Fibroblast growth factor receptor inhibition for succinate dehydrogenase-deficient gastrointestinal stromal tumors: a phase 2 trial
|
Abstract While most gastrointestinal stromal tumors are driven by oncogenic mutations in KIT or PDGFRA, 10–15% exhibit functional loss of the succinate dehydrogenase (SDH) complex and genome-wide DNA hypermethylation. Excess methylation in SDH-deficient gastrointestinal stromal tumors disrupts genomic insulators, inducing aberrant expression of oncogenic ligands FGF3, FGF4, and activating an autocrine signaling loop mediated through FGFR1.

Balancing Local Control With Long-Term Cosmesis in Re-Irradiation
In the management of patients with locoregionally advanced breast cancer, the clinical mandate often shifts toward prioritizing local control, sometimes at the expense of long-term cosmesis. However, for survivors, the chronic sequelae of aggressive retreatment, particularly debilitating skin changes and fibrosis, can profoundly impact quality of life years after the final fraction is delivered. During the 2026 American College of Radiation Oncology (ACRO) Summit, CancerNetwork® sat down with J.
CAR-T 2.0: Expanding Access, Speed, and Precision
Following the 2026 Tandem Meetings, Yan Leyfman, MD, from NewYork–Presbyterian Hospital and a hematologic malignancies board member for the journal ONCOLOGY®, outlined key advances that emerged from the conference. He noted that this year’s data reflect a critical transition from early proof-of-concept innovation to real-world refinement in safety, access, manufacturing, and durability of cellular therapies.
Redefining Frontline Therapy in Undifferentiated Pleomorphic Sarcoma
For clinicians treating undifferentiated pleomorphic sarcoma (UPS), the integration of immune checkpoint inhibitors remains one of the most debated paths in sarcoma. While the high mutational burden and inflammatory profiles of these tumors make them prime candidates for immunotherapy, the clinical community is currently split: some practitioners adopt pembrolizumab early based on anecdotal success, while others reserve it strictly for the refractory setting due to a lack of randomized data.
Advances in Immunotherapy for Breast Cancer
Introduction Breast cancer is the most commonly diagnosed malignancy in women, with more than 2.3 million new cases in 2022.1 Historically, breast cancer was treated with cytotoxic chemotherapy and was initially thought to be less responsive to immunologic agents due to factors creating a poorly immunogenic tumor environment, such as lower tumor mutational burden (TMB) and fewer tumor-infiltrating lymphocytes (TILs).2 However, as cancer treatment has evolved, there has been increased interest...

Seth Pollack, MD, on Overcoming T-cell Trafficking Barriers in Sarcoma
In the landscape of soft tissue sarcomas, the "immune-cold" tumor microenvironment remains one of the most significant hurdles to durable immunotherapy responses. For clinicians treating subtypes like synovial sarcoma, the challenge isn’t just activating T cells but ensuring they can effectively traffic into a hostile or indifferent tumor niche. In this discussion, Seth M. Pollack, MD, Steven T.

Tissue samples from 156 patients were included in the study. Tumor samples were all reviewed by two pathologists (JG and FO) and tumors were only included if there was full concordance in their assessment. Concordance was seen in 153 samples (99%) and only these samples were included for further analysis.

NBA Twitter is dying but we still have the friends we made along the way
Photo by Jakub Porzycki/NurPhoto via Getty Images I’ve been thinking about Twitter lately. Maybe you have too since giant manbaby Elon Musk closed his (heavily leveraged with Saudi money) $40 billion acquisition of the social media platform. I am not at all qualified to accurately predict what will happen to the company as it faces mountains of debt and huge staff layoffs.
The pathetic response to Kyrie Irving’s antisemitic lies blasted by Charles Barkley and Reggies Miller
Photo by Jesse D. Garrabrant/NBAE via Getty Images Charles Barkley, speaking on Tuesday night’s TNT ‘Inside the NBA’ broadcast, did not mince words when commenting on the on-going Kyrie Irving situation (detailed here), “I can’t believe that we ain’t talking about basketball. We’re talking about this idiot.” While calling Brooklyn Nets guard Kyrie Irving an idiot is the pithy one-liner, his most effective fire was directed at NBA commissioner Adam Silver.

Combination Doxorubicin and Pembrolizumab in Patients With Advanced Anthracycline-Naive Sarcoma-Reply
In Reply We thank Drs Baa and Rastogi for their comments regarding our recent Brief Report.1 First, they commented on the starting dose of 45 mg/m2 of doxorubicin in the phase 1 component and the 3 + 3 design, correctly pointing out that a more traditional approach would have been to dose escalate the novel component to a fixed-dose standard of care. However, pembrolizumab lacks a traditional dose-response/toxic effects curve. Dosing is based on translational models and measurement.
Heat vs Pelicans recap: Miami sets record with impressive win over New Orleans
The Miami Heat love the big stage and playing on Christmas Day is as big as it gets for the NBA. Almost. I can think of a bigger stage and that’s the one the Heat are trying to reclaim this summer. We’re a long way from the NBA Finals but as the cliche says, all journeys start with one step and this game against the talented young New Orleans Pelicans presented an early obstacle to the Heat goal.

What to expect during the NBA’s return, in a 5-minute read
The NBA is almost officially back, kicking off its season on July 30. Teams have been playing in scrimmages over the last week and we are already drooling over Bol Bol balling (nods to the Pun Master Prada) and shaking our heads at Lou Williams “dining” decisions. This is your standard mix of on-court small sample size overreaction and off-court drama stories providing some measure of normalcy in these un-normal times.
14 things to know about the NBA’s return

NBA trades 2019: The important stuff that did (and didn’t) happen at the deadline
One big thing didn’t happen, but a few other important things did. Here’s everything you need to know. By Mike Prada, Matt Ellentuck, Kristian Winfield, and Seth Pollack Feb 7, 2019, 4:19pm EST The 2019 NBA trade deadline is now in the books. As there often is, there was a flurry of late activity, featuring a combination of actual significant moves, rearranging of deck chairs, and important non-deals. But even compared to past years, 2019 trade season was a chaotic, beautiful mess.
Amare Stoudemire to Golden State Warriors For Corey Maggette?
There was a time that I might have been hesitant to post such a thinly sourced rumor but if Chad Ford can drop random references to "locker room tension" on ESPN.com then really, are we doing anything more than following the leader? No seriously, we still have our standards. This "rumor" comes from an anonymous source on a message board, but he/she has proven over the years to have gotten some things right.
Robin Lopez Expects To Play In Spurs Series
Robin Lopez was in a much better mood today. Gone were scary scowls and one-word answers and in their place was a much more engaging and happier big man. After reports just yesterday listed him as doubtful for this series, today Robin was much more optimistic. "I definitely think that I will be able to put in some time this series. What game I will first come in, I'm not sure yet. I'm going to keep working in practice and we'll see how I feel," Lopez said.
NBA Mock Draft: L.A. Lakers Select Aaron Holiday
You could have made the argument for a few players at this spot, like Jacob Evans, whose defensive versatility and shooting would have made him a great fit on this Lakers squad, but I decided to take the best player available and in my opinion, that was Aaron Holiday. After drafting Lonzo Ball in last year’s draft, the Lakers finally have a long-term answer at the point guard position. However, they have little depth behind Ball, which was a weakness for this team last season.

Novel therapeutic approaches in chondrosarcoma
Chondrosarcoma is a malignant tumor of bones, characterized by the production of cartilage matrix. Due to lack of effective treatment for advanced disease, the clinical management of chondrosarcomas is exceptionally challenging. Current research focuses on elucidating the molecular events underlying the pathogenesis of this rare bone malignancy, with the goal of developing new molecularly targeted therapies.
Emerging Targeted and Immune-Based Therapies in Sarcoma
Soft tissue and bone sarcomas are malignancies of mesenchymal origin, and more than 50 subtypes are defined. For most sarcomas, locally advanced or unresectable disease is still treated with cytotoxic chemotherapy. Recently, our understanding of subtype-specific cancer biology has expanded, and it has revealed distinct molecular alterations responsible for tumor initiation and progression.
