acute myeloid leukemia (AML) is a type of aggressive blood malignancy characterized by the stagnant transition of immature myeloid cells to mature cells in the bone marrow and peripheral blood (Thomas and Majeti, 2017). Despite great advancements in therapies for most blood cancer types, treatment options for AML are still limited over decades. Therapeutic resistance and a high rate of relapse lead to a low 5-year survival rate (Jemal et al., 2017; Marando and Huntly, 2020).

Carbon dioxide derived carbonized polymer dots for multicolor light-emitting diodes
Carbon dioxide derived carbonized polymer dots for multicolor light-emitting diodes Carbonized polymer dots (CPDs) have received great interest from researchers in recent years. However, the solid-state photoluminescence quenching of CPDs is a big challenge owing to the resonance energy transfer or π–π* interaction. Herein, we report a new type of carbon dioxide (CO2) derived CPD exhibiting quantum yields of 46.2% in solution and 11.3% as a solid.

ALOX12 is required for p53-mediated tumour suppression through a distinct ferroptosis pathway
Abstract It is well established that ferroptosis is primarily controlled by glutathione peroxidase 4 (GPX4). Surprisingly, we observed that p53 activation modulates ferroptotic responses without apparent effects on GPX4 function. Instead, ALOX12 inactivation diminishes p53-mediated ferroptosis induced by reactive oxygen species stress and abrogates p53-dependent inhibition of tumour growth in xenograft models, suggesting that ALOX12 is critical for p53-mediated ferroptosis.
The role of microRNA-146a in regulating the expression of IRAK1 in cerebral ischemia-reperfusion injury
MicroRNA-146a (miR-146a) is reportedly implicated in the pathogenesis of ischemia–reperfusion (I/R) injury; however, its role in cerebral I/R injury is unclear and requires further investigation. In this study, cerebral I/R injury was established in mice via middle cerebral artery occlusion, and the expression of miR-146a was detected in the brain tissue via quantitative real-time PCR. We found that the expression of miR-146a was upregulated.
