A comprehensive transcriptome and metabolome dataset for Erigeron breviscapus (Asteraceae) response to different light intensities
Abstract Erigeron breviscapus is a widely utilized medicinal herb in traditional Chinese medicine, characterized by high annual demand and notable pharmacological activities. Light intensity is a critical environmental factor influencing secondary metabolism in medicinal plants. However, the effects of light intensity on the accumulation of active ingredients in E. breviscapus remain largely unexplored. In this study, we examined the transcriptomic and metabolomic responses of E.
Exploring the technical possibilities of metabolomic versus genomic tests for diagnosis of inborn errors of metabolism in NICU
Abstract Inborn errors of metabolism (IEMs) exhibit substantial biochemical and genetic heterogeneity and cause severe, life-threatening conditions in neonatal intensive care unit (NICU) newborns with high morbidity and mortality. Early diagnosis via high-throughput technology is crucial for optimizing neonatal IEM prognosis.
PRPF19 drives hepatic stellate cell activation and liver fibrosis via PKM2-mediated glycolytic reprogramming
Abstract Activation of hepatic stellate cells (HSCs) is a pivotal event driving fibrogenic remodeling of the injured liver. However, the upstream regulators that integrate post-translational signaling with metabolic reprogramming during HSC activation remain poorly defined. Here, we identify pre-mRNA processing factor 19 (PRPF19) as a ubiquitination-linked metabolic regulator that promotes HSC activation.

Self-disclosure significantly predicted PPR (β = 0.510, p < .001).
Five New Iridoids From the Leaves of Viburnum macrocephalum f . macrocephalum and Their Free Radical Scavenging Activity
1 Introduction Iridoids constitute a structurally diverse and biologically significant class of bicyclic monoterpenes derived biosynthetically from iridodial. The first iridoid, iridomyrmecin, was identified in 1925 from the ant Iridomyrmex humilis. Iridoids are universally characterized by a cyclopenta[c]pyran skeleton [1]. Recently, iridoids have attracted increasing attention due to their antimicrobial [2], antiviral [3], anti-inflammatory [4], and antioxidant activities [5].

Type I interferon in psoriasis: mechanisms and clinical implications
Abstract Psoriasis is a chronic, recurrent, immune-mediated inflammatory skin disease in which the TNF/IL-23/IL-17 axis plays a central role in the maintenance of mature plaques. However, this classical framework does not fully explain the early immune events triggered by skin injury, barrier disruption, infection, and the release of self-nucleic acids. It also does not fully explain selected clinical phenotypes.

Rbm5 sustains leukemia stem cells through a Myc-driven transcriptional circuitry
Abstract Acute myeloid leukemia (AML) represents a type of malignant hematological disease that is usually caused by the dysregulated developmental program of leukemia stem cells (LSCs). Here, we report that an unappreciated RNA-binding protein, Rbm5, selectively promotes murine leukemogenesis, maintains LSC self-renewal in vivo, and is dispensable for normal hematopoiesis.
Ultra-flexible optoelectronic stimulator converts tissue-attenuated weak light into electrical signals for cardiac remodeling
Abstract Precise spatiotemporal resolution and wireless multi-site modulation of excitable tissues via optoelectronics offers transformative potential for bioelectronic medicine, yet clinical translation is hindered by the rigidity of conventional silicon-based devices and their reduced performance under tissue-attenuated illumination, where the weak light reaching implants often fails to generate sufficient stimulation voltage.
Efficient in vivo cytosine base editing using virus-like particles with uracil DNA glycosylase inhibition
Abstract Virus-like particles (VLPs) are promising for delivering genome editors, yet the in vivo in vivo efficacy of VLP-mediated cytosine base editing remains limited. Here we identified insufficient inhibition of uracil DNA glycosylases as the underlying mechanism of low cytosine base editor (CBE) editing efficiencies in vivo.
Immune cell-specific genetic architecture of Alzheimer’s disease revealed by multi-omics analysis for therapeutic target discovery and prioritization
Abstract Alzheimer’s disease (AD) is a multifactorial neurodegenerative condition in which accumulating genetic and molecular evidence implicates dysregulation of peripheral immune processes in disease pathogenesis. Nevertheless, the contribution of distinct peripheral immune cell subsets and associated gene regulatory landscapes to AD risk remains incompletely defined.

TopoFlow: topography-aware pollutant Flow learning for high-resolution air quality prediction
Abstract We propose TopoFlow (Topography-aware pollutant Flow learning), a physics-guided neural network for efficient, high-resolution air quality prediction. To explicitly embed physical processes into the learning framework, we identify two critical factors governing pollutant dynamics: topography and wind direction. Complex terrain can channel, block and trap pollutants, while wind acts as a primary driver of their transport and dispersion.
Sequential CD19/CD22 CAR-T cell therapy following ASCT shows improved efficacy versus CAR-T alone in relapsed/refractory large B-cell lymphoma
To the editor: Patients with relapsed/refractory (R/R) large B-cell lymphoma (LBCL) continue to face a poor prognosis, with autologous stem cell transplantation (ASCT) after salvage chemotherapy curing only ≤30% of eligible patients [1, 2]. While CD19-directed chimeric antigen receptor T (CAR-T) cell therapy has revolutionized treatment with 50–80% response rates, 40–70% of patients still relapse due to antigen escape or other resistance mechanisms [3, 4].
Clinical application of base editing for treating β-thalassaemia
Abstract β-Thalassaemia is caused by reduced or absent production of β-haemoglobin1,2,3,4. Previously, we performed laboratory-scale electroporation of CD34+ haematopoietic stem and progenitor cells from patients with β-thalassaemia using a transformer base editor5,6. The aim was to target the binding motif of the transcription repressor BCL11A in the HBG1 and HBG2 promoters7 to reactivate fetal haemoglobin (HbF) production.
Alternative donors, particularly haploidentical donors, are safe and effective for elderly patients with adverse-risk AML: a multicenter TROPHY study
Dear editor, Adverse-risk acute myeloid leukemia (AML) is one of the most life-threatening hematologic malignancies in adults [1], and allogeneic stem cell transplantation (allo-HSCT) is the standard consolidation therapy after achieving complete remission (CR) [2]. However, the incidence of AML increases with advancing age, with a median age at diagnosis ranging from 65 to 72 years [3].

International testing and refinement of AI algorithms predicting acute leukemia subtypes from routine laboratory data
|
Abstract Despite advances for patients with acute leukemia health disparities limit access to diagnosis and treatment. Artificial Intelligence (AI) approaches may address some disparities. We retrospectively assemble a diverse, international cohort of 6206 leukemia patients from 20 centers to test an AI tool designed to support leukemia diagnosis using standard laboratory results. Executing the pretrained algorithm results in varying accuracy metrics.

Lessons from Pyre that Shaped Pyrefly
Pyrefly is a next-generation Python type checker and language server, designed to be extremely fast and featuring advanced refactoring and type inference capabilities. This isn’t the Pyrefly team’s first time building a type checker for Python: Pyrefly is a successor to Pyre, the previous type checker our team developed. A lot of Pyrefly’s design comes directly from our experience with Pyre. Some things worked well at scale, while other things were harder to live with day-to-day.

HMGCS2-dependent β-OHB/H3K9bhb ameliorates synaptic plasticity and cognition in Alzheimer’s disease - Experimental & Molecular Medicine
Abstract Ketogenic diet (KD) can significantly ameliorate cognition in Alzheimer’s disease (AD), but the specific mechanism is not clear. Histone3-lysine9-β-hydroxybutyrylation (H3k9bhb), a novel histone modification mark induced by ketogenesis-generated β-hydroxybutyrate (β-OHB), may be involved in the prevention and treatment of AD. Here we report that β-OHB and H3K9bhb were reduced in the hippocampus of triple transgenic AD male mice (3xTg-AD) mice.
Python Type Checker Comparison: Empty Container Inference
Empty containers like [] and {} are everywhere in Python. It's super common to see functions start by creating an empty container, filling it up, and then returning the result. Take this, for example: defmy_func(ys:dict[str,int]): x ={} for k, v in ys.items(): if some_condition(k): x.setdefault("group0",[]).append((k, v)) else: x.setdefault("group1",[]).append((k, v)) return x This seemingly innocent coding pattern poses an interesting challenge for Python type checkers.
Fine‐Scale Estimation of Urban Biogenic CO2 Fluxes: A Novel Framework Integrating Multiple Versions of Vegetation Photosynthesis and Respiration Models and In Situ Measurements
1 Introduction As the main driver of global warming, the growth rate of anthropogenic carbon dioxide (CO2) emissions has become a pressing global issue (Meyer et al., 2014). Although only a fraction of fossil fuel CO2 emissions physically occur within city boundaries, urban areas are estimated to account for up to 70% of global CO2 emissions when indirect emissions from energy use and supply chains are considered (Seto et al., 2014; Turnbull et al., 2019; Velasco & Roth, 2010).
