Managing Metabolic Syndrome and Prostate Cancer
In the Original Investigation by Shaver et al,1 electronic health records of more than 16 000 men with prostate cancer and without metabolic syndrome in the prior 6 months were analyzed for the development of metabolic syndrome following initiation of androgen deprivation therapy (ADT) and androgen receptor pathway inhibitors (ARPIs). Data were obtained from what was collected as routine standard-of-care evaluations.
Darolutamide Plus Androgen-deprivation Therapy in Metastatic Hormone-sensitive Prostate Cancer by Disease Volume and Risk Subgroups in the Phase 3 ARANOTE Trial.
In ARANOTE (NCT04736199), darolutamide plus androgen-deprivation therapy (ADT) significantly reduced the risk of radiologic progression or death versus placebo plus ADT in patients with metastatic hormone-sensitive prostate cancer, with favorable safety. Here, we report on post hoc outcomes by disease volume/risk. Randomized patients received darolutamide plus ADT or placebo plus ADT (2:1). High-volume (HV) and high-risk (HR) disease were defined by the CHAARTED and LATITUDE criteria, respectively.

Early favorable prostate-specific antigen response prediction in metastatic hormone sensitive prostate cancer - Nature Communications
Abstract There is an unmet need for a tool that could predict early favorable prostate-specific antigen (PSA) response in metastatic hormone sensitive prostate cancer (mHSPC) patients receiving androgen receptor pathway inhibitor (ARPI). Here, we train and validate a multivariable logistic regression model to predict early favorable PSA response (≤0.2 ng/mL by 6 months) in these patients.
A plain language summary of the EORTC 1333/PEACE-3 study of enzalutamide alone vs enzalutamide plus radium-223 in patients with metastatic castrationresistant prostate cancer (mCRPC) and bone metastases
Plain Language Summary What is this summary about? Enzalutamide and radium-223 are two different types of medicines normally used separately to treat prostate cancer that has spread to other parts of the body (metastatic castration-resistant prostate cancer; mCRPC). Both medicines are usually given with androgen deprivation therapy (ADT). Radium-223 is only used in people with mCRPC whose cancer has spread to their bones (called bone metastases).
Optimizing therapy for high-risk biochemically recurrent non-metastatic prostate cancer: Current and emerging strategies
Abstract Prostate cancer is a leading malignancy affecting men globally and in Canada. Biochemical recurrence (BCR), marked by rising prostate-specific antigen (PSA) levels post-curative-intended local treatment, is prevalent in nearly one-third of prostate cancer patients and is associated with increased risk of metastases and mortality. The management of patients with BCR is evolving rapidly, highlighting the need for practical guidance.
