Abstract Cellular metabolism dynamically adapts to nutrient fluctuations, yet the regulatory mechanisms underlying this plasticity remain incompletely understood. In particular, the adaptive activation mechanism of enoyl-CoA hydratase short-chain 1 (ECHS1), a key mitochondrial enzyme for fatty acid and amino acid catabolism, is poorly characterized.
GID4-Recruiting PROTACs for BRD4 Degradation Overcome Resistance Driven by CRBN and VHL Deficiency Click to copy article link Article link copied!
Abstract Click to copy section linkSection link copied! Despite the identification of over 600 E3 ligases, current proteolysis-targeting chimeras (PROTACs) predominantly rely on CRBN and VHL. Recently, GID4, a substrate receptor of the CTLH E3 ligase complex, emerged as a promising alternative handle. Herein, through structural simplification of a known GID4 ligand, we developed compound a11 as a potent GID4-recruiting BRD4 degrader.

SMAD2 S-palmitoylation promotes its linker region phosphorylation and TH17 cell differentiation in a mouse model of multiple sclerosis
Editor’s summary Despite being activated by the same receptors and sharing considerable homology, SMAD2 promotes proinflammatory TH17 cell differentiation, whereas SMAD3 drives anti-inflammatory Treg cell generation. Zhang et al. uncovered a SMAD-specific posttranslational modification. SMAD2, but not SMAD3, underwent dynamic palmitoylation that promoted its phosphorylation and activation, as well as its interaction with the transcription factor STAT3, leading to TH17 cell generation in vitro.
YAP controls cell migration and invasion through a Rho GTPase switch
Editor’s summary The transcriptional coactivator YAP promotes development and wound healing and is implicated in various diseases. Shah et al. found that YAP promoted cell motility by inducing a switch in the activity of Rho family GTPases. YAP-TEAD4 complexes mediated the induction of TRIO, a guanine nucleotide exchange factor that activated Rac1 and consequently inhibited RhoA, resulting in increased cell migration and invasion.
