Results To avoid confounding from chromosomal abnormalities in prior PMSE patient iPSC lines (Dang et al., 2021), we used CRISPR/Cas9 to generate STRADA loss-of-function KO lines with compound heterozygous truncating variants and isogenic controls (Figures S1A and S1B). Lines expressed pluripotency markers (Figure S1C) and showed no karyotypic abnormalities (data not shown). Two STRADA KO (M1 and M2) and two isogenic control (C1 and C2) lines were used for all experiments.
