Abstract Standard genome-wide association studies (GWAS) and rare variant burden tests are essential tools for identifying trait-relevant genes1. Although these methods are conceptually similar, by analysing association studies of 209 quantitative traits in the UK Biobank2,3,4, we show that they systematically prioritize different genes. This raises the question of how genes should ideally be prioritized.

Discussion What determines the genetic architecture of complex traits? Does genetic variation in a given trait primarily reflect the idiosyncrasies of its biology–or alternatively, does it reflect processes that are shared among different traits? Here we describe a principled approach to tackle these questions. Our point of departure is an evolutionary model of genetic architecture based on empirically motivated and interpretable biological assumptions.

Making sense of the polygenicity of complex traits - Nature Reviews Genetics

In the early twentieth century, scientists faced a puzzle: if hereditary material is transmitted in discrete units, how can traits such as height show continuous or quantitative variation? This puzzle was largely resolved by R. A. Fisher in 1918, who showed that multiple genes, each inherited according to Mendelian rules, together with environmental variation, could produce a continuous trait distribution.
