Genetically encoded fluorescent reporters to visualize α-synuclein pathology in live brain
To determine whether α-Syn-6H-EGFP could be used to explore the pathological effects of α-Syn inclusions on transcription, we performed scRNA-seq on SNc dopaminergic neurons. Single neurons were collected using the patch-seq approach74 (Figure 5A). Neurons with α-Syn-6H-EGFP inclusions were identified by their filamentous structures and enhanced fluorescence (Figure S5A).
