Structural topology optimization of railway vehicle wheel damping plates considering acoustic-vibration interaction
Abstract A topology optimization method for railway wheel damping plates is proposed to suppress the acoustic radiation caused by structural vibration, with the acoustic–vibration interaction explicitly taken into account. A configuration optimization framework for damping wheels is constructed by combining this method with evolutionary structural optimization (ESO) and vehicle dynamics.
Integrated transcriptomic and metabolomic analyses identify key metabolites and genes in sepsis-associated acute lung injury
Abstract Introduction: Sepsis-associated acute lung injury (SALI) is a life-threatening pulmonary complication of sepsis with high mortality. This study aimed to prioritize serum metabolites and genes potentially involved in SALI by integrating multi-omics datasets. Methods: A rat SALI model treated with Xuanbai Chengqi Decoction (XCD) was subjected to untargeted serum metabolomics.
Intestinal Source Control of Lipid Metabolism by Enzyme-Probiotic Encapsulated, Spatiotemporal Crosslinked, and Small Intestine-Adhesive Hydrogel Microspheres
1 Introduction Lipids, serving as essential structural building blocks and energy sources for cells, tissues, and organs, play a pivotal role in sustaining normal physiological functions [1]. However, chronic high-fat diets frequently induce a cluster of metabolic abnormalities, including atherosclerosis, hepatic steatosis, insulin resistance, intestinal microecological imbalance, and obesity [2, 3].
High-fidelity bioassembly of organoids and spheroids using inertial droplet microfluidics for precision oncology and tumor microenvironment modeling
Abstract The translation of 3D multicellular systems into clinical applications has been constrained by the need to balance physiological relevance and scalability. Current biofabrication methods primarily depend on passive cell aggregation or capillary- and viscosity-limited segmentation, resulting in stochastic heterogeneity that limits high-throughput screening (HTS).
HIV-1 uncoating location dictates sites of integration
Abstract HIV-1 cores enter the nucleus and undergo capsid disassembly (uncoating) near their integration site. Although most viral cores are localized to nuclear speckles (NSs), the spatial relationship between the uncoating site and integration site remains unclear.

Critical Review on the Molecular Behavior of Surfactants at the Oil-Water Interface During Oil Displacement
1 Introduction Surfactants are amphiphilic molecules characterized by the presence of both hydrophilic and hydrophobic moieties (Figure 1), enabling them to significantly influence interfacial phenomena [1]. Initially discovered and employed in everyday products such as soaps and detergents, surfactants have since found broader applications across various industrial and scientific sectors [2].
Sensitive and unbiased genome-wide profiling of base-editor-induced off-target activity using CHANGE-seq-BE - Nature Biotechnology
Abstract Detection of the off-target effects of base editors is important for identifying their safety risks but current methods for understanding their global activities have limitations in terms of sensitivity or bias by computationally selecting a subset of sites for experimental analysis. We present CHANGE-seq-BE, a method to assess the guide RNA-dependent off-target profile of both adenine and cytosine base editors that is simultaneously sensitive and unbiased.
Nanostructure-mediated manipulation of light propagation has enabled the demonstration of advanced metamaterial absorbers that have improved the performance of optoelectronic and nanophotonic energy harvesting systems. In this work, MXene Ti3CNTx thin films are prepared and demonstrated as an addition to the plasmonic material database in the near-infrared (NIR) region.
Disrupting bile acid metabolism by suppressing Fxr causes hepatocellular carcinoma induced by YAP activation - Nature Communications
Abstract Disruption of bile acid (BA) metabolism causes various liver diseases including hepatocellular carcinoma (HCC). However, the underlying molecular mechanism remains elusive. Here, we report that BA metabolism is directly controlled by a repressor function of YAP, which induces cholestasis by altering BA levels and composition via inhibiting the transcription activity of Fxr, a key physiological BA sensor.

Characterization of tumor evolution by functional clonality and phylogenetics in hepatocellular carcinoma - Communications Biology
Abstract Hepatocellular carcinoma (HCC) is a molecularly heterogeneous solid malignancy, and its fitness may be shaped by how its tumor cells evolve. However, ability to monitor tumor cell evolution is hampered by the presence of numerous passenger mutations that do not provide any biological consequences.

Fertility-preserving myeloablative conditioning using single-dose CD117 antibody-drug conjugate in a rhesus gene therapy model - Nature Communications
Abstract Hematopoietic stem cell (HSC) gene therapy has curative potential; however, its use is limited by the morbidity and mortality associated with current chemotherapy-based conditioning. Targeted conditioning using antibody-drug conjugates (ADC) holds promise for reduced toxicity in HSC gene therapy. Here we test the ability of an antibody-drug conjugate targeting CD117 (CD117-ADC) to enable engraftment in a non-human primate lentiviral gene therapy model of hemoglobinopathies.

Lentivector cryptic splicing mediates increase in CD34+ clones expressing truncated HMGA2 in human X-linked severe combined immunodeficiency - Nature Communications
|
Abstract X-linked Severe Combined Immunodeficiency (SCID-X1) due to IL2RG mutations is potentially fatal in infancy where ‘emergency’ life-saving stem cell transplant may only achieve incomplete immune reconstitution following transplant. Salvage therapy SCID-X1 patients over 2 years old (NCT01306019) is a non-randomized, open-label, phase I/II clinical trial for administration of lentiviral-transduced autologous hematopoietic stem cells following busulfan (6 mg/kg total) conditioning.

Optimization of the optical particle counter for online particle measurement in pressure-changing natural gas
Abstract Online measurement of particulate matter in high-pressure natural gas is of great significance to the purification process and safe operation of long-distance pipelines. However, pressure changes in high-pressure natural gas pipelines are complicated. The optical particle counter (OPC) is affected by the change in the refractive index of natural gas, which causes an error of the measured particle size.

Optimization of the optical particle counter for online particle measurement in high-pressure gas
Abstract With the aim to achieve fast and accurate online measurement of particle size and concentration in different high-pressure gas, the application of an optical particle counter (OPC) is extended from atmospheric pressure to high-pressure conditions (more than 2 MPa). But with the increase of the gas pressure, the performance of the OPC is reduced, resulting in the error of the tested particle size.

CRISPR-Cas9 gene repair of hematopoietic stem cells from patients with X-linked chronic granulomatous disease
Targeted gene therapy has been hampered by the inability to correct mutations in stem cells that can reconstitute the immune system after transplant into patients. De Ravin et al. now report that CRISPR, a DNA editing technology, corrected blood stem cells from patients with an immunodeficiency disorder (chronic granulomatous disease) caused by mutations in NOX2.
