Enhanced electrochemically-directed oligonucleotide synthesis via porous silica thin-film for DNA data storage
Abstract DNA-based data storage has emerged as a promising alternative for long-term information archiving and its implementation accelerates the development for on-chip large scale oligonucleotide synthesis. Among various technological routes, electrochemically directed synthesis offers exceptional synthesis-site density thanks to its compatibility with semiconductor microfabrication processes.
A scalable deep mixed-effects learning framework for high-dimensional and multi-modal biomarker integration and complex disease outcome prediction
Abstract The integration of high-dimensional and multi-modal biomarker data remains a central challenge in precision medicine, hindered by noise, weak individual signals, and heterogeneous data structures. We propose AdaMixNet, an adaptive mixed-effects deep learning framework that unifies nonlinear fixed-effects modeling with kernel-based random-effects estimation to robustly and accurately predict complex disease outcomes.
Sensitivity analysis of cluster cable current-carrying capacity correction factor
Abstract As urban power grid loads keep rising, clustered multi-circuit laying of 110 kV XLPE cables has become prevalent. Thermal interaction between adjacent cable circuits substantially degrades cable ampacity. Conventional engineering calculation approaches fail to fully account for diverse operating scenarios, leading to notable errors in ampacity correction factors.
An extracellular vesicle-mediated mitochondrial transfer network critical for testosterone synthesis - Nature Cell Biology
|
Abstract Testosterone production by testicular Leydig cells (LCs) in male mammals is energetically demanding and prone to mitochondrial damage. Despite these challenges, LCs exhibit remarkable longevity and minimal turnover, suggesting the existence of specialized mechanisms that maintain LC mitochondrial homeostasis under such constrains. Here we identify a mitochondrial transfer network between LCs and different testicular macrophage (tMac) subpopulations.

Brexanolone infusion produces sustained anti-inflammatory and neurotrophic effects in patients with postpartum depression that predict symptom improvement | Translational Psychiatry
Abstract Postpartum depression (PPD) is linked to neuroimmune dysregulation. Brexanolone, an intravenous formulation of the neurosteroid allopregnanolone and the first FDA-approved treatment for PPD, produces rapid and sustained antidepressant effects. However, its long-term mechanisms of action remain unclear. This study evaluated brexanolone’s prolonged impact on two groups of biomarkers in whole blood: inflammatory mediators and growth/differentiation/neurotrophic factors.

Single-cell RNA sequencing reveals transcriptomic landscape and potential targets for human testicular ageing
The testis is an essential male reproductive organ, acting as the source of sperm and a principal provider of testosterone (Makela et al., 2019). Therefore, the testis is critical for maintaining male fertility and ensuring endocrine homeostasis. However, testicular function experiences a gradual decline as men age.

Precise Correction of Lhcgr Mutation in Stem Leydig Cells by Prime Editing Rescues Hereditary Primary Hypogonadism in Mice
1 Introduction Leydig cells (LCs), which are located in the interstitial compartment of the testes and nestled among the seminiferous tubules, are primarily responsible for the production of testosterone.[1] Thus, LCs are indispensable for the development and maintenance of the masculine phenotype, endocrine homeostasis, and reproductive function.[2, 3] Hereditary primary hypogonadism (HPH) is caused by malfunction at the level of the testes due to genetic causes which impair LCs function.[4]...

Editorial: Aging and male hypogonadism
Male hypogonadism, characterized by testosterone deficiency, is common in males over 40 years old (1, 2). Male hypogonadism can lead to sexual and reproductive dysfunction, as well as age-related diseases such as cardiovascular diseases and metabolic disorders, seriously affecting male quality of life and health. Currently, exogenous testosterone replacement therapy is the main clinical treatment for male hypogonadism, but it can only partially alleviate symptoms and cannot cure the disease.

AAV-mediated gene therapy produces fertile offspring in the Lhcgr-deficient mouse model of Leydig cell failure
Highlights • AAV8 targets progenitor Leydig cells through interstitial injection in the testes • AAV8-Lhcgr partially increases testosterone production and initiates sexual development • AAV8-Lhcgr recovers spermatogenesis and effectively produces fertile offspring • These favorable therapeutic effects could be reproduced in adult male Lhcgr−/− mice Summary Leydig cell failure (LCF) caused by gene mutation results in testosterone deficiency and infertility.
Infant gut microbiome composition is associated with non-social fear behavior in a pilot study
Abstract Experimental manipulation of gut microbes in animal models alters fear behavior and relevant neurocircuitry. In humans, the first year of life is a key period for brain development, the emergence of fearfulness, and the establishment of the gut microbiome. Variation in the infant gut microbiome has previously been linked to cognitive development, but its relationship with fear behavior and neurocircuitry is unknown.
Sorting sub-150-nm liposomes of distinct sizes by DNA-brick-assisted centrifugation
Abstract In cells, myriad membrane-interacting proteins generate and maintain curved membrane domains with radii of curvature around or below 50 nm. To understand how such highly curved membranes modulate specific protein functions, and vice versa, it is imperative to use small liposomes with precisely defined attributes as model membranes.
Selective Nitrogen Reduction to Ammonia on Iron Porphyrin-based Single-Site Metal-Organic Frameworks
Selective Nitrogen Reduction to Ammonia on Iron Porphyrin-based Single-Site Metal–Organic Frameworks Constructing efficient catalysts for the N2 reduction into value added ammonia under ambient conditions is a considerable challenge. Herein, well-defined single-site metal–organic frameworks (MOF, M-TCPP; M = Fe, Co, or Zn) were constructed and evaluated as electrocatalysts for N2 reduction. The prepared Fe-TCPP exhibited prominent performance with a high NH3 yield of 44.77 µg h−1 mg−1cat.

Microstructural and Photoconversion Efficiency Enhancement of Compact Films of Lead-Free Perovskite Derivative Rb3Sb2I9 While lead-based perovskites have held center stage in photovoltaic and optoelectronic research over the past decade, their toxicity has raised significant concerns, spurring the search for lead-free alternatives with similar potential.
Copper regulates the interactions of antimicrobial piscidin peptides from fish mast cells with formyl peptide receptors and heparin
Phagocytic cells in fish secrete antimicrobial peptides (AMPs) such as piscidins, glycosaminoglycans such as heparin, and copper ions as first-line immune defenses. Recently, we established that Cu2+-coordination by piscidins 1 (P1) and 3 (P3) enhances their antibacterial activity against membranes and DNA.
