Risk stratification and relapse pattern in triple-negative breast cancer with pathological complete response after neoadjuvant treatment: the European GAMBIT real-world study
Abstract Pathologic complete response after neoadjuvant treatment is considered a surrogate of cure in triple-negative breast cancer, yet around 10% of patients still relapse. Whether baseline stromal tumor-infiltrating lymphocytes can stratify this residual risk is unknown.
Ciltacabtagene Autoleucel in High-Risk Smoldering Multiple Myeloma: The CAR-PRISM Phase 2 Trial
In a single-center phase 2 study (CAR-PRISM; NCT05767359), researchers from the Dana Farber Cancer Institute have shown that a single infusion of BCMA-directed CAR T cell therapy with ciltacabtagene autoleucel (cilta-cel) produced rapid, sustained minimal residual disease (MRD)-negative remissions in 20 patients with high-risk smoldering multiple myeloma (HR-SMM). Published in Nature Medicine, PRISM is the first prospective evaluation of CAR T cell therapy in a precursor plasma cell disorder.
GVHD Prophylaxis After Cord Blood Transplantation in Patients With High-risk AML: A Nationwide Japanese Cohort Study
In a nationwide, retrospective cohort study, researchers from St. Mary’s Hospital, Kurume, and a nationwide consortium of Japanese transplant centers have shown that mycophenolate mofetil (MMF) accelerates neutrophil engraftment compared to methotrexate (MTX) in cord blood transplantation (CBT). Unfortunately, this was accompanied by significantly higher rates of graft-versus-host disease (GVHD) and viral complications.

Clinical and histopathological characterization of metastatic lobular breast cancer: lessons learned from post-mortem tissue donation programs - npj Breast Cancer
|
Abstract While primary invasive lobular carcinoma (ILC) is well characterized, metastatic ILC remains understudied. Within the post-mortem tissue donation programs, UPTIDER (Belgium) and Hope for Others (USA), we first aimed to explore intra-patient heterogeneity of key prognostic and predictive markers (stromal tumor-infiltrating lymphocytes (sTIL), estrogen receptor (ER), progesterone receptor (PR), human epidermal growth factor receptor 2 (HER2) and KI67).

Expression of antibody-drug conjugate targets in post-mortem samples of breast cancer metastases and normal tissue - Nature Communications
Abstract While many Antibody Drug Conjugates (ADCs) are investigated in solid tumors, their target expression and heterogeneity is poorly documented in metastatic breast cancer. Here, we perform RNA sequencing to investigate expression of 72 ADC targets in 909 samples from 30 female patients from our post-mortem tissue donation program UPTIDER (NCT04531696). We report that the highest median expression in metastases is seen for FN1, MUC1, LAMP1, HER3 and SLC39A6.
ctDNA detectability and representativeness in seven body liquids from patients with metastatic breast cancer - Nature Communications
Abstract Liquid biopsies enable non-invasive monitoring and characterization of metastatic cancer, primarily through circulating tumor DNA (ctDNA) in blood. The representativeness of all metastatic sites in these liquid biopsies and the clinical relevance of other body fluids remain uncertain.
Palbociclib and endocrine therapy diminish adaptive anti-tumor immunity in early breast cancer: The NeoRHEA phase 2 study - Nature Communications
Abstract The NeoRHEA was a single-arm phase 2 study that included patients with estrogen receptor positive / human epidermal factor receptor 2 negative early breast cancer that received 4 cycles of neoadjuvant palbociclib and endocrine therapy. The primary outcome was baseline biomarkers of treatment resistance and secondary outcome was post-treatment transcriptional and epigenetic changes of tumor, immune and stromal cells.
