Prospective multicenter study of ctDNA versus tumor tissue guiding FGFR-targeted therapy in metastatic urothelial cancer - Nature Communications
|
Abstract Fibroblast growth factor receptor (FGFR) alterations are targetable in metastatic urothelial carcinoma (mUC). Identifying FGFR alterations currently requires tissue testing, which is limited by sample availability and cancer heterogeneity. Plasma circulating tumor DNA (ctDNA) offers a complementary testing strategy, but the added value of ctDNA relative to conventional FGFR alteration assessment remains unclear.

Canadian Prostate Cancer Trends in the Context of PSA Screening Guideline Changes
1. Introduction Prostate cancer is the most common cancer in Canadian men aside from non-melanoma skin cancers. A projected 27,900 cases were expected to be diagnosed in Canada in 2024, accounting for 22% of all cancers among males [1]. Prostate cancer is the third leading cause of cancer deaths in Canadian men, ranking only behind lung and colorectal cancers, and is responsible for 11% of all cancer deaths in men. The treatments for prostate cancer have improved over time (Figure 1).

Multiomics and cellular senescence profiling of aging human skeletal muscle uncovers Maraviroc as a senotherapeutic approach for sarcopenia - Nature Communications
Abstract Cellular senescence is a hallmark of organismal aging but how it drives aging in human tissues is not fully understood. Here we leverage single nucleus multiomics to profile senescence in mononucleated cells of human skeletal muscle and provide the first senescence atlas. We demonstrate the intra- and inter-populational transcriptomic and epigenomic heterogeneity and dynamics of cellular senescence.

Two case reports of eosinophilic fasciitis with onset after immune checkpoint inhibitor cessation
Abstract Immune checkpoint inhibitors (ICIs) are known to cause a wide spectrum of immune-related adverse events (irAEs). Among these, eosinophilic fasciitis (EF) is a rare, fibrosing disorder causing inflammatory infiltration of subcutaneous fat and fascia. It is characterized clinically by edema and subsequent induration and tightening of the skin and subcutaneous tissues. Several case reports have documented EF secondary to ICIs in patients on active treatment.

3 Steps to Improved Cardiac Health
Firefighting has come a long way in the past few decades. Advances in technology, apparatus, building construction and firefighter fitness have contributed to a more effective and efficient fire service. The days of charcoal breathing canisters and Browder Life Nets have given way to state-of-the-art breathing apparatus and thermal imaging cameras. Yet despite these advances, there remains one factor we haven’t mastered that continues to fatally plague our personnel: heart attacks.
A randomised trial of 4- versus 12-weekly administration of bone-targeted agents in patients with bone metastases from breast or castration-resistant prostate cancer
Highlights • Four- versus 12-weekly therapy of bone-targeting agents in patients with bone metastases from breast or prostate cancer. • BTAs used in the study are pamidronate, zoledronate and denosumab. • Twelve-weekly therapy was non-inferior to 4-weekly therapy for the primary outcome of health-related quality of life. • No significant difference was seen in symptomatic skeletal event (SSE) rates and SSE-free survival between arms.
Evolution in upfront treatment strategies for metastatic RCC
The treatment paradigm for treatment-naive metastatic clear cell renal cell carcinoma continued to evolve in 2019 as three phase III randomized controlled trials were reported comparing sunitinib with combination immune checkpoint inhibitors and VEGF-targeted therapy. Pembrolizumab plus axitinib and avelumab plus axitinib became new treatment options for untreated patients, irrespective of risk group or PD-L1 expression.

A new approach to evaluating the cost-efficiency of complex hedging strategies: an application to electricity price-volume quanto contracts
Abstract Electricity quanto contracts improve the efficiency of financial risk management by accommodating the correlated price and volumetric risks of energy suppliers. The use of electricity quanto contracts, however, suffers from the high risk premiums associated with such tailor-made and illiquid financial instruments. As a result, it is of great interest to properly evaluate the cost-efficiency of these complex hedging deals.

Promoting Sleep to Improve Delirium in the ICU
A night in the ICU is often characterized by a chaotic whirlwind of beeping machines, staff conversations, bright lights, medical interventions, and visits from care providers. Mix in mind-altering drugs, uncomfortable medical devices, an unfamiliar environment and pain, and sleep becomes markedly fragmented and devoid of the restorative stages considered vital for repair and recovery (1).

Effectiveness of Remote Patient Monitoring After Discharge of Hospitalized Patients With Heart Failure
Importance It remains unclear whether telemonitoring approaches provide benefits for patients with heart failure (HF) after hospitalization. Objective To evaluate the effectiveness of a care transition intervention using remote patient monitoring in reducing 180-day all-cause readmissions among a broad population of older adults hospitalized with HF.
National Differences in Trends for Heart Failure Hospitalizations by Sex and Race/Ethnicity
Methods and Results— The National Inpatient Sample (NIS) was used to estimate the crude and age-standardized rates of HF hospitalization between 2002 and 2013 by sex and race/ethnicity. Direct standardization was used to age-standardize rates to the 2000 US standard population. Relative differences between subgroups were reported. The national age-adjusted HF hospitalization rate decreased 30.8% from 526.86 to 364.66 per 100?000 between 2002 and 2013.
