Caco-2-derived spheroids and organ-on-a-chip for food safety risk assessment: mechanisms of intestinal inflammation induced by 20 nm polystyrene nanoplastics
Abstract Microplastics released from food packaging have become an emerging food safety concern and may contribute to intestinal inflammation. This study employed Caco-2-derived spheroids combined with self-developed microfluidic organ-on-a-chip technology to systematically investigate the effects of 20 nm polystyrene nanoplastics (PS-NPs) on oxidative stress and functional homeostasis in the three-dimensional (3D) intestinal epithelial microenvironment.
Glioma-derived ULBP2 triggers microglial extracellular traps formation via PAD4 lactylation to drive autophagy-dependent malignant growth
Abstract The microglial extracellular traps(MiETs) were appeared in glioblastoma multiforme (GBM), but the underlying molecular mechanisms and functions were unknown. In this study, we aimed to explore the ability of GBM-derived UL16-binding protein 2 (ULBP2) on activating MiETs and manipulating microglia to support GBM development. Herein, ULBP2 was identified as a key upregulated protein in GBM and triggered MiETs formation. In turn, MiETs accelerated GBM malignant growth.
PINK1 agonist MTK458 attenuates cerebral edema by protecting blood-brain barrier integrity in intracerebral hemorrhage mice
Abstract Secondary blood-brain barrier (BBB) disruption and vasogenic edema drive neurological deterioration after intracerebral hemorrhage (ICH), but targeted therapies remain lacking. PTEN-induced putative kinase 1 (PINK1) is a neuroprotective kinase, yet its role in ICH-induced BBB damage is unclear. This study investigates whether MTK458, a novel PINK1 agonist, protects BBB integrity after ICH and elucidates mechanisms.

FAM96A functions as a novel pace controller for iron uptake to maintain iron homeostasis and erythropoiesis
Abstract Iron is indispensable for various biological processes, among which erythropoiesis is a privileged iron consumer to synthesize hemoglobin. FAM96A, also known as cytosolic iron-sulfur protein assembly 2 A (CIA2A), is a conserved protein involved in cytosolic iron-sulfur cluster assembly but also harboring a canonical signal peptide for secretion. Here, we unveil a previously unrecognized role of autocrine FAM96A in maintaining iron homeostasis and erythroid differentiation.
Immune checkpoint inhibitor therapy in advanced cancer: clinical association of irAEs type, inflammatory markers and efficacy
Introduction Over the past decade, immune checkpoint inhibitors (ICIs) have significantly enhanced outcomes and increased survival for patients with various advanced solid tumors through blocking key immune regulators such as PD-1/PD-L1 and CTLA-4 (1, 2). The finding has had a significant impact on the fields of cancer treatment. Although ICIs increase anti-tumor immunity, they can also disrupt immune tolerance, which can lead to immune-related adverse events (irAEs).

Fluid Correlation: A Novel Nonparametric Metric to Assess the Dynamic Association
Abstract The dynamic association between stochastic processes provides crucial information to science and industrial enterprises. Classic methods only provide a fixed and static metric that represents the global association of the stochastic processes. The metric that characterizes the dynamic association is still lacking. Developing such a metric is challenging since the temporal dependence of the stochastic processes could introduce an additional layer of complexity.
