Developing a competency framework for operating room nurse specialist training instructors: a Delphi study
Abstract Objective: To develop a competency evaluation framework for OR nurse specialist training instructors. Methods: Guided by the Iceberg Model and the Onion Model of competency, a preliminary indicator framework was developed through a literature review, behavioral event interviews, and expert group discussions. A two-round Delphi survey involving 31 operating room nursing experts was conducted to refine the indicators.
Synthesis and Activity-Based Protein Profiling Identifies Aldo-Keto Reductase 1C3 as Target Protein of Myxoglucamides
1 Introduction The study of natural products has been highly beneficial for the progression of biological research [1-3]. They have not only been a highly prolific source of new drugs until today [4], but also disclosed original cellular targets and mechanisms that were subsequently exploited therapeutically by the natural product itself, or by other molecular formats [5].
Hypertensive disorders of pregnancy contribute to developmental and metabolic multimorbidity in preterm infants
Abstract Preterm birth poses a global health challenge, frequently resulting in complex infant multimorbidity. While the adverse effects of maternal hypertensive disorders of pregnancy (HDP) on offspring are well recognized, their specific impact on distinct multimorbidity patterns in very preterm infants remains underexplored. In this multicenter cohort study, we utilize the Sino-northern Neonatal Network (SNN) database to systematically characterize these profiles.
Modified Huangqi Chifeng decoction alleviates IgA nephropathy inflammation and renal fibrosis via miR-146a/TLR4/NF-κB signaling modulation
Abstract Background: Immunoglobulin A nephropathy (IgAN) is the most prevalent primary glomerulonephritis worldwide and a leading cause of end-stage renal disease, posing a significant threat to human health. Modified Huangqi Chifeng decoction (MHCD) has shown efficacy in ameliorating IgAN; however, its underlying regulatory mechanisms remain incompletely understood.
Atypical signaling, ligand recognition and selective agonist discovery of complement receptor C5aR2
Abstract C5a, the most potent anaphylatoxin in the complement system, exerts its effects through the canonical G protein-coupled receptor C5aR1 and the arrestin-coupled receptor C5aR2. Despite the critical role of C5aR2 in immunomodulation, the molecular mechanisms underlying its biased signaling, ligand recognition, and associated pathophysiology remain poorly understood.
Fringe-width encoded line-shifting method for complex 3D measurement
Complex 3D measurements, including reflective, dark, transparent, humid, and dynamic surfaces, pose challenges for existing fringe projection profilometry (FPP). Phase-shifting methods face difficulties in accurately measuring the surfaces of objects that are reflective, dark, transparent, or humid, due to its reliance on the surface light intensity. Thus, line-shifting methods emerge as a viable solution, leveraging binary patterns to enhance robustness.
Dynamic neutrophil-to-lymphocyte ratio predicts prognosis in patients with soft tissue sarcoma: a retrospective study of 231 cases
Abstract Background: Soft tissue sarcomas (STS) exhibit significant heterogeneity and are classified as rare tumors with a high risk of metastasis. The neutrophil-to-lymphocyte ratio (NLR), a hematological marker indicative of systemic inflammation, has gained broad recognition for its prognostic utility in oncology. This ratio can be used to evaluate the dynamic changes in inflammatory markers during the diagnosis and treatment of tumors.
Python metabolomics uncovers a conserved postprandial metabolite and gut–brain feeding pathway
|
Abstract Most mammals consume small and frequent meals. By contrast, pythons are ambush predators that exhibit extreme feeding and fasting patterns and provide a unique model for uncovering molecular mediators of the postprandial response1,2,3. Using untargeted metabolomics, we show that circulating levels of the metabolite para-tyramine-O-sulphate (pTOS) are increased more than 1,000-fold in pythons after a single meal.

Systemic Pressures and Nurse Decision‐Making in ICU Missed Nursing Care: A Meta‐Synthesis
3.3.2.3 Organisational-Level Constraints At a deeper structural level, nurses identified the absence of institutional support, inadequacies in staffing management mechanisms, and the lack of effective systems for monitoring, feedback and incident reporting. Some nurses remarked that ‘when something goes wrong, it's always the nurses who bear the consequences’, highlighting a tacit organisational tolerance and normalisation of missed care.

DSS1 inhibits autophagy to activate epithelial-mesenchymal transition in a pro-metastatic niche of renal cell carcinoma - Nature Communications
Abstract The mechanisms underlying clear cell renal cell carcinoma (ccRCC) metastasis remain largely unexplored. We demonstrate that Deleted in Split hand/Split foot protein 1 (DSS1), a critical cofactor of BRCA2 in DNA repair, is upregulated in metastatic ccRCC and promotes both tumor growth and distant metastasis. Mechanistically, DSS1 interacts with LC3 and promotes its degradation via TRIM25-mediated Lys63 (K63)-linked polyubiquitination at LC3B-K51.

Radical Footprinting in Mammalian Whole Blood
Abstract Hydroxyl Radical Protein Footprinting (HRPF) is a powerful tool to probe protein higher-order structure, as well as protein-protein and protein-carbohydrate interactions. It is mostly performed in vitro, but recent advances have extended its use to live cells, nematodes, and 3D cultures. However, application in living mammalian tissues has not been accomplished.

Temporal mapping of the anti-tumor effects of nanobody-based MSLN.CAR-T cell therapy in metastatic solid tumors
Abstract Studies on the dynamic changes occurring in the tumor microenvironment (TME) following CAR-T cell therapy have been confounded by host lymphodepletion, multiple dosing and immunodeficient models. Here, a nanobody-based, mouse mesothelin-targeting CAR-T cell (A101) was developed, achieving effective primary tumor suppression, metastasis reduction, and improved survival after a single dose in immunocompetent, syngeneic mouse models without lymphodepletion.
Genetic variation in IL-4 activated tissue resident macrophages determines strain-specific synergistic responses to LPS epigenetically - Nature Communications
Abstract How macrophages in the tissue environment integrate multiple stimuli depends on the genetic background of the host, but this is still poorly understood. We investigate IL-4 activation of male C57BL/6 and BALB/c strain specific in vivo tissue-resident macrophages (TRMs) from the peritoneal cavity.

The mechanisms and factors that induce trained immunity in arthropods and mollusks
1 Introduction The immune system evolved under coevolutionary selection and is the backbone of animal resistance to pathogen attack (1). The immunity of an organism is divided into adaptive immunity and innate immunity. Adaptive immunity evolved independently in vertebrates (2) and is the only one that has memory. However, a growing number of studies have shown that innate immunity can enhance immune responses to secondary infection, which imply that innate immunity has memory (3).
