A suite of enhancer AAVs and transgenic mouse lines for genetic access to cortical cell types
Keywords epigenetics transcriptomics enhancer cell type neocortex AAV transgenic recombinase mouse human Introduction To understand how the brain works, we need to establish the structure-function relationships between the units that comprise it and the outputs of its activity.1 One approach is to gain experimental access to each building block (e.g., a cell type) and manipulate it to observe effects on an animal’s physiology and behavior.2,3,4 Historically, the definition of building blocks...
The inherent mutational tolerance and antigenic evolvability of influenza hemagglutinin
Abstract Influenza is notable for its evolutionary capacity to escape immunity targeting the viral hemagglutinin. We used deep mutational scanning to examine the extent to which a high inherent mutational tolerance contributes to this antigenic evolvability. We created mutant viruses that incorporate most of the ≈104 amino-acid mutations to hemagglutinin from A/WSN/1933 (H1N1) influenza.
