Functional evaluation and clinical classification of BRCA2 variants - Nature
Abstract Germline BRCA2 loss-of function variants, which can be identified through clinical genetic testing, predispose to several cancers1,2,3,4,5. However, variants of uncertain significance limit the clinical utility of test results. Thus, there is a need for functional characterization and clinical classification of all BRCA2 variants to facilitate the clinical management of individuals with these variants.
Predictors of response to CDK4/6i retrial after prior CDK4/6i failure in ER+ metastatic breast cancer - npj Breast Cancer
Abstract After disease progression on endocrine therapy (ET) plus a CDK4/6 inhibitor, there is no standardized sequence for subsequent treatment lines for estrogen receptor positive (ER+) metastatic breast cancer (MBC). CDK4/6i retrial as a treatment strategy is commonplace in modern clinical practice; however, the available prospective data investigating this strategy have had inconclusive results.
Dose dense doxorubicin plus cyclophosphamide in a modified KEYNOTE522 regimen for triple negative breast cancer - npj Breast Cancer
Abstract The KEYNOTE-522 (KN522) regimen for neoadjuvant treatment of triple negative breast cancer (TNBC) utilized q3week dosing for doxorubicin plus cyclophosphamide (AC); however, dose-dense AC (ddAC) has demonstrated superior overall survival (OS) compared to q3week AC in anthracycline and taxane-based regimens.

Biospecimen Repositories in Low- and Middle-Income Countries: Insights From an American University of Beirut and Memorial Sloan Kettering Collaboration | JCO Global Oncology
Cancer is a global disease with geographic and ethnic differences contributed by different genetic make-up, background, and environmental exposures.1 The genomic complexity of individual tumors can now be characterized through a detailed analysis of the tissue acquired for the purposes of standard pathologic and cytologic evaluation.

The AUB-MSK biospecimen repository has provided a unique resource for interrogation. Patient participation rate was high, and analyses of those who elected not to consent (29%) provide important insights into educational need and the local and cultural awareness and norms. Of 850 patients approached, 704 (70.8%) elected to consent and 293 (29.5%) declined participation. The number of declined consents led to an amendment permitting the documentation of reasons for same.
On Impact
If Eliot is right and April is the cruelest month, we had all better armor up on March 31, because January has been a beast. We should take a mulligan and drop the ball again on February 1rst. But, despite all of the disappointments and challenges brought by Omicron, some things have continued on, perennial as the grass. The sun rises and sets, the tides ebb and flow, and here in academic medicine, we complete annual reviews.

Golden opportunity missed due to collapse of European Super League
Watching the shambolic launch of a party to which Newcastle United were not invited, has been hilarious. The thought of the Premier League’s ‘Super Six’ leaving the top tier actually filled me with joy. These are six of the clubs I dislike most in the world, so I would have been more than happy for them to vanish into a show pony content fest, where they played with each other in a European Super League behind a paywall for all eternity.
New Premier League rules coming in...(***Satire)
Talksport’s Jim White recently asked if watching football makes lockdown more bearable. My response is no, it has made lockdown more miserable. That’s because I’m a Newcastle fan. However, watching a lot of football has made me realise that “The Beautiful Game” could be improved in many constructive ways to make it a better television spectacle.
Dear Santa, as a Newcastle United fan I'm not asking for much, but...
Dear Santa, I hope you and the reindeer are well. I was worried you might not be able to come into Tier 4 with my presents but I have written to the Prime Minister and he has promised me you can travel as long as you all wear masks and disinfect in Norway so that’s OK. And a Prime Minister always honours their promises… I suppose I don’t really want many presents this year.

Cancer therapy shapes the fitness landscape of clonal hematopoiesis
Abstract Acquired mutations are pervasive across normal tissues. However, understanding of the processes that drive transformation of certain clones to cancer is limited. Here we study this phenomenon in the context of clonal hematopoiesis (CH) and the development of therapy-related myeloid neoplasms (tMNs). We find that mutations are selected differentially based on exposures.

A long season for Newcastle United - Few manifestations of the Magic Triangle but plenty rubbish
Here is a very basic theory of bad football, especially with regard to Newcastle United. In a typical game, how do we know that a team is really poor and has little ambition to win? Well, we watch how they perform and conclude from the evidence before us that they were rubbish, from back to front and all over.

Broad Application of Multigene Panel Testing for Breast Cancer Susceptibility
Testing for inherited breast cancer (BC) susceptibility is an integral part of the practice of oncology. The initial purpose of testing was for cancer risk assessment to inform decisions about enhanced surveillance or risk-reducing surgery.

| Management of Women With Breast Cancer and Pathogenic Variants in Genes Other Than BRCA1 or BRCA2. Our ability to identify pathogenic variants predisposing towards breast cancer is increasing. However, guidance on how to incorporate variants beyond high penetrance cancer syndrome genes into clinical decision making is not well established. The original report highlighted in this discussion looks at a novel approach using polygenic risk score to further risk stratify these patients.
When push came to shove, Newcastle had these 4 options at Crystal Palace | NUFC The Mag
One of the more worrying aspects of the Newcastle United performance against Crystal Palace, was the sudden evaporation of any belief, determination and positivity to build on what was looking to be a promising performance. When Palace stepped up their game, Newcastle United did not respond. There didn’t seem to be any enthusiasm to match Palace’s sudden burst of energy.
Olaparib for Metastatic Breast Cancer in Patients with a Germline BRCA Mutation
The oral PARP inhibitor olaparib is approved for the treatment of patients with recurrent ovarian cancer and a BRCA mutation, and it has been shown to provide clinically meaningful benefit among such patients. 10,11 Olaparib has also been shown to have promising activity in patients with metastatic breast cancer and a germline BRCA mutation.
Commodity Hedging - the advent of a new paradigm
Commodity Hedging - the advent of a new paradigm The events of 2008 have shaken up the risk management approach of multinational corporations (MNCs) and have given renewed emphasis to the way they handle a range of financial risks, such as commodity purchasing, liquidity, FX and funding. These issues have come to the fore for heavy users of commodities following the rollercoaster ride in the prices of oil, metals, and soft commodities.

Reliable Detection of Mismatch Repair Deficiency in Colorectal Cancers Using Mutational Load in Next-Generation Sequencing Panels
|
Purpose Tumor screening for Lynch syndrome is recommended in all or most patients with colorectal cancer (CRC). In metastatic CRC, sequencing of RAS/BRAF is necessary to guide clinical management. We hypothesized that a next-generation sequencing (NGS) panel that identifies RAS/BRAF and other actionable mutations could also reliably identify tumors with DNA mismatch repair protein deficiency (MMR-D) on the basis of increased mutational load.

Behind the numbers: Researching Syria's killed journalists
This year, CPJ researchers confirmed that at least 29 journalists died while covering the Syrian conflict. How did we arrive at that number? Our research begins with the collection of possible cases to investigate. By closely following reports from news outlets, local journalist associations, press freedom groups, social media accounts, and human rights organizations, CPJ compiled a list of 159 names of potential cases where journalists died covering the conflict this year.
