1 Introduction Spirocyclic molecules are highly valued in medicinal chemistry due to their unique structural characteristics. The rigid 3D structure imposed by the spirocycle can enhance the molecule binding affinity to biological targets.[1] In particular, the spirolactone moiety is a structural motif that can be found in a wide array of natural and unnatural compounds with biological activities.
