Contextual security defenses prevent AI agents from taking rogue actions by synthesizing a task-specific policy and enforcing it on the agent's tool calls. In multi-step tasks, however, which actions are valid often depends on what the agent has already done and learned. We present Sapien, a policy engine for enforcing stateful contextual policies.
Oxygen-Gradient-Responsive Photosensitizing Gadolinium Phthalocyaninate for Self-Adaptive Type II/I Photodynamic Therapy Toward Heterogeneous Tumor Suppression
1 Introduction Solid tumors establish an intricate internal landscape characterized by staggered biochemical profiles, a multifaceted heterogeneity that serves as a primary catalyst for metastasis and disease recurrence [1-5]. Particularly, the non-uniform gradients of oxygen partial pressure throughout this architecture act as a decisive barrier, fundamentally constraining the reach and lethality of oxygen-reliant interventions like clinically approved photodynamic therapy (PDT) [6-10].

1 Introduction Optical frequency combs with gigahertz repetition rates are essential tools in modern photonics, underpinning applications in optical clocks, dual-comb spectroscopy, coherent communications, and frequency referencing [1-4]. A ∼ $\sim$ 1 GHz pulse repetition rate (PRR) comb provides a mode spacing that is resolvable on standard radio frequency (RF) electronics, reducing mode ambiguity and simplifying phase locking for optical frequency metrology [5, 6].
Metabolic–epigenetic crosstalk in latent autoimmune diabetes in adults: potential roles of lactate-induced histone lactylation in immune regulation and pancreatic β-cell fate
Abstract Latent autoimmune diabetes in adults (LADA) is clinically between type 2 and type 1 diabetes with progressive autoimmune loss of β-cells and variable insulin dependence. Lactate was once thought to be a glucan byproduct and is now recognized as a signaling enzyme regulating gene expression via lysine lactylation. One of these marks is histone H3 lysine 18 lactylation (H3K18la), which is enriched at active enhancers and links immune genes.

Abstract Mercury-associated minimal change disease (M-MCD) is a rare form of secondary minimal change disease (MCD) caused by chronic mercury exposure. Given its rarity and nonspecific clinical presentation, M-MCD is frequently underrecognized in clinical practice. We report the case of a 32-year-old woman who presented with nephrotic syndrome following 4 months of applying a skin-whitening cream.
Control of associative learning by the mediodorsal thalamus-orbitofrontal cortex circuit
Abstract Mediodorsal thalamus (MD) projections to the prefrontal cortex are crucial for adaptive cognitive functions. Using a Go/No-Go olfactory discrimination task in male mice, we investigate how MD input to the orbitofrontal cortex (OFC) regulates associative learning and modulates cue-value and outcome representations in OFC neurons. We show that inactivating MD projections to the OFC impairs learning but does not affect performance in the well-trained phase.

POLY-Senolytic nanoplatform for tumor-specific eradication of senescent tumor cells and mitigation of radiotherapy-induced immune resistance of cancer
Abstract Radiotherapy (RT) efficacy is limited by RT-induced immune resistance. Here we show that RT upregulates programmed death ligand 1 (PD-L1) on senescent tumor cells (STCs) via bromodomain-containing protein 4 (BRD4) signaling, thereby promoting immune evasion. To counter this, we develop POLY-Senolytic, a polymeric senolytic nanoparticle formed by conjugating an acid-responsive polymer to a peptide-based BRD4 PROteolysis-TArgeting Chimera via a reduction-cleavable disulfide bond.

Lactobacillus paracasei L9 Ameliorates Pulmonary Fibrosis in Aged Mice via Gut-Lung Axis-Mediated Regulation of Immune Cell Migration
1 Introduction In recent years, numerous studies have shown the lung may develop fibrosis with age (Moss et al. 2022). And because of the aggravation of global aging, the medical burden of pulmonary fibrosis (PF) increases rapidly (Hutchinson et al. 2015). Most patients with PF have a slow progressive decline in lung function that eventually leads to respiratory failure, while 10%–15% of patients experience an unusually rapid decline over several months (Ley et al. 2011).
Tuning Reaction Pathways via Symmetric Fluorination Enables High-Temperature and High-Voltage Electrolytes
1 Introduction To increase the energy density of lithium-ion batteries (LIBs), research has increasingly focused on high-voltage and Ni-rich layered oxide cathode materials (LiNixCoyMnzO2, x ≥ 0.8) owing to their high specific capacity and relatively low cost [1-3]. However, under high voltage, the highly oxidative Ni4+ species catalytically destabilizes the electrolyte at the cathode particle surface, leading to the formation of an unstable cathode-electrolyte interphase (CEI) [4-6].
