Berzosertib enhances the sensitivity of pediatric diffuse midline glioma H3K27-altered cells to radiotherapy
Abstract Diffuse midline glioma H3K27M-altered (DMG) remains a fatal pediatric brain cancer driven by a global loss of histone H3K27 trimethylation. Radiotherapy comprises the most important treatment modality and significantly improves overall survival. Novel therapeutic strategies for DMG patients without or with radiotherapy are urgently needed. Here, we aimed to gain insights into potential radiation response modulators.

Deep transfer learning approach for automated cell death classification reveals novel ferroptosis-inducing agents in subsets of B-ALL - Cell Death & Disease
Abstract Ferroptosis is a recently described type of regulated necrotic cell death whose induction has anti-cancer therapeutic potential, especially in hematological malignancies. However, efforts to uncover novel ferroptosis-inducing therapeutics have been largely unsuccessful. In the current investigation, we classified brightfield microscopy images of tumor cells undergoing defined modes of cell death using deep transfer learning (DTL).
Linker histone H1-0 is a specific mediator of the repressive ETV6::RUNX1 transcriptional landscape
Abstract ETV6::RUNX1 is the most common oncogenic fusion in pediatric B cell precursor acute lymphoblastic leukemia (BCP-ALL). It induces a clinically silent preleukemic state that requires secondary mutations for progression to leukemia. However, the molecular mechanisms contributing to the characteristic quiescence of ETV6::RUNX1+ preleukemic cells remain elusive.
mTORC1 regulates cell survival under glucose starvation through 4EBP1/2-mediated translational reprogramming of fatty acid metabolism - Nature Communications
Abstract Energetic stress compels cells to evolve adaptive mechanisms to adjust their metabolism. Inhibition of mTOR kinase complex 1 (mTORC1) is essential for cell survival during glucose starvation. How mTORC1 controls cell viability during glucose starvation is not well understood. Here we show that the mTORC1 effectors eukaryotic initiation factor 4E binding proteins 1/2 (4EBP1/2) confer protection to mammalian cells and budding yeast under glucose starvation.
The EIF4EBP1 gene encoding 4EBP1 is transcriptionally upregulated by MYC and linked to shorter survival in medulloblastoma
Abstract Medulloblastoma (MB) is the most common malignant brain tumor in childhood and is stratified into four molecular groups ‒ WNT, SHH, Group 3 and Group 4. Group 3 MB patients exhibit the poorest prognosis, with a 5-year overall survival of <60%, followed by Group 4 MB patients. Apart from MYC amplification in a subset of Group 3 MBs, the molecular pathomechanisms driving aggressiveness of these tumors remain incompletely characterized.
Co-targeting HSP90 alpha and CDK7 overcomes resistance against HSP90 inhibitors in BCR-ABL1+ leukemia cells - Cell Death & Disease
Abstract HSP90 has emerged as an appealing anti-cancer target. However, HSP90 inhibitors (HSP90i) are characterized by limited clinical utility, primarily due to the resistance acquisition via heat shock response (HSR) induction. Understanding the roles of abundantly expressed cytosolic HSP90 isoforms (α and β) in sustaining malignant cells’ growth and the mechanisms of resistance to HSP90i is crucial for exploiting their clinical potential.

Tacedinaline (CI-994), a class I HDAC inhibitor, targets intrinsic tumor growth and leptomeningeal dissemination in MYC-driven medulloblastoma while making them susceptible to anti-CD47-induced macrophage phagocytosis via NF-kB-TGM2 driven tumor inflamma…
Discussion Amplification of MYC constitutes a widely accepted biomarker indicating poor prognosis and driver of aggressive tumor biology in patients with MB, as demonstrated in several clinical trial cohorts and retrospective analyses.7 23 24 Hence, patients with MYC-driven MB are commonly treated with intensified high-dose chemotherapy and radiotherapy protocols.4 However, 5-year overall survival does not exceed 40% in this patient cohort, and the aggressive treatment strategies often result...
Characterization of an RNA binding protein interactome reveals a context-specific post-transcriptional landscape of MYC-amplified medulloblastoma - Nature Communications
Abstract Pediatric medulloblastoma (MB) is the most common solid malignant brain neoplasm, with Group 3 (G3) MB representing the most aggressive subgroup. MYC amplification is an independent poor prognostic factor in G3 MB, however, therapeutic targeting of the MYC pathway remains limited and alternative therapies for G3 MB are urgently needed.

(PHOTOS) Darth Vader, R2-D2 and Boba Fett toys run amuck in the modern world.
Keeping those Star Wars collectibles in their original packaging increases their value over time, but it isn’t much fun. They’re toys, not collectibles, after all, and meant to be played with. Photographer Daniel Picard totally gets that, and has a ball with Figures & Statues. For the last five years, Picard has taken his action figures out of their packages and thrown them into all kinds of funny situations.
