Tumor mutations predict HER2-targeted therapy resistance in primary HER2-positive breast cancer
|
Abstract The presented study investigated the relevance of mutations in 17 cancer genes and response to neoadjuvant chemotherapy in two clinical cohorts of HER2+ breast cancer. 364 samples from HER2+ tumors of the neoadjuvant studies GeparTrio (no anti-HER2 treatment, n = 71) and GeparSepto (dual HER2 blockade and randomization for paclitaxel vs. nab-paclitaxel, n = 293) were analyzed by targeted next generation sequencing of hot spot regions of 17 genes.

Expanding adjuvant therapy with ribociclib in HR-positive, HER2-negative early breast cancer
Despite advances in adjuvant endocrine therapy (ET), residual recurrence risk remains in patients with hormone receptor (HR)–positive, HER2-negative early breast cancer (eBC).
Treatment decisions in post-neoadjuvant HER2-positive early breast cancer
Patients HER2-positive (HER2+) breast cancer (BC) who achieve pathological complete response (pCR) have significantly better event-free survival (EFS), yet pCR cannot overcome the prognostic impact of baseline tumour size and nodal status.

21 Ribociclib (RIB) + Nonsteroidal Aromatase Inhibitor (NSAI) as Adjuvant Treatment in Patients (pts) With HR+/HER2- Early Breast Cancer (EBC): Final Invasive Disease-Free Survival (iDFS) Analysis From the NATALEE Trial
Background Interim results from the phase 3 NATALEE trial showed that ribociclib added to standard-of-care adjuvant nonsteroidal aromatase inhibitor (NSAI) had a significant invasive disease-free survival (iDFS) benefit in patients with stage II/III HR+/HER2- early breast cancer (EBC) at risk of recurrence, including those with node-negative disease. We present the final protocol-specified analysis of iDFS (primary end point).
Palbociclib for Residual High-Risk Invasive HR-Positive and HER2-Negative Early Breast Cancer-The Penelope-B Trial
Patients with hormone receptor–positive (HR+), human epidermal growth factor receptor 2–negative (HER2−) primary breast cancer who receive chemotherapy as part of their primary treatment are at higher risk of relapse.1 About a third of the patients with residual invasive disease after neoadjuvant chemotherapy (NACT) will relapse despite adjuvant endocrine therapy (ET).2 The risk of relapse can be assessed more accurately using the clinical pathological staging-estrogen receptor grading...

Survival Analysis After Neoadjuvant Chemotherapy With Trastuzumab or Lapatinib in Patients With Human Epidermal Growth Factor Receptor 2-Positive Breast Cancer in the GeparQuinto (G5) Study (GBG 44): Journal of Clinical Oncology: Vol 0, No 0
Patients with HER2-positive tumors (n = 615) received neoadjuvant treatment with epirubicin (E) plus cyclophosphamide (C), followed by docetaxel (T) in combination with either lapatinib (L) or trastuzumab (H; ECH-TH arm: n = 307; ECL-TL arm: n = 308). All patients received adjuvant trastuzumab for a total of 12 months and 18 months in the ECH-TH and ECL-TL arms, respectively. Median follow-up was 55 months.
