Natural product-derived azaphilones are selective scaffolds for covalent lysine liganding in live cells
Abstract Lysine-directed covalent liganding offers a promising strategy to expand the covalently ligandable proteome beyond cysteine. However, broadly applicable and selective lysine-reactive scaffolds remain limited. Here, we designed SCL-alkyne, a simplified azaphilone probe derived from the natural product sclerotiorin (SCL), for the global profiling of lysine reactivity in bacteria. SCL-alkyne displayed high intrinsic selectivity for lysine in vitro and in live Escherichia coli cells.
Prediction of tracheal intubation in patients with Pierre Robin sequence: development of a machine learning-based model
Abstract Patients with Pierre Robin sequence (PRS) frequently require multiple attempts for airway stabilization. However, there is currently no standardized algorithm to identify anatomical landmarks that predict the difficulty of tracheal intubation and its clinical impact.

A Biweekly GLP-1R Agonist Designed With Drug-Free Intervals for Enhanced Efficacy, Receptor Homeostasis and Gastrointestinal Tolerability
1 Introduction Glucagon-like peptide-1 receptor agonists (GLP-1RAs) have revolutionized the therapeutic landscape for type 2 diabetes and obesity [1]. Semaglutide, a paradigm-shifting GLP-1RA, exemplifies this success by achieving a prolonged half-life of 6.3 days in human through targeted structural modifications—notably, fatty acid side chain conjugation—while maintaining high receptor potency (EC50 = 6.2 ± 0.6 pM) [2, 3].
Benchmarking of pathology foundation models for assessing HER2 amplification and CEP17 polysomy in breast cancer
Abstract Accurate assessment of Human Epidermal Growth Factor Receptor 2 (HER2) amplification and Chromosome 17 (CEP17) copy number is critical for breast cancer treatment, yet diagnostically equivocal (IHC 2+) cases remain a clinical bottleneck. This study systematically benchmarks 14 feature encoders, including 12 Pathology Foundation Models (PFMs), using 2393 whole slide images across four diverse cohorts.
Hyper-O-GlcNAcylation destabilizes ARC to unleash NLRP3-mediated pyroptosis in diabetic cardiomyopathy
Abstract Diabetic cardiomyopathy (DCM) is a severe complication of diabetes characterized by myocardial dysfunction and inflammatory cell death. While apoptosis repressor with caspase recruitment domain (ARC) is a known inhibitor of apoptosis, its potential role in regulating pyroptosis, a critical driver of diabetic cardiac injury-remains unexplored.
Dual branch negotiation model for robust water resource monitoring with spectral ambiguity and boundary degradation
Abstract Despite the remarkable progress in remote sensing water-body segmentation, existing methods still struggle to jointly preserve fine boundary fidelity and achieve robust semantic disambiguation under cross-domain shifts, undermining applications like flood risk assessment and urban water monitoring. This limitation largely stems from purely data-driven fitting with little explicit control over aliasing artifacts and spectrally induced physical ambiguities.
A N-methylated antimicrobial peptide targets the fructose transporter FruA of Staphylococcus aureus
Abstract Antimicrobial peptides (AMPs) represent promising alternatives to conventional antibiotics, but their therapeutic application is often hindered by suboptimal stability and poorly defined mechanisms of action. To overcome these limitations, we engineered a peptide named CAMP502NC3 through chemical modification of its parent peptide, CAMP502, which originates from marine biofilm microorganisms.

Plasma Proteome-Wide Mendelian Randomization with Multi-Layer Triangulation Identifies Druggable Targets for Uterine Leiomyoma
Introduction Uterine leiomyomas (fibroids) are benign monoclonal smooth-muscle tumors of the myometrium that affect up to 70% of women by the age of 50 and are the single leading indication for hysterectomy.1,2 They cause heavy menstrual bleeding, pelvic pain, bulk symptoms, subfertility and pregnancy complications, and impose a substantial economic and quality-of-life burden.

β-Arrestin condensates regulate G-protein-coupled receptor function
|
Abstract β-Arrestins 1 and 2 are multifunctional adaptor proteins1 that regulate the signalling of G-protein-coupled receptors (GPCRs), the largest class of receptors, which impact nearly all aspects of physiology and are one of the most common drug targets2. Although β-arrestins interact with a wide array of signalling effectors at many GPCRs, it is unclear how β-arrestins promote such varied functions.

Assessment and dynamic characteristics of herbaceous vegetation condition in steppe regions of the forest-steppe ecotone in Northeastern China
Abstract Introduction: The Forest-Steppe Ecotone (FSE) is a highly heterogeneous and climate-sensitive transition zone facing intensified degradation due to increasing disturbances. Conventional single-index remote sensing methods often fail to adequately capture the complex, multi-dimensional ecological dynamics of these sensitive ecotones. This study investigated the Baiyin Aobao Forest Farm, an FSE in Northeastern China, to develop a more robust monitoring method.

Regulating Interfacial Dynamic Self‐Adjusting Hydrophobic Layer via Cationic Molecular Structure for Ultra‐Stable Zinc Metal Anode
The structure of solution additives significantly affects the polarization and dendrite behavior of metal batteries. In this study, we propose, for the first time, the “hydrophilic-lipophilic balance (HLB) value-spatial configuration synergistic regulation” strategy by selecting cations with different structures as additives, successfully revealing the structure-activity relationship between additive structures and the stability of the anode interface adsorption layer.

A crack detection and quantification framework for high‐resolution images using Mamba and unmanned devices
1 INTRODUCTION Structural defects detection is essential for assessing the safety of civil infrastructure, aiming to evaluate structural integrity and service life. Cracks, as one of the structural defects, are also a primary focus (Cha et al., 2024; Ferraris et al., 2023). The occurrence of cracks often signals the deterioration of structural performance and potential safety risks (Ahila Priyadharshini et al., 2023).
