Prevalence of depressive symptoms and associated factors among empty-nest elderly in China: a meta-analysis
Abstract The empty-nest elderly may be particularly vulnerable to mental health problems, including depressive symptoms. However, estimates of the prevalence of depressive symptoms and associated factors among the empty-nest elderly in China vary substantially across studies. This meta-analysis was conducted in accordance with the PRISMA guidelines.

Discovery of actinators, actin-derived bioactive peptides that modulate cytoskeleton and actin-related cellular activities
Abstract The cortical actin cytoskeleton serves as a structural support and force-generating machinery in cells, but the actin meshwork can also act as a barrier, hindering the delivery of extracellular substances. To address the issue of the cortical actin barrier, we engineered synthetic libraries of human β-actin–derived peptides to promote actin dynamics.
The resting and ligand-bound states of the membrane-embedded human T-cell receptor–CD3 complex - Nature Communications
Abstract The T-cell receptor (TCR) initiates T-lymphocyte activation, but the mechanism of TCR activation remains uncertain. Here, we present cryogenic electron microscopy structures for the unliganded and human leukocyte antigen (HLA)-bound human TCR–CD3 complex in nanodiscs that provide a native-like lipid environment.

Yan Leyfman: Boosting CAR-T/NK Cell Persistence via the FAS Pathway
Yan Leyfman, Medical Correspondent of OncLive, shared a post on LinkedIn about an article by Fei Yi et al.:“Boosting CAR-T/NK Cell Persistence via the FAS Pathway Chimeric antigen receptor (CAR) T and NK cell therapies have transformed treatment for B-cell malignancies – but limited cell persistence remains a key barrier. In a new study led by Dr. Christopher A.
CAR-engineered lymphocyte persistence is governed by a FAS ligand–FAS autoregulatory circuit - Nature Cancer
Abstract Chimeric antigen receptor (CAR)-engineered lymphocytes treat B cell malignancies; however, limited persistence can restrain the full therapeutic potential of this approach. FAS ligand (FAS-L)/FAS interactions govern lymphocyte homeostasis. Knowledge of which cells express FAS-L in patients with cancer and whether these sources compromise CAR persistence remains incomplete.

The resting and ligand-bound states of the membrane-embedded human T-cell receptor-CD3 complex
Abstract The T-cell receptor (TCR) is central to the ligand-dependent activation of T lymphocytes and as such orchestrates both adaptive and pathologic immune processes. However, major questions remain regarding the structure and function of the human TCR. Here, we present cryogenic electron microscopy structures for the unliganded and HLA-bound human TCR-CD3 complex in nanodiscs that provide a native-like lipid environment.

Abstract The T-cell receptor (TCR) is central to the ligand-dependent activation of T lymphocytes and as such orchestrates both adaptive and pathologic immune processes. However, major questions remain regarding the structure and function of the human TCR. Here, we present cryogenic electron microscopy structures for the unliganded human TCR-CD3 complex in a native-like lipid bilayer, revealing two related conformations that are distinct from its structure in detergent.

CAR-engineered lymphocyte persistence is governed by a FAS ligand/FAS auto-regulatory circuit
Abstract Chimeric antigen receptor (CAR)-engineered T and NK cells can cause durable remission of B-cell malignancies; however, limited persistence restrains the full potential of these therapies in many patients. The FAS ligand (FAS-L)/FAS pathway governs naturally-occurring lymphocyte homeostasis, yet knowledge of which cells express FAS-L in patients and whether these sources compromise CAR persistence remains incomplete.

Erasing iatrogenic neoantigens from in vivo CRISPR screens
Dubrot et al. present the design of a scalable CRISPR vector system that removes Cas9 and other antigens after genome editing. They perform an in vivo genetic screen for immunotherapy dependencies in a murine renal cancer model that is sensitive to vector antigen expression.
The deacetylation-phosphorylation regulation of SIRT2-SMC1A axis as a mechanism of antimitotic catastrophe in early tumorigenesis
Abstract Improper distribution of chromosomes during mitosis can contribute to malignant transformation. Higher eukaryotes have evolved a mitotic catastrophe mechanism for eliminating mitosis-incompetent cells; however, the signaling cascade and its epigenetic regulation are poorly understood. Our analyses of human cancerous tissue revealed that the NAD-dependent deacetylase SIRT2 is up-regulated in early-stage carcinomas of various organs.
Non-dispersive infrared multi-gas sensing via nanoantenna integrated narrowband detectors
Abstract Non-dispersive infrared (NDIR) spectroscopy analyzes the concentration of target gases based on their characteristic infrared absorption. In conventional NDIR gas sensors, an infrared detector has to pair with a bandpass filter to select the target gas. However, multiplexed NDIR gas sensing requires multiple pairs of bandpass filters and detectors, which makes the sensor bulky and expensive.
Autism-associated SHANK3 haploinsufficiency causes Ih channelopathy in human neurons
SHANK3 is a widely expressed scaffolding protein that is enriched in postsynaptic specializations. In mutant mice, SHANK3 mutations cause autism-like behavioral changes and exhibit alterations in synaptic transmission. Yi et al. produced human neurons lacking SHANK3 but not other genes that are also involved in the autism-like disease Phelan-McDermid syndrome.
Autism-associated SHANK3 haploinsufficiency causes Ih channelopathy in human neurons
Heterozygous SHANK3 mutations are associated with idiopathic autism and Phelan-McDermid syndrome. SHANK3 is a ubiquitously expressed scaffolding protein that is enriched in postsynaptic excitatory synapses. Here, we used engineered conditional mutations in human neurons and found that heterozygous and homozygous SHANK3 mutations severely and specifically impaired I channels.
