Results Overcoming key bottlenecks in engineered myeloid cell therapy requires a scalable source of myeloid progenitors. We developed a chemically defined culture formulation, termed SCF/2i, composed of stem cell factor (SCF), GDC-0879 (BRAF inhibitor),18 and SKL2001 (a reported Wnt/β-catenin modulator),19 that enabled log-phase expansion of primary mouse bone marrow cells for more than 50 days (Figure 1A; see STAR Methods).
