Single-cell spatiotemporal dissection of the human maternal–fetal interface
Abstract The human maternal–fetal interface is characterized by mosaic intermingling of maternal and fetal cells1. Yet the underlying cellular, molecular and spatial programmes remain incompletely defined.
KMT2C/KMT2D-dependent H3K4me1 mediates changes in DNA replication timing and origin activity during a cell fate transition
Results To understand how RT is regulated during cellular differentiation, we used a previously established ESC model following the transition from the naive to the EpiLC state reflecting the physiologically relevant cell fate transition of the embryonic epiblast during the peri-implantation period of mammalian development (mouse embryonic day [E]4.5–E5.5).24,25,26,27 This EpiLC state is consistent with the recently hypothesized formative pluripotency state, which immediately precedes...
Coordinate post-transcriptional regulation by microRNAs and RNA binding proteins is critical for early embryonic cell fate decisions
Abstract Post-transcriptional control by RNA binding proteins (RBPs) and microRNAs play central roles in mRNA stability and translation (1). However, little is known about how RBPs and microRNAs coordinate in developmental time to regulate cell fate. Here, we show that coordinate RBP and microRNA control of a single transcript, Profilin 2 (Pfn2), is essential for differentiation of embryonic stem cells (ESCs) into the primary germ layer lineages.
MLL3/MLL4 enzymatic activity shapes DNA replication timing
Abstract Mammalian genomes are replicated in a precise order during S phase, which is cell-type-specific and correlates with local transcriptional activity, chromatin modifications and chromatin architecture. However, the causal relationships between these features and the key regulators of DNA replication timing (RT) are largely unknown.
The roles of microRNAs in mouse development - Nature Reviews Genetics
Abstract Hundreds of microRNAs (miRNAs) are expressed in distinct spatial and temporal patterns during embryonic and postnatal mouse development. The loss of all miRNAs through the deletion of critical miRNA biogenesis factors results in early lethality. The function of each miRNA stems from their cumulative negative regulation of multiple mRNA targets expressed in a particular cell type. During development, miRNAs often coordinate the timing and direction of cell fate transitions.
MicroRNA-dependent inhibition of PFN2 orchestrates ERK activation and pluripotent state transitions by regulating endocytosis
Viruses harbored by bats and rodents, considered high-risk reservoirs, are no more likely to infect humans than viruses carried by other hosts, a study suggests.
