International Trip Planning
Dubrovnik approach Majuro Flying your own aircraft to foreign countries is especially challenging, yet an extremely rewarding experience. It is not difficult, just requires a bit more planning and thought. First thing is to decide where to go – a quick trip to Mexico or Canada, or an around-the-world adventure. You are only limited by your imagination, your heart, and of course your wallet.
What we learned from building a community-owned solar farm
Last week the Goulburn Community Solar farm was officially opened. It was the culmination of 12 years of planning, consultation, designing, and building, resulting in a 1.4MW solar farm with a 4MWh battery on the outskirts of the NSW town of Goulburn. More than 200 members of the Goulburn Community Energy Cooperative that owns the farm attended a joyous celebration of finally being able to generate energy into the grid.
A new MU-2 in the Caribbean
The front office Owning aircraft has been a large part of my life. I have previously owned either fully or in partnership: Beechcraft Travel Air, Beechcraft E55, Beechcraft Staggerwing, Beechcraft King Air 200, and Commander 690A. Also, two Mig 21s, but that’s another story. Ownership gives the freedom to travel when and where you like with no strings attached; of course, it also comes with the costs and sometimes headaches.
Dual Bcl-2/Bcl-xl inhibition via AZD0466 combines with immune checkpoint blockade to enhance anti-tumour activity - Cell Death & Disease
Abstract Small molecule inhibitors designed to specifically target oncogenic proteins have demonstrated potent anti-tumour activities due to direct effects on tumour cells survival and/or proliferation. However, the effects of these compounds on normal cells, specifically immune cells and their potential to impede or enhance anti-cancer immunotherapies has yet to be fully explored.
Flying Africa: A Backcountry Story
Entering the desert from Sossousvlie Under the wing of my rented 1967 Cessna 182, the canyon meanders through this spectacular and empty country in southern Namibia. Photos and story by Peter Fraser A dirt strip stretches out as nothing but wild animals and endless African landscapes expand to the horizon. Pushing the throttle toward the blue, the cloudless sky beckons. Gaining speed, a thirsty flat six is fed more nourishment. Pull back until the lightness of the nose wheel breaks firmament.
A CDK11-dependent RNA polymerase II pause-checkpoint precedes CDK9-mediated transition to transcriptional elongation

Journal Entries: Flying Around South America
Bariloche view of the lake and Andes mountains from our room To be able to do a trip like this is a true gift, only available to those of us lucky enough to own and fly airplanes. Time to start dreaming, planning, then just go! Some songs just make you want to get up and go – and so it was with Frank Sinatra’s “Come Fly with me” ringing in my ears as we headed off for a memorable three-month swing through the Caribbean, South, and Central America.
p53 rapidly restructures 3D chromatin organization to trigger a transcriptional response - Nature Communications
Abstract Activation of the p53 tumor suppressor triggers a transcriptional program to control cellular response to stress. However, the molecular mechanisms by which p53 controls gene transcription are not completely understood. Here, we uncover the critical role of spatio-temporal genome architecture in this process.
Single cell Hi-C identifies plastic chromosome conformations underlying the gastrulation enhancer landscape - Nature Communications
Abstract Embryonic development involves massive proliferation and differentiation of cell lineages. This must be supported by chromosome replication and epigenetic reprogramming, but how proliferation and cell fate acquisition are balanced in this process is not well understood. Here we use single cell Hi-C to map chromosomal conformations in post-gastrulation mouse embryo cells and study their distributions and correlations with matching embryonic transcriptional atlases.
Beavertown Brewery launches creative collaboration with Queens Of The Stone Age

Check it out: The collaboration between Beavertown Brewery and Queens of the Stone Age combines musical artistry with unique illustrations, bringing to life a surrealist visual world in the form of a music video.
DeepLoop robustly maps chromatin interactions from sparse allele-resolved or single-cell Hi-C data at kilobase resolution - Nature Genetics
Abstract Mapping chromatin loops from noisy Hi-C heatmaps remains a major challenge. Here we present DeepLoop, which performs rigorous bias correction followed by deep-learning-based signal enhancement for robust chromatin interaction mapping from low-depth Hi-C data. DeepLoop enables loop-resolution, single-cell Hi-C analysis. It also achieves a cross-platform convergence between different Hi-C protocols and micrococcal nuclease (micro-C).
Fonterra great? It can barely afford to be ordinary
In examining Fonterra’s latest capital restructuring proposal, Peter Fraser asks whether it is a case of ‘third time lucky‘ or has Fanshaw Street latched onto something else entirely? Fonterra. Outside of largely rural gatherings of the cooperative faithful, the very name is likely to elicit a groan. This is not surprising. Fonterra is like that kid from the good family – the one who got all the chances – yet never lived up to the wunderkind promises.
Heartland confounds the doubters with profit upgrade
It's always a dangerous thing to bet against a company with an established track record of doing what it says it will do.Last December, Forsyth Barr analysts clearly didn't believe the message Heartland Group gave the annual shareholders' meeting that the company was tracking ahead of expectations.Heartland has just proven them wrong in the best possible way; it raised its full-year earnings guidance after reporting it had 75% or more of the higher annual forecast under its belt from the...
Preno wants to help hotels fly autopilot
A hotel management software company with over 500 customers in 25 countries is seeing a rise in direct hotel bookings in New Zealand post-lockdown.Amelia Gain, the co-founder and chief executive of software company Preno, told BusinessDesk more New Zealanders are shunning sites like Booking.com than they were before covid-19.“Guests are becoming more aware of the commission that Booking.com takes, so [they are] also trying to book direct with properties as much as they can.

It's Not Too Late to Plant a Summer Victory Garden
This summer's not going to look like what we thought. We'll be working at home, making and eating more meals at home, and perhaps even taking "staycations" at home. But there's one place we can all safely go to get away from the daily grind: our yards. One throwback idea that's currently bubbling up in backyards from Maine to Mississippi? The Victory garden.
âDonald Trump Cut From Home Alone 2 Broadcast
Peter Fraser in Entertainment Last updated 11:49 AM, Thursday December 26 2019 GMT Never mind impeachment, US President Donald Trump has really been hit where it hurts this Christmas after apparently being chopped from a version of Home Alone 2. The POTUS famously made a blink-and-you-miss-it cameo in the Christmas classic movie from 1992 but Canadian broadcaster CBC cut him out. Check out his appearance below.
âDon't F**k With Cats Viewers Think Documentary Left Some Unanswered Questions
Peter Fraser in Entertainment Last updated 1:28 PM, Thursday December 26 2019 GMT Don't F**k With Cats: Hunting An Internet Killer is Netflix's most recent hit but fans are claiming that the harrowing documentary has left some unanswered questions. The gruesome show (check it out in the trailer above if you're not easily shocked) is about Luka Magnotta and the amateur detective work carried out to catch the Canadian murderer after he posted a video of himself killing two kittens online.
Hi-C as a tool for precise detection and characterisation of chromosomal rearrangements and copy number variation in human tumours
Abstract Chromosomal rearrangements occur constitutionally in the general population and somatically in the majority of cancers. Detection of balanced rearrangements, such as reciprocal translocations and inversions, is troublesome, which is particularly detrimental in oncology where rearrangements play diagnostic and prognostic roles.
More of a good thing - is surplus renewable electricity an opportunity for early decarbonisation?
We are entering a world where renewables will make up an increasing share of our electricity supply –the electricity sector was the leading sector for energy investment in 2018, the third year in a row that this has occurred. This trend is set to continue. In WEO 2018’s New Policies Scenario, 21% of global electricity production is projected to come from variable renewables by 2040, up from 7% in 2018, supported by about $5.3 trillion of investment. The EU share is even higher at around 39%.
Capturing Three-Dimensional Genome Organization in Individual Cells by Single-Cell Hi-C
Protocol First Online: 13 September 2017 Part of the Methods in Molecular Biology book series (MIMB, volume 1654) Abstract Hi-C is a powerful method to investigate genome-wide, higher-order chromatin and chromosome conformations averaged from a population of cells. To expand the potential of Hi-C for single-cell analysis, we developed single-cell Hi-C.
âLiverpool Win The Champions League After Beating Tottenham
LADbible http://www.ladbible.com/assets/images/theme/logo-blk.png 35 seconds ago 0 Shares Liverpool have won the Champions League after beating Tottenham Hotspur 2-0 in Saturday night's final in Madrid. Mohamed Salah's penalty in just the second minute and Divock Origi's late goal settled what was, let's be honest, a naff game between the two Premier League clubs in Spain's capital.
Long-range enhancer–promoter contacts in gene expression control - Nature Reviews Genetics
Abstract Spatiotemporal gene expression programmes are orchestrated by transcriptional enhancers, which are key regulatory DNA elements that engage in physical contacts with their target-gene promoters, often bridging considerable genomic distances. Recent progress in genomics, genome editing and microscopy methodologies have enabled the genome-wide mapping of enhancer–promoter contacts and their functional dissection.
Sparrow (Passer Domesticus)
fish. Then we were in a shopping arcade, the man started to gut the fish. I had a go, yuk it wasn't my kind of thing. I remember the inside of the fish being a nice flower or star shape when it's head was cut off. I kept looking at this shape on and off. I really didn't want to be doing the gutting. People came by and bought pilchards as we worked. I just wanted out of that job and was pleased when my boss asked me to go and get him a drink. I brought coffee and a very expensive apple drink.
Promoter Capture Hi-C: High-resolution, Genome-wide Profiling of Promoter Interactions
Promoter Capture Hi-C is designed to specifically enrich Hi-C libraries for interactions involving promoters. These interactions comprise only a subset of ligation products present in a Hi-C library. Capture Hi-C can easily be modified to enrich Hi-C libraries for any genomic region or regions of interest by changing the capture system.

'Call Of Duty: Modern Warfare 2' Voted Best COD Game
Let's start with a simple disclaimer - there's no such thing as an objectively correct list. People have different tastes in different things, whether that's music, film, art or computer games. Except when it comes to computer games, there's a pretty universal opinion that Call Of Duty: Black Ops isn't very good. It's not that it's awful, but it's just not that great, compared to the other games in the franchise.

Sixty years of genome biology
In 1977, Rich Roberts and Phil Sharp independently reported that genes can be interrupted by introns [6,7]. In my opinion, this is the biggest and most surprising discovery in genome biology since the structure of DNA was reported by Watson and Crick on 25 April 1953 [1]. Accordingly, Roberts and Sharp were awarded the Nobel Prize in Physiology or Medicine in 1993 for this discovery.
