IN YOUR GLASS: One Formula, Three Classics
by ANDERS ERICKSON photos by AZUSA INABA Classic cocktails aren’t complicated. They’re just good at hiding how simple they really are. Many favorites follow clear formulas, and once you recognize a pattern, the recipes feel less like rules and more like variations. The Sidecar, margarita and Pegu Club may look like entirely different cocktails, but they’re built on the same idea: a base spirit, fresh citrus and orange liqueur. Learn one, and you’ve essentially learned all three.
Androgen activity in the male embryonic hindbrain drives lethal PFA ependymoma
|
Abstract Posterior fossa type A (PFA) ependymoma is an unusual infantile brain tumour with few known somatic mutations, thought to be driven by epigenetic mechanisms1. PFA ependymoma has a markedly higher incidence and worse prognosis in male children than in female children2. The mechanisms that underlie these sex differences are at present unknown. Here we show that the cellular hierarchy of PFA ependymoma is less differentiated in male individuals than it is in female individuals.

Brain metastasis burden and management in patients with small cell lung cancer in Canada: a retrospective, population-based cohort study
Keywords Small cell lung cancer Brain metastasis Stereotactic radiosurgery Whole brain radiation therapy Evidence before this study We searched PubMed for peer-reviewed studies from database inception until June 2, 2022 using search terms related to “small cell lung cancer”, “brain metastasis”, and “management”. A total of 826 studies were retrieved of which 39 focused on brain metastasis (BM) in small cell lung cancer (SCLC).
Retrospective: A Producer’s Guide to Team-Buy-In
A game producer facilitates an efficient work environment by assigning tasks, clearing blockers, and planning schedules. However, all this work can fall by the wayside if the team isn’t invested in the vision of the game. But, in an industry full of creative and passionate minds, how do you get everyone’s interests aligned?
Inferred Developmental Origins of Brain Tumors from Single Cell RNA-Sequencing Data
Abstract The reactivation of neurodevelopmental programs in cancer highlights parallel biological processes that occur in both normal development and brain tumors. Achieving a deeper understanding of how dysregulated developmental factors play a role in the progression of brain tumors is therefore crucial for identifying potential targets for therapeutic interventions.

Failure of human rhombic lip differentiation underlies medulloblastoma formation - Nature
|
Abstract Medulloblastoma (MB) comprises a group of heterogeneous paediatric embryonal neoplasms of the hindbrain with strong links to early development of the hindbrain1,2,3,4. Mutations that activate Sonic hedgehog signalling lead to Sonic hedgehog MB in the upper rhombic lip (RL) granule cell lineage5,6,7,8. By contrast, mutations that activate WNT signalling lead to WNT MB in the lower RL9,10.

Assessing the Association of Targeted Therapy and Intracranial Metastatic Disease
Key Points Question What is the association of targeted therapy with survival for patients with intracranial metastatic disease (IMD)? Findings In this cohort study of 26 676 patients with IMD, prolonged survival was observed in patients with IMD and ERBB2 (formerly HER2)–positive breast cancer, EGFR-positive lung and bronchus cancer, or BRAF-positive melanoma who received targeted therapy compared with those who did not.
Dual- versus single-agent HER2 inhibition and incidence of intracranial metastatic disease: a systematic review and meta-analysis
Abstract Observational studies have suggested that HER2 inhibition with trastuzumab may be associated with an increased incidence of intracranial metastatic disease (IMD) due to its ability to prolong survival. We hypothesized that prolonged survival associated with dual-agent HER2 inhibition may be associated with an even higher incidence of IMD.
