SARS-CoV-2-specific monoclonal antibody AER002 in Long COVID: an exploratory randomized phase 2a mechanistic trial
|
Abstract Persistence of SARS-CoV-2 has been proposed as a biological driver of Long COVID, a disabling chronic illness with no proven treatments. We conducted an exploratory, placebo-controlled, double-blind, 2:1 randomized mechanistic trial (NCT05877508) of the SARS-CoV-2-specific monoclonal antibody AER002 in 36 participants who met the World Health Organization case definition of Long COVID. After baseline characterization, participants received a single infusion and were followed for 360 days.

Potent type-specific de novo antibodies complement broadly reactive imprinted antibodies in immune responses to SARS-CoV-2 variants
|
Abstract For rapidly mutating viruses such as influenza viruses and severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), immune memory recalled by antigenically drifted variants primarily comprises antibodies that cross-react to the priming strain rather than de novo elicited responses, a phenomenon termed original antigenic sin or immune imprinting. The composition and functionality of de novo responses elicited by variant exposures remain unclear.

I was right then, and I'm right now. Trump must be removed | Opinion Original
April 28, 2026, 1:46 p.m. ET In February of 2017, I said here that Donald Trump must be impeached. Some people said I was premature, in that he had only begun violating the Constitution. Others said I was just nuts. I said it again when he was in fact impeached but not convicted − twice − because Republican senators would rather condone treason than imperil their party or their persons. By then, there could be no doubt that Trump was a major threat to our country and our democracy.
Publisher Correction: Tissue-resident exhausted and memory CD8+ T cells have distinct ontogeny, function and roles in disease - Nature Immunology
Correction to: Nature Immunology https://doi.org/10.1038/s41590-025-02352-y, published online 29 December 2025. In the version of this article initially published, there was a typographical error in the title, where “roles in disease” appeared originally as “role in disease.” Also, the name of Alexander C. Huang was shown incorrectly (Huang C. Alexander). The name and title are now amended in the HTML and PDF versions of the article. About this article Park, S.L., Painter, M.M., Manne, S. et al.
Tissue-resident exhausted and memory CD8+ T cells have distinct ontogeny, function and role in disease - Nature Immunology
Abstract The presence of CD8+ T cells coexpressing residency and exhaustion molecules in chronic diseases often correlate with clinical outcomes; however, the relationship between these cells and conventional tissue-resident memory (TRM) cells or exhausted CD8+ T (TEX) cells is unclear. Here we show that chronic antigen stimulation drives development of tissue-resident TEX (TR-TEX) cells that are distinct from TRM cells generated after antigen clearance.

Cleanet: Robust Doublet Detection in Cytometry Data Based on Protein Expression Patterns
1 Introduction Flow and mass cytometry studies are becoming increasingly complex as they involve higher-dimensional data and larger sample sizes. In order to scale to thousands of samples, studies would benefit from automating as much of the analysis tasks as possible.

Automated cytometric gating with human-level performance using bivariate segmentation - Nature Communications
Abstract Recent advances in cytometry have enabled high-throughput data collection with multiple single-cell protein expression measurements. The significant biological and technical variance in cytometry has posed a formidable challenge during the gating process, especially for the initial pre-gates which deal with unpredictable events, such as debris and technical artifacts.
Cleanet: robust doublet detection in cytometry data based on protein expression patterns
Abstract Flow and mass cytometry experiments are essential for profiling immune cells at single cell resolution. Better understanding of human immunology increasingly involves analyzing studies at the scale of hundreds or thousands of samples, with data analysis a significant bottleneck. This trend increases the demand for automated analysis methods.
Opinion: Hamilton County did ourselves proud with election choices
Mark P. Painter It is not our fault. Hamilton County, where l was born at Christ Hospital and have lived all my life (so far), did everything right in this election. If you did not appreciate the outcome of the national elections, voters in other counties and states are to blame. While Ohio has fallen to the barbarians, our county abides in harmony with truth, justice, and the American way. More:What is the Trumpiest area of Hamilton County?

County prosecutor's win-at-all-cost mentality elevates convictions over justice | Opinion
Mark P. Painter In the recent Enquirer article, "Critics dismiss Hamilton County integrity unit as 'political stunt,'" two things caught my attention. First, that the "integrity unit" supposedly established by our prosecutors' office is totally fake. It’s not anything similar to "real" integrity units that other prosecutors have established; it is a mirror image. If the best practice is to have someone from outside be the unit’s head, then the one here is one of the longest-serving employees.
Immunological imprinting shapes the specificity of human antibody responses against SARS-CoV-2 variants
The spike glycoprotein of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) continues to accumulate substitutions, leading to breakthrough infections of vaccinated individuals. It remains unclear if exposures to antigenically distant SARS-CoV-2 variants can overcome memory B cell biases established by initial SARS-CoV-2 encounters.

Prior vaccination promotes early activation of memory T cells and enhances immune responses during SARS-CoV-2 breakthrough infection - Nature Immunology
Abstract Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection of vaccinated individuals is increasingly common but rarely results in severe disease, likely due to the enhanced potency and accelerated kinetics of memory immune responses. However, there have been few opportunities to rigorously study early recall responses during human viral infection.

Make the railroad deal; spend some money | Opinion
Cincinnatians will vote on selling our railroad in November. I will vote yes. The deal is too good to pass up. By far. I trust the Cincinnati Southern Railway Board of Trustees. They have studied, negotiated and debated the deal. Two lawyers, an experienced CPA, a former P&G executive, two former mayors, a former councilwoman, two Republicans and three Democrats. All in a total of five people. And I trust the present mayor. Our mayor and all present council members support the deal. I trust the deal.
SARS-CoV-2 reservoir in post-acute sequelae of COVID-19 (PASC) - Nature Immunology
Abstract Millions of people are suffering from Long COVID or post-acute sequelae of COVID-19 (PASC). Several biological factors have emerged as potential drivers of PASC pathology. Some individuals with PASC may not fully clear the coronavirus SARS-CoV-2 after acute infection. Instead, replicating virus and/or viral RNA—potentially capable of being translated to produce viral proteins—persist in tissue as a ‘reservoir’.
Should Investors Worry About China?
The rise of stock market indices has caught many investors by surprise this year. In fact, most research firms were anticipating a recession and the positioning in managed funds coming into 2023 was on the defensive side. While there are many factors that drive asset prices in the short term, over the long term asset prices are related and can tell investors a lot about the current backdrop and also expectations of the future.
An educated population is a public good; so is forgiving student debt | Opinion Original
Mark P. Painter The U.S. Supreme Court just issued a major decision on federal student loans. Specifically, whether the president had the power to forgive the first $20,000 owed. It's more complicated, but that's the gist. I’m not going to comment much on the legal dispute −I had predicted that the court would invalidate the plan on the separation of powers issue − whether Congress or the president can make the decision. That is what happened. In my view, the court was correct legally.

A tale of two countries and two parties
by Mark P. Painter In 2016 one country elected a narcistic, opportunist, bigoted, congenital liar, claiming populism but born to wealth and privilege, a cheating and mostly failed businessman, to head the party and the government. He proceeded to appoint grifters and incompetents to the cabinet, valued loyalty to himself above loyalty to the country. He lied to the country, his colleagues, courts, and then lied about the lies.

Opinion: COVID-19 should never have become political
Mark P. Painter For 18 months, we have known about COVID-19. Not much at first. We were equal in our ignorance. But the USA leads the world in medical science. Our doctors, scientists and medical techs went to work and learned how to help prevent the spread – by masks and distancing. Not totally effective, but much better than nothing. We could only do masks and distancing until our medical people developed a vaccine. Applying those measures helped. But not fully doing so let the virus gain on us.

Rapid induction of antigen-specific CD4+ T cells is associated with coordinated humoral and cellular immunity to SARS-CoV-2 mRNA vaccination
Highlights • mRNA vaccines generate antigen-specific T cells in a coordinated immune response • Vaccine-induced T cells resemble the durable memory cells primed by infection • Th1 and cTfh cell responses to the first dose correlate with second-dose responses • SARS-CoV-2-recovered individuals benefit from the first but not the second dose Summary SARS-CoV-2 mRNA vaccines have shown remarkable clinical efficacy, but questions remain about the nature and kinetics of T cell priming.
Opinion: Can lawsuits save democracy?
Mark P. Painter Rupert Murdoch has a lot to answer for. The Iraq war, which his news empire fostered. The near death of truth in journalism. Roger Ailes, who, with Murdoch’s connivance, successfully elevated entertainment, and lies, over truth in TV news. All leading to the Orwellian conditions that led to the rise of a demagogue like Donald Trump. Not only did Murdoch’s empire lead to Trumpism – constant lies told as truth – but enabled it once it grew from the Murdoch seed.

Distinct antibody and memory B cell responses in SARS-CoV-2 naïve and recovered individuals following mRNA vaccination
Abstract Novel mRNA vaccines for SARS-CoV-2 have been authorized for emergency use. Despite their efficacy in clinical trials, data on mRNA vaccine-induced immune responses are mostly limited to serological analyses. Here, we interrogated antibody and antigen-specific memory B cells over time in 33 SARS-CoV-2 naïve and 11 SARS-CoV-2 recovered subjects.

Opinion: Trump might be the last Republican president
Mark P. Painter Abraham Lincoln was right: You can fool some of the people all the time. He was right, in that more than 60% of the party he headed now believe in a complete myth – that this presidential election was somehow "stolen." Never mind that about 60 courts, including the Supreme Court – which Donald Trump himself stacked – have all but laughed at Rudy Giuliani’s fevered dreams. Even his own – now resigned – attorney general said there was no fraud.
Opinion: Trump has no conscience, no moral compass
Mark P. Painter Donald Trump is not a racist. Neither is he a Fascist. Sure, he shows many of the same traits – and people are free to use those as evidence of racism or Fascism. But deep down, he is neither, because deep down he is shallow. Trump is not a Republican. Nor is he a Democrat. Yes, he claims to be a Republican, but he has repudiated the Republican principles I grew up with – balanced budgets, free trade, fiscal responsibility, civil rights, fair dealing, honesty.

Opinion: Not removed, still incompetent and illiterate
Mark P. Painter, Opinion contributor Published 10:44 a.m. ET March 2, 2020 In the early days of American democracy, being a wealthy, white, male landowner was a prerequisite to having a voice in how the country was run. So it should come as no surprise that many of America’s leaders have been quite wealthy. Winning a bid for the presidency requires a significant amount of hard work and good luck, but it is also often helpful to have another key resource -- wealth.
Opinion: Uncle Sam is drowning in debt
Mark P. Painter, Opinion contributor Published 5:38 p.m. ET Nov. 22, 2019 National debt (Photo: Ramirez) Now that Trump’s perfidy is proved beyond cavil, we should look at some of non-impeachable damage he has caused to our country. During his campaign, our now-president said that he would pay off the national debt in eight years. It would be easy, like winning trade wars. Trump does know a lot about payoffs and debts, though the two are usually not associated in his mind. But facts matter.
