The role of R-loop aberrations in lower-grade gliomas: prognostic, immune, and metabolic implications from multi-omics and machine learning analysis
Abstract Background: The prognostic and therapeutic implications of R-loop regulators in lower-grade glioma (LGG) remain largely unexplored. Methods: We constructed a R-loop prognostic index (RLPI) by benchmarking 9 machine-learning algorithms through nested cross-validation. Associations between RLPI and clinical outcomes, immune microenvironment, and metabolic features were assessed across –11 bulk transcriptomic, 2 single-cell RNA-sequencing, and 1 spatial transcriptomic LGG cohorts.

1. Introduction Continuous functions are an important object of study in classical calculus, and one class of them is known as fractal functions. Fractal functions are an important part of fractal geometry, and their graphs are characterized by fractals. A class of classical fractal functions is known as Weierstrass functions. The Weierstrass function was proposed in 1872. Prior to this, mathematicians believed that continuous functions were continuous everywhere except for a few special instances.

Here, we discover a 9-bp nucleotide sequence that enables efficient polycistronic gene expression in yeasts and filamentous fungi. Coupling polycistronic expression to multiplexed, markerless, CRISPR/Cas9-based genome editing, we develop a strategy termed HACKing (Highly efficient and Accessible system by CracKing genes into the genome) for the assembly of multigene pathways.
