The new frontier of cancer neuroscience
Growing evidence that the nervous system can influence tumour behaviour highlights the need to better understand the biological basis and consequences of interactions between cancer cells and the host, both to elucidate mechanisms of disease progression and to identify new therapeutic opportunities (Nature 2023;618:467–479). However, the neuroscience of cancer remains comparatively underexplored today.
The next 25 years of cancer research: emerging perspectives and priorities
Over the past 25 years, cancer research has undergone profound conceptual and technological advances, yet major challenges remain. As Nature Reviews Cancer marks its 25th anniversary, we turn our attention to the future of the field. Believing that emerging investigators are uniquely positioned to challenge prevailing assumptions, adopt new interdisciplinary approaches and redefine research priorities, we asked them to share how they envision the future of cancer research.
Nociceptive innervation limits tertiary lymphoid structures to promote lung cancer
|
Results We first performed a stage-wise characterization of the innervation landscape in a GEMM of LUAD (Ad5-Spc-Cre, KrasLSL-G12D/+, Trp53fl/fl; Rosa26LSL-tdTomato [KP] model), which recapitulates the LUAD tumorigenesis process observed in patients.26 Staining for the pan-neuronal marker β3-tubulin revealed that pulmonary innervation in the normal lung is mainly concentrated in the large airways (Figures 1A and S1A).

The CCL2-CCR2 astrocyte-cancer cell axis in tumor extravasation at the brain
Abstract Although brain metastases are common in cancer patients, little is known about the mechanisms of cancer extravasation across the blood-brain barrier (BBB), a key step in the metastatic cascade that regulates the entry of cancer cells into the brain parenchyma. Here, we show, in a three-dimensional in vitro BBB microvascular model, that astrocytes promote cancer cell transmigration via their secretion of C-C motif chemokine ligand 2 (CCL2).
Identification of DHODH as a therapeutic target in small cell lung cancer
A big step against small cell cancer Small cell lung cancer is a particularly aggressive tumor subtype, for which no specific targeted treatments are available. Li et al. used a CRISPR screening approach to identify a metabolic vulnerability and determine that small cell lung cancer cells are particularly reliant on the pyrimidine biosynthesis pathway and therefore sensitive to its disruption.
GKAP Acts as a Genetic Modulator of NMDAR Signaling to Govern Invasive Tumor Growth
Dlgap1 Is the Most Differentially Expressed Candidate Modifier Gene between the Invasive B6 and Non-invasive C3H Backgrounds Cancer Cells from the Invasive B6 Background Have Higher NMDAR Pathway Activity than Those from the Non-invasive C3H Background GKAP Expression Is Associated with Differential NMDAR Activity and Sensitivity to Pharmacological Inhibition In Vivo GKAP Knockdown Phenocopies the “Low NMDAR Activity” Status and Reduces Invasiveness of mPanNET Cells In Vitro Gene Expression...
