Saturation mutagenesis enables RAD51D clinical variant interpretation and identifies distinct biochemical functions
Abstract The tumor suppressor RAD51D is essential for homologous recombination (HR). Pathogenic variants in RAD51D are associated with breast and ovarian cancers. However, most clinical missense variants are of uncertain significance. Here, we report a multiplex assay of variant effect to test 6,888 RAD51D coding variants for loss-of-function.
CST complex promotes second-strand synthesis in break-induced replication
Abstract Break-induced DNA replication (BIR) is a highly mutagenic recombination pathway used by eukaryotic cells to repair single-ended DNA breaks, mediate mitotic DNA synthesis and promote telomerase-independent telomere maintenance in certain cancers. Leading-strand synthesis in BIR is mediated by a migrating D-loop driven by DNA polymerase δ and Pif1 helicase but the mechanism of second-strand synthesis has remained poorly understood.

Multifaceted roles of PDS5B in RAD51-dependent homology-directed DNA repair and replication fork protection
|
Abstract PDS5B (Precocious Dissociation of Sisters 5B) functions in sister chromatid cohesion and genome organization. Interestingly, PDS5B also associates with RAD51, the recombinase required for DNA damage repair by homologous recombination (HR) and the preservation of stressed DNA replication forks against nucleolytic attrition. We show that PDS5B binds dsDNA preferentially over ssDNA and that it enhances RAD51-mediated DNA strand exchange via the capture of dsDNA.

Structural insight into how RAD51 paralog exchange regulates RAD51 filament formation
|
Abstract Homologous recombination (HR) repairs DNA double-strand breaks and stabilizes stressed replication forks, and HR deficiency promotes genome instability and cancer. HR requires assembly of RAD51 nucleoprotein filaments on single-stranded DNA (ssDNA), a process regulated by the human RAD51 paralogs RAD51C, XRCC3, RAD51D and XRCC2.

Resolution of R-loops and transcription–replication conflicts by SETX–BRCA1–BARD1 complex
|
Abstract Senataxin (SETX), an RNA–DNA helicase, accumulates at transcription pause sites through the tumor suppressor BRCA1. Here, we provide mechanistic insight into how SETX–BRCA1 resolves transcription-associated R-loops to prevent deleterious outcomes. Specifically, we show that full-length SETX unwinds R-loops with broad specificity and that the complex of BRCA1 and its obligatory partner BARD1 binds R-loops and stimulates R-loop unwinding by SETX.

Cryo-EM structures of UBA6 reveal mechanisms of E1–E2 specificity and dual FAT10/ubiquitin thioester transfer - Nature Communications
Abstract UBA1 and UBA6 define parallel ubiquitin (Ub) activation systems that perform non-overlapping roles in Ub and ubiquitin-like protein (Ubl) signaling. Whereas UBA1 supports the canonical Ub pathway, UBA6 also activates the Ubl FAT10, linking Ub signaling to immune-regulated proteostasis. In addition to selective Ub/Ubl activation, UBA1 and UBA6 engage distinct sets of E2s, yet how these enzymes achieve selective E2 engagement has remained unclear.

Why is a Spanish soccer match set to be held in the US so controversial?
It’s something that is very familiar for American sports fans: Your favorite team in your favorite sport is sent to play a regular season game abroad. The NFL, NBA, MLB and NHL all regularly do this and, in some cases, are increasing the amount of games held overseas every year in an effort to expand the footprint of the sport, as well as for economic reasons. Now, European soccer is joining America’s superpower sports in attempting to host fixtures beyond its native shores.

Cryo-EM structures reveal the molecular mechanism of SUMO E1–E2 thioester transfer - Nature Structural & Molecular Biology
Abstract Post-translational modification of proteins by SUMO (small ubiquitin-like modifier) regulates fundamental cellular processes and occurs through the sequential interactions and activities of three enzymes: E1, E2 and E3. SUMO E1 activates SUMO in a two-step process involving adenylation and thioester bond formation, followed by transfer of SUMO to its dedicated E2 enzyme, UBC9. This process is termed E1–E2 thioester transfer (or transthioesterification).
