Results To avoid confounding from chromosomal abnormalities in prior PMSE patient iPSC lines (Dang et al., 2021), we used CRISPR/Cas9 to generate STRADA loss-of-function KO lines with compound heterozygous truncating variants and isogenic controls (Figures S1A and S1B). Lines expressed pluripotency markers (Figure S1C) and showed no karyotypic abnormalities (data not shown). Two STRADA KO (M1 and M2) and two isogenic control (C1 and C2) lines were used for all experiments.

1. Introduction Herpes Simplex Virus type 1 (HSV-1) is a double-stranded DNA (~152 Kb) virus belonging to the Alphaherpesvirinae subfamily of the Orthoherpesviridae family [1,2]. HSV-1 infects humans and causes a variety of clinical manifestations, including neonatal herpes, corneal blindness, herpetic whitlow, meningitis, encephalitis, and genital herpes [3,4]. With the advance of high-throughput sequencing technologies, an increased number of HSV-1 strains have been sequenced.
