Protein phosphatase 1 regulates phosphorylation of Gasdermin D and pyroptosis Ying Li, Debing Pu, Jian Huang, Ying Zhang and Hang Yin Abstract We report that phosphorylation of Gasdermin D (GSDMD)-NT, cleaved by inflammatory caspases, could inhibit pyroptosis. Phosphatase 1 (PP1) is responsible for the regulation of GSDMD phosphorylation and the capacity of GSDMD to induce cell death. Our findings reveal a critical regulatory mechanism as it provides insight into pyroptosis processes.

Highlights • Crystal structure of glucose-bound PfHT1 was determined at 2.6-Å resolution • Crystal structure of PfHT1 bound to a selective inhibitor, C3361, was elucidated • C3361 binding induces a pocket that can be employed for inhibitor optimization • Rational design yielded more potent selective inhibitors with low cytotoxicity Summary Plasmodium species, the causative agent of malaria, rely on glucose for energy supply during blood stage.
