Structural Basis of Amyloid Fibril Assembly by Plant Seed Storage Proteins
Abstract Amyloid fibrils are highly ordered protein assemblies characterized by a cross-β architecture. A wide range of proteins can adopt amyloid states, contributing to both disease-related pathology and normal physiological function. While animal-derived amyloids have been extensively examined in atomic detail, amyloid fibrils formed by plant proteins remain relatively understudied.

Clinical implications of free triiodothyronine levels and diagnostic revisions in antibody-negative autoimmune encephalitis
Abstract Background and objective: Low triiodothyronine (T3) syndrome has been associated with initial clinical severity and poor long-term functional outcomes in autoimmune encephalitis (AE). However, its prognostic implications in antibody-negative AE remain unclear.
Development and validation of a nomogram for early prognostic prediction in childhood aplastic anemia receiving cyclosporine monotherapy
Abstract The prognosis for aplastic anemia (AA) patients undergoing cyclosporine is variable; therefore, a prognostic model is needed to optimize stratified treatment strategies. This observational retrospective cohort study included patients with AA who received immunosuppressive therapy with cyclosporine. A total of 437 patients were randomly assigned to the training cohort (n = 262) or the validation cohort (n = 175) at a ratio of 6:4.
Ultrastructural and atomic characterization of biofilm-associated extracellular filaments in Shewanella oneidensis MR-1
Abstract Biofilms formed by Shewanella oneidensis MR-1 are crucial for metal reduction, underpinning bioremediation and bioenergy applications, yet detailed structural insights into biofilm components remain limited. Here we show that MR-1 biofilms contain abundant filamentous networks, membrane vesicles, and distinct square-shaped aggregates, as revealed by cryo-electron tomography and cryo-electron microscopy.
Rapid rescue of despair behaviors by sonogenetic neuromodulation of the mPFC-DRN pathway
Abstract Sonogenetics combines genetic tools and low-intensity ultrasound to non-invasively modulate specific neuronal populations and circuits, exhibiting potential for treating brain diseases. This study examines sonogenetics’ potential in a mouse depression model, targeting excitatory medial prefrontal cortex (mPFC) neurons projecting to the dorsal raphe nucleus (DRN).
808 nm laser diode with a stepped-structure heat sink for high-efficiency laser wireless power transmission
As the output power of laser diodes increases, the resulting thermal load leads to elevated chip temperatures and reduced electro-optical conversion efficiency. To address this challenge, we design a stepped-structure heat sink integrated with multi-channel water cooling in which 15 laser diode chips are arranged in a stepped configuration to achieve efficient thermal management.
A monoclonal antibody targeting the C-terminal of α-synuclein fibrils mitigates pathology in a Parkinson’s disease model
|
Results To systematically generate monoclonal antibodies that selectively target pathological α-syn fibrils, we established a multiplex screening platform comprising immunization, high-throughput binding assays, and functional validation in neuronal systems (Figure 1A). This resource-oriented platform was designed not only to isolate functional anti-α-syn antibodies but also to provide a generalizable workflow applicable to other amyloid pathologies.

NAD-dependent dehydrogenases enable efficient growth of Paracoccus denitrificans on the PET monomer ethylene glycol - Nature Communications
Abstract Ethylene glycol is a monomer of the plastic polyethylene terephthalate (PET) and an environmental pollutant of increasing concern. Although it is generally accepted that bacteria use ethylene glycol as growth substrate, not all involved enzymes are well understood. Here, we show that Paracoccus denitrificans assimilates ethylene glycol solely via NAD-dependent alcohol and aldehyde dehydrogenases.

PAK5 promotes the trastuzumab resistance by increasing HER2 nuclear accumulation in HER2-positive breast cancer - Cell Death & Disease
Abstract Nuclear HER2 (N-HER2) predicts resistance to HER2-targeted therapy and poor prognosis of breast cancer patients, and the underlying mechanisms remain unclear. Here, we show that high expression of p21-activated kinase 5 (PAK5) is associated with HER2-targeted therapy resistance and poor outcomes of breast cancer patients. Excitingly, we find an increase in N-HER2 protein expression in patients with high PAK5 expression, who demonstrate resistance to trastuzumab treatment.

Promiscuous NAD-dependent dehydrogenases enable efficient bacterial growth on the PET monomer ethylene glycol
Abstract Ethylene glycol is widely used as antifreeze agent and monomer of the ubiquitous plastic PET (polyethylene terephthalate). Its global production amounts to more than 50 million tons per year, and it constitutes an environmental pollutant of increasing concern. Although it is generally accepted that bacteria oxidize ethylene glycol to use it as growth substrate, the enzymes involved in this process are not well understood.

IJMS | Free Full-Text | A Prognostic Model of Head and Neck Cancer Based on Amino Acid Metabolism-Related Signature and Its Implication for Immunosuppressive Microenvironment
Abstract : Amino acid metabolism has been implicated in tumorigenesis and tumor progression. Alterations in intracellular and extracellular metabolites associated with metabolic reprogramming in cancer have profound effects on gene expression, cell differentiation, and tumor immune microenvironment. However, the prognostic significance of amino acid metabolism in head and neck cancer remains to be further investigated.

Calculate Computational Efficiency of Deep Learning Models with FLOPs and MACs
FLOPs (Floating Point Operations) and MACs (Multiply-Accumulate Operations) are metrics that are commonly used to calculate the computational complexity of deep learning models. They are a fast and easy way to understand the number of arithmetic operations required to perform a given computation. For example, when working with different model architectures such as MobileNet or DenseNet for edge devices, people use MACs or FLOPs to estimate the model performance.

Periventricular Microglia Polarization and Morphological Changes Accompany NLRP3 Inflammasome-Mediated Neuroinflammation after Hypoxic-Ischemic White Matter Damage in Premature Rats
White matter damage (WMD) is a primary cause of cerebral palsy and cognitive impairment in preterm infants, and no effective treatments are available. Microglia are a major component of the innate immune system. When activated, they form typical pro-inflammatory (M1) and anti-inflammatory (M2) phenotypes and regulate myelin development and synapse formation. Therefore, they may play a pivotal role in hypoxic–ischemic (HI) WMD.

Peripheral Inflammatory Biomarkers and Cognitive Decline in Older Adults With and Without Alzheimer's Disease: A Systematic Review
Journal of Gerontological Nursing | Peripheral inflammatory biomarkers may play an important role in the cognitive decline of aging and incidence of Alzheimer's disease (AD); however, data from epidemiological studies present conflicting findings. The purpose of the current review was to systematically determine the current state of the science on the association between peripheral inflammatory biomarkers and cognitive decline.
Association of Biotin Ingestion With Performance of Hormone and Nonhormone Assays in Healthy Adults
Question Does oral biotin supplementation interfere with hormone and nonhormone assays that use biotinylation in their design? Findings In this nonrandomized crossover study of 6 healthy adults (2 women, 4 men), 10 mg/d of biotin ingested for 1 week was associated with potentially clinically important assay interferences in some but not all biotinylated hormone and nonhormone assays studied. Meaning Oral biotin use may be associated with false hormone and nonhormone assay results.
