Rewriting CAR-T cell fate: CRISPR/Cas gene editing for solid tumor therapy
Abstract Although chimeric antigen receptor T (CAR-T) cell therapy has achieved remarkable success in hematological malignancies, its therapeutic efficacy in solid tumors remains limited by several challenges, including insufficient tumor infiltration, T cell exhaustion and the immunosuppressive tumor microenvironment (TME). CRISPR/Cas, a third-generation gene editing technology developed in recent years, is characterized by its simplicity and high efficiency.
Engineered Un1Cas12f1 for multiplex genome editing with enhanced activity and targeting scope - Nature Communications
Abstract The compact CRISPR-Cas12f system is promising for AAV-delivered gene therapy, but its application has been constrained by restrictive PAM recognition (e.g., TTTR) and suboptimal editing efficiency. Through bacterial library screening and mammalian cell validation, we engineer evoCas12f, an optimized variant incorporating five key mutations, that dramatically expands PAM recognition to NTNR/NYTR.

Keywords Klebsiella pneumoniae lung injury ethanol production neutrophil reactive oxygen species ROS Introduction Klebsiella pneumoniae (Kpn) mainly colonizes in the upper respiratory tract and intestinal tracts.1,2 We have previously reported that the commensal high-alcohol-producing Kpn (HiAlc Kpn) in the intestinal microbiota can produce high endogenous alcohol through 2,3-butanediol fermentation and might be one of the potential causative agents of endo-alcoholic fatty liver disease.3,4...
Targeting the immune privilege of tumor-initiating cells to enhance cancer immunotherapy
Tumor-initiating cells (TICs) represent a distinct cell population characterized by enhanced stemness and tumorigenic potential.1 They exhibit high plasticity and can arise from the conversion of tumor cells that initially lack TIC-like properties.2,3,4,5 Evidence indicates that TICs possess the ability to evade immunosurveillance.6,7,8,9 This ability is partly mediated by the expression of inhibitory checkpoint molecules, which allows TICs to impair the activity of cytotoxic cells and escape...

Joint Performance of a Continuous Glass Fiber/Polypropylene Composite
4. Results and Discussion This section presents the experimental results and compares the three connection techniques through simulation. 4.1. Adhesive and Mechanical Bonding The load–displacement curve of the adhesive bonding specimen is presented in Figure 6a. Figure 6b shows the fracture surface of the specimen after the experiments. During the linear elastic stage, the bonding strength between the adhesive and the laminate surpasses the internal stress of the adhesive layer.
Regulation on the electronic structure and surface wettability of Co9S8 electrocatalyst by nitrogen and phosphorous co-doping for efficient overall water splitting
Regulation on the electronic structure and surface wettability of Co9S8 electrocatalyst by nitrogen and phosphorous co-doping for efficient overall water splitting Pentlandite cobalt sulfide (Co9S8) has gained great attention as water splitting electrocatalyst on account of its intrinsic metallic conductivity and active sites. However, the formed robust sulfur-hydrogen (S-Hads) bonds on Co9S8’s surface significantly impede the atomic hydrogen desorption and suppress overall HER performance.

Elimination of GPR146-mediated antiviral function through IRF3/HES1-signaling pathway
As the most important host defense against viral infection, interferon (IFN) stimulates hundreds of antiviral genes (ISGs) that together establish an “antiviral state”. However, the antiviral function of most ISGs in viral infection still need further exploration. Here, we demonstrated that the expression of G-protein-coupled receptor 146 (GPR146) is highly increased by both IFN-? and IFN-? in a STAT1-dependent signaling pathway.
