Nonsense-mediated mRNA decay inhibition reshapes the cancer immunopeptidome
Keywords nonsense-mediated mRNA decay non-canonical neoantigens cryptic peptides cancer immunopeptidome SMG1 tumor immunogenicity cancer immunotherapy patient-derived fragments Introduction Effective anti-tumor immunity relies on the ability of T cells to detect cancer-associated peptides presented by the major histocompatibility complex (MHC) molecules and is shaped by both the quantity and quality of neoantigens.1,2 Among these, mutation-derived neoantigens (peptides derived from...

Lymphodepleting chemotherapy potentiates neoantigen-directed T cell therapy by enhancing antigen presentation

Results Utilizing our previously developed data-driven pipeline,26 we prioritized candidate neoantigens from The Cancer Genome Atlas (TCGA) lung cancer dataset and identified the KRAS.G12V-HLA-A∗03:01 combination as a leading candidate. This HLA-mutation combination is found in approximately 94,000 patients worldwide (Figure 1A; Table S1).

Cancer cell-derived arginine fuels polyamine biosynthesis in tumor-associated macrophages to promote immune evasion
The tumor microenvironment (TME) represents a complex ecosystem harboring cancer cells, stromal cells, immune cells, and a diverse array of signaling molecules.1 This complicated network orchestrates crucial aspects of tumor biology, including progression, metastasis, immune evasion, and response to therapy.2,3,4 Central to TME dynamics is its immunosuppressive milieu, often driven by metabolic reprogramming of resident cells.3,5 Cancer cells undergo distinct metabolic alterations,...
