The VPS33B-VPS16B protein complex facilitates Golgi ribbon formation and Golgi-endosome trafficking
Abstract Loss of expression of VIPAS39/VPS16B or the Sec1/Munc18 (SM) family protein VPS33B causes early lethal ARC syndrome in humans. These proteins have been shown to form a complex containing 2 opposed VPS33B molecules known as VPS33B-VPS16B. ARC patient-derived fibroblasts have dispersed Golgi elements and absence of ribbons, an abnormality also observed in VPS16B-knockout HEK293 cells. These observations indicate that VPS33B-VPS16B is involved in Golgi formation/stability.
Listeria promotes systemic infection by targeting the host deubiquitinase CYLD
Abstract Listeria monocytogenes (Lm) is an intracellular pathogen that can cause life-threatening systemic infections. Internalin C (InlC) is a secreted factor required for full Lm virulence in systemic models of infection by mechanisms that remain unclear. Here, we show that InlC binds to CYLD, a host deubiquitinase and regulator of innate immunity. Structurally, we reveal the LRR domain of InlC binds the CAP-Gly2 domain of CYLD, independently of InlC’s binding site for other host target proteins.
Intracellular IL-23R is necessary for mitotic spindle formation and viability in AML
|
Abstract Interleukin-23 receptor (IL-23R) is a cell surface cytokine receptor classically expressed on T cells, where it regulates T cell activation. Here, we discovered a novel intracellular localization and function for IL-23R in Acute Myeloid Leukemia (AML). Compared to normal hematopoietic cells, IL-23R was increased in primary AML samples. IL-23R was predominantly localized intracellularly in AML cells. BioID mass spectrometry identified mitotic spindle proteins as top interactors with IL-23R.

Identification of E3 ligase substrates and PROTAC-induced ubiquitylation sites using proximity-based identification of ubiquitin sites (PrIUS)
Abstract The ubiquitin system regulates virtually all cellular processes, yet the vast majority of ubiquitylation sites identified in the human proteome cannot be attributed to specific E3 ligases. This knowledge gap hampers our understanding of ubiquitin signaling and the development of therapeutics leveraging the ubiquitylation system.
Circular RMST cooperates with lineage-driving transcription factors to govern neuroendocrine transdifferentiation

Keywords circular RNA noncoding RNA neuroedocrine prostate cancer small cell lung cancer transcription factors RNA protein interactions lineage plasticity Introduction Circular RNA (circRNA) is a class of noncoding RNA (ncRNA) primarily generated by backsplicing.2,3 The circularization can be promoted through the binding of reverse complementary repetitive sequences in the flanking introns or the dimerization of RNA-binding proteins.2,4 Due to the lack of poly(A) tail, circRNA is...

S-acylation of ATGL is required for lipid droplet homoeostasis in hepatocytes - Nature Metabolism
Abstract Lipid droplets (LDs) are organelles specialized in the storage of neutral lipids, cholesterol esters and triglycerides, thereby protecting cells from the toxicity of excess lipids while allowing for the mobilization of lipids in times of nutrient deprivation. Defects in LD function are associated with many diseases.
DPCD is a regulator of R2TP in ciliogenesis initiation through Akt signaling
Highlights • DPCD is an adaptor for the R2TP chaperone complex • R2TP-DPCD regulates ciliogenesis initiation of primary cilia • R2TP-DPCD interacts with p-AKT2 to control its phosphorylation levels • DPCD specifically interacts with the DII of RUVBL2 within the R2TP-DPCD complex Summary R2TP is a chaperone complex consisting of the AAA+ ATPases RUVBL1 and RUVBL2, as well as RPAP3 and PIH1D1 proteins.
TBL1XR1 Mutations Drive Extranodal Lymphoma by Inducing a Pro-tumorigenic Memory Fate
Highlights • TBL1XR1 mutation skews the humoral immune response toward producing memory B cells • TBL1XR1 mutant memory cells feature aberrant cyclic reentry to new germinal centers • Mutant TBL1XR1 acts by triggering aberrant targeting of SMRT complex to BACH2 • TBL1XR1 mutation gives rise to extranodal ABC-DLBCLs derived from memory B cells Summary The most aggressive B cell lymphomas frequently manifest extranodal distribution and carry somatic mutations in the poorly characterized gene...
