Discovery of a peripheral Myh11-expressing nucleus pulposus cell population demontrating therapeutic potential for disc degeneration
Abstract Continual turnover of nucleus pulposus (NP) cellularity is critical for maintaining intervertebral disc (IVD) homeostasis; however, the presence and identity of potential progenitor cells within the NP remain unclear. This study identifies NP cells expressing myosin heavy chain 11 (Myh11+ NPCs) through single-cell transcriptomic analysis of human adolescent NP cells and spatial transcriptomic mapping of the murine juvenile IVD.
Reducing city light pollution could improve urban tree and plant growth
New York City and Washington, DC, became darker after adopting measures to reduce artificial light at night, especially blue light. Satellite images showed that seasonal plant growth in those cities shifted closer to its long-term timing than in Newark and Philadelphia. The results are promising but exploratory, since the analysis covered only two city pairs and one year after the policies took effect. City lights do more than brighten streets.
Sensory nerve-derived signaling coordinates oropharyngeal structural organization that supports suckling and vocalization in neonatal mice
Abstract Proper oropharyngeal function is essential for suckling, feeding, and speech, and relies on the coordinated development of the palate, oropharyngeal musculature, and the cranial nerves that control them. Disruption of this integration leads to severe neonatal complications. However, how neuromuscular architecture is developmentally coordinated to support oropharyngeal function remains unclear.
Case Report: zanidatamab-based regimen as a potential later-line therapeutic option in HER2-amplified metastatic colorectal cancer
Abstract Background: Overexpression human epidermal growth factor receptor 2 (HER-2) has been identified as a key oncogenic driver in metastatic colorectal cancer (mCRC), detected in between 2% and 5% of the overall mCRC population, with a higher prevalence identified in RAS/RAF/PI3KCA wild-type patients. Optimal targeted interventions are seldom administered to patients with HER2-positive mCRC due to the absence of definitive anti-HER2 therapeutic regimens in the refractory setting.
KDM6B safeguards mineralized tissue homeostasis from mechanical stress through epigenetic control of PIEZO1-mediated mechanotransduction in the mouse incisor
Abstract Mechanical forces shape the growth and regeneration of mineralized tissues such as bones and teeth, yet how these tissues adapt to sustained mechanical stress remains poorly understood. Here, using mouse incisor models with varying degrees of loading, we identified that the histone demethylase KDM6B is a critical epigenetic regulator that preserves mineralized tissue homeostasis by protecting progenitor transit-amplifying cells from mechanical stress-induced apoptosis.
Ozone mitigates extended growing season and enhanced vegetation greenness driven by environmental change
Abstract Rising temperature and elevated CO2 concentrations lead to extended growing seasons and enhanced vegetation greenness in terrestrial ecosystems, especially across the Northern Hemisphere. However, whether and to what extent surface ozone, an anthropogenic environmental factor, affects vegetation phenology and greenness remains unexplored at a large scale.
Canonical BAF chromatin remodeling complex specifies stem cell fate via cell-type-specific co-factor recruitment - Nature Communications
Abstract Individual subunits within the canonical BAF (cBAF) chromatin remodeling complex are known to regulate stem cell behavior, with some functional redundancy across subunits. Yet, how the cBAF complex directs adult stem cell fate specification and maintains stem cell niches remains unclear. Using the adult mouse incisor, we show that cBAF specifies mesenchymal stem cell (MSC) fate by recruiting distinct transcriptional co-factors to shape cell-type-specific chromatin regulation.
Data-driven subtyping of early Parkinson’s disease via mutual cross-attention fusion of EEG and dual-task gait features - npj Parkinson's Disease
Abstract Parkinson’s disease (PD) exhibits marked clinical heterogeneity, which poses challenges for diagnosis, prognosis, and therapeutic precision, especially for early-stage PD patients. Existing subtyping approaches often rely on subjective clinical scales and single-modality data, which limits their sensitivity in capturing subtle but clinically relevant differences across patients.
Amplified Arctic–boreal fire regimes from permafrost thaw feedbacks - Nature Geoscience
|
Abstract Widespread permafrost thaw—indicated by the thickening of the soil active layer, which undergoes seasonal freeze–thaw cycles above the permafrost—has important implications for the thermal, hydrological and ecological processes of Arctic–boreal ecosystems. Yet, how thaw-driven biophysical and ecological feedbacks alter Arctic–boreal fire regimes remains poorly understood.

Hot droughts in the Amazon provide a window to a future hypertropical climate - Nature
Abstract Tropical forests represent the warmest and wettest of Earth’s biomes, but with continued anthropogenic warming, they will be pushed to climate states with no current analogue1,2. Droughts in the tropics are already becoming more intense as they occur at successively higher temperatures3,4,5. Here we synthesize multiple datasets to assess the effects of hot droughts on a central Amazon forest.
Artificial light at night outweighs temperature in lengthening urban growing seasons - Nature Cities
Abstract Plant growing seasons are largely regulated by light and temperature. Cities are increasingly hot (higher air temperature), from the urban heat island effect, and bright (artificial light at night, ALAN). However, the relative effect of heat and light on the timing of plant growth events, called phenology, is unclear, limiting our understanding under climate change and urbanization.

10 Manga That Are Too Controversial To Receive An Anime Adaption
With a medium as extensive as anime, it's not surprising that it's had a fair amount of controversy over the years. Whether it be from a series being too gory, lewd, or deplorable, a good deal of anime has been banned in certain countries due to its graphic nature. When compared to manga content, however, anime gore is tame. There are many manga with the most explicit content imaginable, such as Dead Tube, Real Account, and Pumpkin Night.

As a vital part of the global supply chain system, China actively develops the manufacturing industry and volunteers to shoulder the international industrial transfer, in an attempt to integrate more deeply into the global supply chain system.
Green with phenology
Temperature, Lights, Action Urban Treescapes Warming Speeds Greening Bright Sky at Night Our Changeable Environment References and Notes Biographies Trees blossom earlier in urban environments. Do you remember when you saw the first buds burst this spring? Trees do not have watches or calendars, but they seem to know when spring arrives better than we do. The timing of seasonal biological events—such as when trees leaf out, flowers open, and leaves turn yellow—is called phenology (1).

Bone mesenchymal stem cells are recruited via CXCL8‐CXCR2 and promote EMT through TGF‐β signal pathways in oral squamous carcinoma
Abstract Objectives Bone mesenchymal stem cells (BMSCs) play critical roles in tumour microenvironment. However, molecular mechanisms of how BMSCs to be recruited and effect subsequent tumour progression are poorly understood in oral squamous cell carcinoma (OSCC). Materials and Methods The distribution of CXCL8 was detected by immunohistochemical staining in OSCC tissues. The chemotaxis of conditioned media from different epithelial cells to BMSCs was examined by trans‐well assay.

Molecular Cell Biology Cheng Zhang, Like Qu, Shenyi Lian, Lin Meng, Li Min, Jiafei Liu, Qian Song, Lin Shen and Chengchao Shou DOI: 10.1158/0008-5472.CAN-18-0520 Published March 2019 Abstract The oncogenic phosphatase PRL-3 is highly expressed in metastatic colorectal cancer but not in nonmetastatic colorectal cancer or noncolorectal cancer metastatic cancers.
Light Pollution Reshapes Growing Seasons | Think Global Health
Across cities in the Northern Hemisphere, it’s not unusual to see trees blooming earlier in the spring or keeping their leaves later in the autumn than trees in the countryside. Those shifts exemplify how cities are quietly changing nature’s calendar. For years, warmer urban temperatures (urban heat island effect) were thought to be the main reason for these seasonal shifts.
PRL-3 promotes ubiquitination and degradation of AURKA and colorectal cancer progression via dephosphorylation of FZR1
Abstract The oncogenic phosphatase PRL-3 is highly expressed in metastatic colorectal cancer (CRC) but not in non-metastatic CRC or non-CRC metastatic cancers. Although the pro-invasive capacity of PRL-3 has been validated in multiple types of cancer, its impact on CRC progression and the underlying mechanisms remain poorly understood.

For the cloning of Na+/H+ antiporter genes, Sau3AI-digested genomic DNA from strain NEAU-ST10-25T was electroporated into E. coli KNabc and its transformants were screened on the LBK medium plates containing 0.2?M NaCl. Of several resultant clones, one recombinant plasmid designated pUC-ZD-6 [pUC18 carrying a 4.2-kb DNA fragment (Fig. 1)] enabled E. coli KNabc to grow on the LBK medium plate containing 0.2?M NaCl, which is the upper limit of NaCl tolerance concentrations for E.
Antibody Array Revealed PRL-3 Affects Protein Phosphorylation and Cytokine Secretion
Phosphatase of regenerating liver 3 (PRL-3) promotes cancer metastasis and progression via increasing cell motility and invasiveness, however the mechanism is still not fully understood. Previous reports showed that PRL-3 increases the phosphorylation of many important proteins and suspected that PRL-3-enhanced protein phosphorylation may be due to its regulation on cytokines.
