Genetic variation in antidiabetic drug targets: associations with Parkinson’s disease risk and age at onset
Abstract To investigate whether antidiabetic drugs have a biological basis to be repurposed in PD prevention, we applied a drug target Mendelian randomization framework to assess associations between genetic variation in antidiabetic drug targets and PD risk or age at onset (AAO).
A multimodal biomarker strategy to enhance diagnostic precision in neurodegenerative parkinsonism
|
Abstract Differential diagnosis of neurodegenerative parkinsonian syndromes is complicated by overlapping clinical features and frequent co-pathology that challenges the interpretation of single-protein biomarkers. We evaluated a multimodal, minimally invasive biomarker strategy integrating dermal α-synuclein and 4-repeat tau seed amplification assays (SAAs) with serum neurofilament light chain.

Genome-wide association study of copy number variations in Parkinson’s disease
|
Abstract We investigated the role of copy number variations (CNVs) in Parkinson’s disease (PD) using genotyping data from 10,815 patients (2731 early-onset PD, EOPD) and 8901 controls from the COURAGE-PD consortium. CNVs were analyzed using a sliding window genome-wide association and burden approach.

The “Hallett Sign” of Functional Jerky Movement Disorder
Mark Hallett (1943–2025), a giant in the fields of movement disorders and functional neurological disorder (FND), will be sadly missed by a global research community.

LRRK2 as a Potential Disease‐Modifying Target in Sporadic Parkinson's Disease
Parkinson's disease (PD) is a complex, multifactorial, and progressive disease characterized by motor and non-motor symptoms.1, 2 Approximately 13% to 15% of PD cases have a genetic link, whereas up to 85% are sporadic (sPD), with no demonstrated single genetic cause.3, 4 In 2002, the PARK8 locus was identified in a large Japanese family with autosomal dominant parkinsonism.5 Two years later, leucine-rich repeat kinase 2 (LRRK2) was identified as the gene implicated in this locus.6, 7 LRRK2...
Disease‐Modifying Trials in Treated Parkinson's Disease: “Stable Treated” Does Not Equate with Biological Stability
The greatest unmet need in Parkinson's disease (PD) is the development of effective disease-modifying therapy (DMT).

Research Priorities on the Role of α‐Synuclein in Parkinson's Disease Pathogenesis
α-Synuclein (αSyn) pathology is a major feature of Parkinson's disease (PD), and the occurrence of pathogenic variants in the SNCA gene1-12 or multiplication of its locus13, 14 supports a possible causative role of αSyn in PD. Yet the nature of this role has long been a matter of debate due to unclear αSyn-mediated mechanisms of cell dysfunction, degeneration, and death.
Concerns with the new biological research criteria for synucleinopathy - Authors' reply
We would like to thank the authors of the letters for their interest in our SynNeurGe biological classification and address the concerns that they raise. Fred B Ketchum and Nathaniel A Chin highlight important ethical issues that need to be considered in any biological classification that will include asymptomatic individuals positive for the biomarkers of interest, many of whom might never develop clinical manifestations. We agree entirely with their concerns.

Concomitant Medications for Progressive Supranuclear Palsy
There are currently no approved therapies for progressive supranuclear palsy (PSP).1-3 In this post hoc secondary analysis of a phase 2/3 randomized clinical trial of davunetide for PSP (NCT01110720),4 we examined PSP progression outcomes for all included medication classes. Methods Included participants had completed the PSP Rating Scale (PSPRS) and provided concomitant medication data. Both davunetide and placebo arms from the trial4 were included, as there was no treatment effect.

Teaching Video NeuroImage: “Weighing” in on an Unusual Tremor
A 73-year-old right-handed man with hypertension, hypercholesterolemia, renal insufficiency, and cyclothymic disorder presented with 1 year of insidious onset tremor in his right hand when carrying his briefcase. Psychotropic drugs included bupropion and lamotrigine. He was neurologically intact except for a very low amplitude bilateral kinetic tremor (L > R) and a much larger amplitude right hand task-specific tremor (video 1) characterized by EMG (figure). Video 1 Tremor phenomenology.

Initiating pharmacotherapy in early Parkinson's disease
We would like to address the Comment that accompanied our Review on the initiation of pharmacotherapy in patients with Parkinson's disease. We agree with Anthony Schapira on the importance of taking into consideration the views of the patient in deciding on treatment. However, it is the responsibility of the treating physician to use the relevant evidence in guiding early treatment decisions.

The evidence for multidisciplinary care in Parkinson’s disease
Introduction Parkinson’s Disease (PD) is a chronic and slowly progressive neurodegenerative disease. Team-based care is required to address and manage the diverse array of motor and non-motor symptoms in PD and related conditions. As the evidence base for the efficacy of non-pharmacological treatment of PD is expanding, many different centers are implementing interdisciplinary models of care with allied health professionals trained in PD.
