CAR signaling instructs divergent metabolic reprogramming and functional fates in αβ and γδ T cells
Abstract γδ T-cell-based immunotherapies have become relevant as alternatives to conventional αβ T-cell products, with preclinical data demonstrating tumor burden reduction and the mitigation of tumor-induced tissue damage. Given that most CAR constructs have been optimized for αβ T cells, we hypothesized that distinct T-cell types may require tailored CAR architectures to achieve optimal function.

In-vivo dendrite injury drives local mitochondrial contraction and dendrite branching
Abstract Mitochondria regulate cellular homeostasis in development and disease, and mitochondrial morphology plays a role in local injury signaling and wound repair. How mitochondria respond during dendrite injury remains an open fundamental question. Here we show that mitochondria contract rapidly and locally after laser dendrotomy. In the proximal intact dendrite, the extent of mitochondrial contraction diminishes with increasing distance from the injury site.
SITC perspective: leveraging patient enrichment biomarkers to accelerate early phase IO drug development
Biomarker Biopsy Tumor Microenvironment Next generation sequencing - NGS Pathology The first wave of immunotherapy (IO)—especially immune checkpoint inhibitors (ICIs) and chimeric antigen receptor T-cell therapy (CAR-T)—transformed cancer care by delivering long-term clinical benefit and even cure to some patients.1 However, not all patients with cancer are eligible for these treatments, and primary or acquired drug resistance to current agents may occur.2 Novel IO drugs that overcome these...

Northwestern team performs liver transplant for terminal cancer patient
A team at Chicago-based Northwestern Medicine has performed a successful liver transplant in a patient with terminal colorectal cancer. The procedure was made possible through Northwestern’s new Colorectal Metastasis to Liver Extraction with Auxiliary Transplant and Delayed Resection program, or CLEAR, according to a March 18 news release from the health system. To make the procedure possible, the liver of a deceased donor split into two parts.
Baptist Health appoints leukemia chief
Baptist Health Miami Cancer Institute has appointed Firas El Chaer, MD, as chief of leukemia. He will also serve as medical director of infusion services, according to a March 18 news release from the health system. Before joining Baptist Health, Dr. El Chaer was the medical director of infusion centers for the cancer service line and medical director of the chronic and acute leukemia research team at Charlottesville-based University of Virginia Comprehensive Cancer Center.
The future of cancer care is outpatient: How leaders can prepare
Cancer isn’t just one disease, but many — each with its own challenges, characteristics, risks and treatments. As we have worked tirelessly to understand this group of diseases, I’ve witnessed a profound shift in how we approach cancer treatment. When I began treating advanced melanoma, patients had few treatment options outside of chemotherapy, surgery and radiation. Early novel therapeutics often required long hospital stays and carried significant toxicities.
Tumor-infiltrating lymphocytes: A new hope
Tumor-infiltrating lymphocytes (TILs) can be massively expanded from resected tumors and used as a cellular treatment for advanced malignancies. TILs require a preparative non-myeloablative chemotherapy followed by an abbreviated course of interleukin-2. Here, we review the historical development of TIL therapy and discuss potential solutions to ongoing roadblocks that may result in broader and improved efficacy for patients afflicted with treatment-refractory, advanced cancer.
Waun Ki Hong, legend and “mentor magnificus,” in his own words
If lifetime impact could be quantified by pages in The Cancer Letter, no person has been more revered than Waun Ki Hong: following Hong’s death in 2019, The Cancer Letter published no fewer than six obituaries. Now, you can hear his story in his own words. “Waun Ki Hong’s contributions to medicine changed the field as we know it,” wrote Fadlo R. Khuri, president of the American University of Beirut. Hong died Jan. 2, 2019. On Jan. 4, The Cancer Letter published three obituaries, but they kept coming.

Why bias blocks Asian Americans from top leadership positions
Recent weeks have seen crimes against Asians and Pacific Islanders make headlines across the U.S. From verbal and physical attacks, to murders and mass shootings, these heinous crimes are forcing us to address a persistent, pervasive and largely unspoken national problem. There is an inherent bias toward the API community in America. In many cases, it’s an unconscious bias, rooted in an ignorance of our cultures and diversity.

Indoleamine 2,3-Dioxygenase Production by Human Dendritic Cells Results in the Inhibition of T Cell Proliferation
Patrick Hwu, Mark X. Du, Réjean Lapointe, My Do, Milton W. Taylor and Howard A. Young J Immunol April 1, 2000, 164 (7) 3596-3599; DOI: https://doi.org/10.4049/jimmunol.164.7.3596 Abstract Dendritic cells (DCs) play a key role in the activation and regulation of B and T lymphocytes. Production of indoleamine 2,3-dioxygenase (IDO) by macrophages has recently been described to result in inhibition of T cell proliferation through tryptophan degradation.
CXCL9, CXCL10 and IFNγ favor the accumulation of infused T cells in tumors following IL-12 plus doxorubicin treatment
The infiltration of cytotoxic CD8+T cells suggests a positive prognosis in a number of established tumors. T cell therapy is rapidly growing in the past decade, and achieved big success in treating hematological tumors. Despite autologous TIL infusion made remarkable breakthroughs in treating refractory melanoma, the highest respond rates are only around 50%. Therefore, approaches which guide TILs to reach tumor sites more efficiently are to be developed.
