USP24-dependent STAT2 stabilization mediates physiologic and pathologic bone formation
Abstract Osteoblast development must be precisely regulated, as insufficient bone formation results in low bone mass and skeletal fragility, whereas excessive osteogenesis drives heterotopic ossification (HO), the ectopic formation of bone in soft tissues. Here, we identify the deubiquitinating enzyme ubiquitin-specific peptidase 24 (USP24) as a key regulator of both physiological and pathological ossification.

Notably, IleuTPS features a relatively unusual E 366 IDD motif. Given this alteration of the prototypical catalytic acid motif, the role of E366 in IleuTPS was investigated, even though a previous report indicated that substitution of glutamate for the first aspartate does not alter product output in at least one DTC. (14) Thus, an E366D variant was constructed and subjected to ex vivo analysis ( Table S2 ).
The One Ring: A Monocycle Producing Class II Diterpene Cyclase from Isodon leucophyllus Click to copy article link Article link copied!
Abstract Click to copy section linkSection link copied! Class II diterpene cyclases (DTCs) have long been recognized to mediate distal cyclization of geranylgeranyl pyrophosphate (GGPP) into bicyclic labdane-related products through an acid–base catalytic mechanism, leaving the pyrophosphate ‘head’ group intact. Although distal monocyclic diterpenoids naturally occur, these are historically attributed to tetraterpenoid carotenoid degradation rather than direct enzymatic cyclization.
Long-term comparative analysis of AAV9-mediated gene replacement therapies for spinal muscular atrophy in mice
|
Abstract Spinal muscular atrophy (SMA) results from a deficiency of the survival motor neuron (SMN) protein. Zolgensma, an adeno-associated virus (AAV)-based SMN1 gene-replacement therapy, is approved for SMA, though its long-term efficacy and safety remain uncertain. This study compares a Zolgensma-like benchmark vector with a 2nd-generation vector featuring a codon-optimized SMN1 transgene under the control of an endogenous SMN1 promoter.

In vivo base editing rescues liver pathophysiology and peroxisome dysfunction in a mouse model of Zellweger spectrum disorder
|
Abstract Zellweger spectrum disorder (ZSD) is caused by biallelic loss-of-function variants in PEX genes required for peroxisome biogenesis, which is critical for normal cellular metabolism and signalling. The PEX1-p.G843D (c.2528G>A) allele, present in approximately 30% of individuals with ZSD, frequently results in chronic liver disease that can progress to cirrhosis, hepatocellular carcinoma and degraded neurological health.

Since then, investor attention to environmental, social, and governance (ESG) factors has grown rapidly.
Exploring a Unique Class II Diterpene Cyclase: The Modified Catalytic Acid Motif Contributes to Ring Contraction in Premutilin Synthase
For decades, semisynthetic derivatives of the fungal diterpenoid pleuromutilin have demonstrated potent clinical efficacy in both veterinary and human medicine, and are distinguished by their low rates of cross-resistance. (1) The complex tricyclic “propellane-like” hydrocarbon backbone of pleuromutilin originates from its progenitor, premutilin, which is formed by cyclization of general diterpenoid precursor (E,E,E)-geranyl-geranyl pyro-phosphate (GGPP, 1).
Environmental, Social, and Governance Factors as Tools for Improving Market Efficiency: A Study on Equity Misvaluation
5.1.2.1 ESG Performance and Misvaluation Deviation Our global study extends signaling theory to ESG performance and equity misvaluation. We find that strong ESG performance serves as a credible signal of good governance and stakeholder alignment, supporting the theories of Armstrong et al. (2010) and Freeman (1984). The results align with costly signaling theory, suggesting that high-quality ESG performers more effectively signal their value.
