Structural and biochemical basis for cannabinoid cyclase activity in marine bacterial flavoenzymes
Abstract The marine bacterial flavoenzymes Clz9 and Tcz9 can process cannabigerolic acid to the minor cannabinoid, cannabichromenic acid (CBCA); however, the mechanistic details of this extrinsic transformation are still obscure. Here we report a thorough analysis of CBCA formation by Clz9 and Tcz9 through high-resolution crystallographic characterization, biochemical analysis and spectroscopic interrogation.

CRISPR PERSIST-On enables heritable and fine-tunable human gene activation
Abstract Current technologies for upregulation of endogenous genes use targeted artificial transcriptional activators but stable gene activation requires persistent expression of these synthetic factors. Although general "hit-and-run" strategies exist for inducing long-term silencing of endogenous genes using targeted artificial transcriptional repressors, to our knowledge no equivalent approach for gene activation has been described to date.
MOSAIC enables in situ saturation mutagenesis of genes and CRISPR prime editing guide RNA optimization in human cells
Abstract CRISPR prime editing offers unprecedented versatility and precision for the installation of genetic edits in situ. Here we describe the development and characterization of the Multiplexing Of Site-specific Alterations for In situ Characterization (MOSAIC) method, which leverages a non-viral PCR-based prime editing method to enable rapid installation of thousands of defined edits in pooled fashion.
How to Setup a Basic Test-Driven Typescript Environment
If you’re starting out learning Typescript, then the implications to project structure are just as daunting as (if not more so) than cleaning up your JS code to be type-safe. Moreover, if you’re looking to publish your project to NPM then there are even more considerations with how you setup your development space. Since one of the main reasons to use Typescript is to produce safer code, it’s also a great time to throw in automated testing… but isn’t that overwhelming?
Strapi: How to Restrict Users to Edit Only Themselves
If you’re reading this article, you’re probably mid-development in one of your first Strapi projects that includes user registration/authentication. That’s where I was when I needed this article. tl;dr: this article is a synthesis of a StackOverflow post I found on the topic, which can be found here. Here’s the situation we’re in. We want our users to be able to update their own user information such as name or email.
How to Use Customer Acquisition Cost (CAC) and Customer Lifetime Value (CLV)
Customer Acquisition Cost (CAC) and Customer Lifetime Value (CLV) are two of the most basic metrics a business can track as it relates to customer revenue. That being said, I’m continually surprised by how many companies do not explicitly use these labels as well as how many use the labels, but don’t do anything with the data. In this article, we’ll introduce CAC and CLV.
Response to Letter to the Editor
To the editor, We thank Kataoka et al. for their comments regarding our recently published article on catheter ablation of atrial fibrillation in very elderly patients (≥80 years old).1 While the unadjusted time-to-event analysis may be more reflective of real-world clinical practice, we adjusted for multiple potential confounders to more clearly assess the association of catheter ablation with all-cause hospitalizations and mortality in the very elderly.

Highlights • Ruptured SVA into LA is a rare disorder and has high mortality. • Wide pulse pressure without severe AR is concerning for SVA rupture. • Continuous turbulent Doppler flow by echo can identify the rupture of SVA. • Severe MR without structural valve abnormality raises the possibility of SVA rupture. Introduction Sinus of Valsalva aneurysm (SVA) is a rare disorder. Ruptured SVA has high morbidity and in-hospital death. Early identification of ruptured SVA is crucial in patient outcome.
Human-specific regulation of neural maturation identified by cross-primate transcriptomics
Unique aspects of human behavior are often attributed to differences in the relative size and organization of the human brain: these structural aspects originate during early development. Recent studies indicate that human neurodevelopment is considerably slower than that in other nonhuman primates, a finding that is termed neoteny. One aspect of neoteny is the slow onset of action potentials. However, which molecular mechanisms play a role in this process remain unclear.

Transcription factor competition at the γ-globin promoters controls hemoglobin switching
Abstract BCL11A, the major regulator of fetal hemoglobin (HbF, α2γ2) level, represses γ-globin expression through direct promoter binding in adult erythroid cells in a switch to adult hemoglobin (HbA, α2β2). To uncover how BCL11A initiates repression, we used CRISPR–Cas9, dCas9, dCas9-KRAB and dCas9-VP64 screens to dissect the γ-globin promoters and identified an activator element near the BCL11A-binding site.
PrimeDesign software for rapid and simplified design of prime editing guide RNAs
Abstract Prime editing (PE) is a versatile genome editing technology, but design of the required guide RNAs is more complex than for standard CRISPR-based nucleases or base editors. Here we describe PrimeDesign, a user-friendly, end-to-end web application and command-line tool for the design of PE experiments. PrimeDesign can be used for single and combination editing applications, as well as genome-wide and saturation mutagenesis screens.
Scalable characterization of the PAM requirements of CRISPR-Cas enzymes using HT-PAMDA
Abstract The continued expansion of the genome-editing toolbox necessitates methods to characterize important properties of CRISPR–Cas enzymes. One such property is the requirement for Cas proteins to recognize a protospacer-adjacent motif (PAM) in DNA target sites. The high-throughput PAM determination assay (HT-PAMDA) is a method that enables scalable characterization of the PAM preferences of different Cas proteins.

I don’t know what you do, I know what I do and it’s physics stuff. So, in physics we make graphs — like all the time. Position vs. time, electric current vs. voltage, and even velocity vs. position. We pretty much love them. But you know, like a good love — not like crazy love. I mean, it’s just a graph. Fine, but how do you make these? Let’s do it. How to make graphs in python. Python, Glowscript, and Matplotlib I’m going to use Glowscript. Yes, it’s still python — but it’s python with more.

No-Go on the Radio
What Not to Say by Susan K. Parson, FAA Safety Briefing Editor A few years ago, I occasionally (okay, regularly) found myself watching a now defunct and highly formulaic “reality” show called “What Not to Wear.” The fact that it appeared on The Learning Channel assuaged some of the I-shouldn’t-be-watching-this guilt (educational, right?). I also rationalized by saying I was learning about better fashion. Sort of. For those unfamiliar with the show, here’s the shtick.

Stop Using Square Bracket Notation to Get a Dictionary’s Value in Python
Go beyond the traditional method and safeguard your code Photo by Pixabay on Pexels A dictionary is an unordered set of terms and definitions. That means that: Each data point has an identifier (term) and a value (definition). The terms must be unique to that dictionary — no repetitions. The terms have no explicit order — unlike a list. To define a dictionary, use curly braces and separate each term/definition pair with a comma.

Left Atrial Appendage Electrical Isolation as a Target in Atrial Fibrillation
Although the benefit of catheter ablation of the pulmonary veins is well established in paroxysmal atrial fibrillation (AF), the optimal ablation strategy for persistent AF is yet to be determined. Pulmonary vein isolation in persistent AF often results in AF recurrence and the success rate may be low even after multiple procedures. Recurrent AF risk may be secondary to non–pulmonary vein triggers, and adjunctive therapies to better control persistent AF are under investigation.
External risks remain key threat to developed Asia-Pacific economies

LAA Occlusion in AF Patients Without Anticoagulants
Stroke is the most feared complication of atrial fibrillation (AF). Although oral anticoagulation with non-vitamin K antagonist and non-vitamin K antagonist oral anticoagulants (NOACs) have been established to significantly reduce risk of stroke, real-world use of these agents are often suboptimal due to concerns for adverse events including bleeding from both patients and clinicians. Particularly in patients with previous serious bleeding, oral anticoagulation may be contraindicated.
