Monte Carlo modeling of the formation and organization of ion channel clustering
Discussion In this study, we have developed a stochastic computational model to investigate the principles governing ion channel organization on cell membranes. By systematically varying key parameters such as channel density, trafficking rates, and interaction energies, we have uncovered several fundamental properties of channel clustering that provide insight into the mechanisms underlying membrane domain formation.
The influence of intercalated disk nanostructure on local ionic currents and cardiac conduction
Significance Cardiac conduction critically depends on the nanoscale organization of the cell-cell junction, the intercalated disk (ID). We employed a refined 3D finite element mesh model of the ID nanostructure, integrating key ion channels and gap junctions and their nonuniform distributions, thus facilitating the quantification of the relative role of these properties on cardiac conduction.
Nebulized Treprostinil in Mechanically Ventilated Children
To the Editor, Pulmonary hypertension (PH) is attributable to lung disease in approximately one-third of affected children [1], many of whom require mechanical ventilation. For patients with heterogeneous lung disease, systemic pulmonary vasodilators may worsen ventilation-perfusion matching. The resultant hypoxemia can be mitigated by transitioning the route of administration from systemic (enteral or parenteral) to inhaled.

Recurrent Endobronchial Coil Migration Associated With Needle Biopsy Following Pulmonary Artery Pseudoaneurysm Embolization
1 Introduction Endobronchial coil migration is a rare complication of pulmonary artery embolization [1]. It occurs when coils erode through a vessel into an airway and has been described after embolization of pulmonary artery aneurysms [2-6], pulmonary arteriovenous malformations [7-12], pulmonary artery pseudoaneurysms (PAPs) [1, 13-15], and bronchial arteries [16].
Role for inflammatory markers in predicting right ventricular failure in mechanical assist device recipients
Introduction It is well established that heart failure (HF) is a global epidemic, which remains a major focus for research towards developing efficacious preventive and treatment algorithms.1-3 Approximately 10% of HF patients meet the criteria for advanced HF, defined as the presence of structural heart disease with a left ventricular ejection fraction (LVEF) of <25% and severe symptoms refractory to medical treatment.4 Orthotopic heart transplantation is the gold standard treatment for...
Nicotine inhalation and metabolism triggers AOX-mediated superoxide generation with oxidative lung injury
Keywords aldehyde oxidase electronic cigarettes superoxide free radicals EPR spin-trapping Introduction Nicotine (NIC) is the active and addicting element of tobacco cigarettes and electronic cigarettes (EC). In view of the harmful effects of tobacco cigarettes, EC were first proposed as a safe alternative; however, a number of recent studies have shown that EC can also cause a variety of toxicities (1-4).

Endothelial cell clonality, heterogeneity and dysfunction in pulmonary arterial hypertension
1 Introduction In most organs, endothelial cells (ECs) play a central role as a main component of the capillaries to provide oxygen and nutrition to the parenchymal cells (1). In the lung however the function of ECs is more complex, as the vascular system has dichotomous functions compared to any other vascular system in the body (2).

Emergent Activity, Heterogeneity, and Robustness in a Calcium Feedback Model of the Sinoatrial Node
The sinoatrial node (SAN) is the primary pacemaker of the heart. SAN activity emerges at an early point in life and maintains a steady rhythm for the lifetime of the organism. The ion channel composition and currents of SAN cells can be influenced by a variety of factors. Therefore, the emergent activity and long-term stability imply some form of dynamical feedback control of SAN activity.
Protein 14-3-3 Influences the Response of the Cardiac Sodium Channel Nav1.5 to Antiarrhythmic Drugs
Abstract The cardiac sodium channel Nav1.5 is a key contributor to the cardiac action potential, and dysregulations in Nav1.5 can lead to cardiac arrhythmias. Nav1.5 is a target of numerous antiarrhythmic drugs (AADs). Previous studies identified the protein 14-3-3 as a regulator of Nav1.5 biophysical coupling. Inhibition of 14-3-3 can remove the Nav1.5 functional coupling and has been shown to inhibit the dominant-negative effect of Brugada syndrome mutations.

Saliva and Lung Microbiome Associations with Electronic Cigarette Use and Smoking
The microbiome has increasingly been linked to cancer. Little is known about the lung and oral cavity microbiomes in smokers, and even less for electronic cigarette (EC) users, compared with never-smokers. In a cross-sectional study (n = 28) of smokers, EC users, and never-smokers, bronchoalveolar lavage and saliva samples underwent metatranscriptome profiling to examine associations with lung and oral microbiomes. Pairwise comparisons assessed differentially abundant bacteria species.

Initiation and Entrainment of Multicellular Automaticity via Diffusion Limited Extracellular Domains
ABSTRACT Electrically excitable cells often spontaneously and synchronously depolarize in vitro and in vivo preparations. It remains unclear how cells entrain and autorhythmically activate above the intrinsic mean activation frequency of isolated cells with or without pacemaking mechanisms. Recent studies suggest that cyclic ion accumulation and depletion in diffusion limited extracellular volumes modulates electrophysiology by ephaptic mechanisms (non-gap junction or synaptic coupling).
Mechanisms underlying age-associated manifestation of cardiac sodium channel gain-of-function
Highlights • Long QT syndrome type 3 can present early in life or be concealed to adulthood. • Age-dependent properties regulate LQT3-associated Na + channel gain-of-function. • Cell size and Na + channel density increases promote arrhythmias in neonatal tissue. • Intercellular cleft width increases promote arrhythmias in adult tissue. • Cellular property variability can lead to arrhythmias for all developmental stages.
The Effects of a 6-Week Controlled, Hypocaloric Ketogenic Diet, With and Without Exogenous Ketone Salts, on Body Composition Responses
Introduction Ketogenic diets (KDs) have been demonstrated to promote weight loss, often to a greater extent than low-fat diets, and without explicit instructions to limit calories (1–4). Some authors have suggested that low-carbohydrate diets may facilitate a metabolic advantage by increasing energy expenditure (5), but others have argued against such an effect (6).

IRAK-M Regulates Monocyte Trafficking to the Lungs in Response to Bleomycin Challenge
Previous work has demonstrated that macrophages derived from recruited monocytes promote pulmonary fibrosis (6, 8, 9); however, the mechanism by which these cells traffic to the lungs following bleomycin challenge had not been defined. In this study, we sought to determine the mechanism by which the presence or absence of IRAK-M enhances monocyte recruitment to the lung.

Treatment with insulin is associated with worse outcome in patients with chronic heart failure and diabetes
We examined two datasets: 24?012 patients with HF from four large randomized trials and an administrative database of 4 million individuals, 103?857 of whom with HF. In the former, survival was examined using Cox proportional hazards models adjusted for baseline variables and separately for propensity scores. Fine–Gray competing risk regression models were used to assess the risk of hospitalization for HF.
Aliskiren alone or in combination with enalapril vs. enalapril among patients with chronic heart failure with and without diabetes: a subgroup analysis from the ATMOSPHERE trial - Kristensen - 2017 - European Journal of Heart Failure - Wiley Online Librar
Because of concerns about the safety of aliskiren in patients with diabetes, study treatment was stopped prematurely in the Aliskiren Trial of Minimizing OutcomeS for Patients with HEart failuRE (ATMOSPHERE). We examined outcomes and treatment effect in these patients compared with those without diabetes. ATMOSPHERE included 7016 patients with heart failure and a reduced ejection fraction (HFrEF) randomly assigned to enalapril plus aliskiren, aliskiren alone, or enalapril.
