Abstract Natural killer (NK) cell-based immunotherapies are promising for cancer treatment due to their ability to eliminate cancer cells independently of antigen presentation and “off-the-shelf” utility. However, molecular determinants governing tumor susceptibility to NK cytotoxicity remain incompletely understood. Here we employ CRISPR activation (CRISPRa) screening to identify cancer cell surface regulators of NK killing.

Editor’s summary Natural killer (NK) cell–based therapies for cancer have shown promise; however, there remains the need to identify the most effective antitumor NK cell subpopulation. Here, Horowitz et al. identified a population of tissue-resident, cytotoxic CD49a+CD103+ NK cells that were distinguishable from their more immunosuppressive counterparts by coexpression of CD39.
