Genetic encoding and mutagenesis of RNA droplet phenotypes
Abstract RNA droplets assembled from cotranscriptionally folded nanostructures have recently emerged as a promising platform for constructing protocells and minimal synthetic cell models. A custom-designed DNA template encodes an RNA nanostar that self-assembles into droplets by kissing-loop interactions. Here we show that such RNA droplets encapsulate their DNA template, creating a genotype–phenotype link.
Abundant CD8 + tumor-infiltrating lymphocytes in glioblastoma harbor distinct exhaustion signatures in potentially reactive subsets

Abstract The effectiveness of immunotherapy against glioblastoma (GBM) remains limited, suggesting that antigen-specific tumor-infiltrating lymphocytes (TILs) are rare. This study aimed to identify tumor-specific T cells in GBM as the basis for antigen-specific immunotherapy. Tumor specimens were collected from 56 patients newly diagnosed with IDH-wildtype GBM.

Brain barriers at the crossroads of glioma immune surveillance and immunotherapy response
Abstract Immunotherapies have substantially advanced cancer treatment; however, their efficacy in gliomas remains limited. This observation cannot be fully explained by tumour-intrinsic factors and may rather be linked to the distinct relationship between the central nervous system (CNS) and the immune system, commonly described as CNS immune privilege.

IDH1 -mutant vaccine in newly diagnosed astrocytoma: final analysis of the multicenter, single-arm, open-label, first-in-human phase 1 NOA16 trial
|
Abstract The clonal glioma driver mutation IDH1R132H gives rise to a major histocompatibility class II-restricted neoepitope. A multicenter, first-in-human phase 1 trial met its prespecified primary endpoints by demonstrating safety and immunogenicity of an IDH1-R132H peptide vaccine (IDH1-vac) integrated into standard of care in 33 participants with newly diagnosed grade III and IV (World Health Organization classification 2007) IDH1-R132H+ astrocytomas (NOA16).

Regulation of immune checkpoint molecules in cancer immune evasion and therapy
Abstract Immune checkpoint molecules are essential regulators of immune homeostasis, maintaining the balance between activation and tolerance. In cancer, tumours exploit checkpoint pathways to suppress antitumour immunity and promote progression.
Type I interferon drives T cell responses to amyloid beta in the central nervous system
Abstract Amyloid beta (Aβ) plaque deposition in the central nervous system (CNS) is a hallmark of Alzheimer’s disease (AD) and cerebral amyloid angiopathy (CAA), triggering robust innate immune responses. However, the role of the adaptive immune system remains less well understood. Here we show the immune microenvironment dynamics in APP23 transgenic (APP23-tg) mice modelling CNS amyloid pathology, using single-cell transcriptomics.

A microenvironment-determined risk continuum refines subtyping in meningioma and reveals determinants of machine learning-based tumor classification - Nature Genetics
|
Abstract Classification of tumors in neuro-oncology today relies on molecular patterns (mostly DNA methylation) and their machine learning-supported interpretation. Understanding the process of algorithmic interpretation is essential for safe application in clinical routine. This is paradigmatically true for the most common primary intracranial tumor in adults, meningioma.

Immunosuppressive mechanisms and therapeutic interventions shaping glioblastoma immunity - Nature Cancer
Abstract Plasticity is a hallmark of aggressive tumors, including glioblastoma (GBM), enabling tumor cells and the tumor microenvironment (TME) to adapt to diverse niches and evade treatment. Here, we discuss how innate and adaptive immune players cooperate in time and space to create an immunosuppressive TME that supports GBM growth and confers resistance to conventional treatments and immunotherapies.

Molecularly matched targeted therapies plus radiotherapy in glioblastoma: the phase 1/2a N2M2 umbrella trial - Nature Medicine
Abstract Advances in molecular understanding and diagnostic precision of glioblastoma enable the identification of key genetic alterations in a timely manner and, in principle, allow treatments with targeted compounds based on molecular markers. Here we report the results of the phase 1/2 umbrella trial NCT Neuro Master Match (N2M2), which evaluated targeted treatments in 228 patients with newly diagnosed glioblastoma without O6-methylguanine DNA-methyltransferase promoter hypermethylation.
Vaccine-induced T cell receptor T cell therapy targeting a glioblastoma stemness antigen - Nature Communications
Abstract T cell receptor-engineered T cells (TCR-T) could be advantageous in glioblastoma by allowing safe and ubiquitous targeting of the glioblastoma-derived peptidome. Protein tyrosine phosphatase receptor type Z1 (PTPRZ1), is a clinically targetable glioblastoma antigen associated with glioblastoma cell stemness. Here, we identify a therapeutic HLA-A*02-restricted PTPRZ1-reactive TCR retrieved from a vaccinated glioblastoma patient.
Bottom-up synthetic immunology - Nature Nanotechnology
Abstract Infectious diseases and cancer evade immune surveillance using similar mechanisms. Targeting immune mechanisms using common strategies thus represents a promising avenue to improve prevention and treatment. Synthetic immunology can provide such strategies by applying engineering principles from synthetic biology to immunology. Synthetic biologists engineer cells by top-down genetic manipulation or bottom-up assembly from nanoscale building blocks.
Cranioencephalic functional lymphoid units in glioblastoma - Nature Medicine

L-RNA aptamer-based CXCL12 inhibition combined with radiotherapy in newly-diagnosed glioblastoma: dose escalation of the phase I/II GLORIA trial - Nature Communications
Abstract The chemokine CXCL12 promotes glioblastoma (GBM) recurrence after radiotherapy (RT) by facilitating vasculogenesis. Here we report outcomes of the dose-escalation part of GLORIA (NCT04121455), a phase I/II trial combining RT and the CXCL12-neutralizing aptamer olaptesed pegol (NOX-A12; 200/400/600 mg per week) in patients with incompletely resected, newly-diagnosed GBM lacking MGMT methylation.
Dietary L-Tryptophan consumption determines the number of colonic regulatory T cells and susceptibility to colitis via GPR15 - Nature Communications
Abstract Environmental factors are the major contributor to the onset of immunological disorders such as ulcerative colitis. However, their identities remain unclear. Here, we discover that the amount of consumed L-Tryptophan (L-Trp), a ubiquitous dietary component, determines the transcription level of the colonic T cell homing receptor, GPR15, hence affecting the number of colonic FOXP3+ regulatory T (Treg) cells and local immune homeostasis.

A H3K27M-targeted vaccine in adults with diffuse midline glioma - Nature Medicine
Abstract Substitution of lysine 27 to methionine in histone H3 (H3K27M) defines an aggressive subtype of diffuse glioma. Previous studies have shown that a H3K27M-specific long peptide vaccine (H3K27M-vac) induces mutation-specific immune responses that control H3K27M+ tumors in major histocompatibility complex-humanized mice. Here we describe a first-in-human treatment with H3K27M-vac of eight adult patients with progressive H3K27M+ diffuse midline glioma on a compassionate use basis.
Short‐term brain atrophy evolution after initiation of immunotherapy in a real‐world multiple sclerosis cohort
Abstract Background and Purpose In multiple sclerosis (MS), brain atrophy measurements have emerged as an important biomarker reflecting neurodegeneration and disability progression. However, due to several potential confounders, investigation of brain atrophy in clinical routine and even in controlled clinical studies can be challenging.
Correcting the drug development paradigm for glioblastoma requires serial tissue sampling - Nature Medicine
A breakthrough in drug discovery for glioblastoma requires serial collection of tissue from the central nervous system via window of opportunity trials Therapeutic drug development in glioblastoma has remained static despite numerous clinical trials, advances in tumor biology, and substantial public interest. Since 2005, more than 1,250 interventional glioblastoma trials have been registered on ClinicalTrials.gov. Of these, 1,100 are phase 1 or 2 trials, most of which are dose-escalation studies.
Spatial probabilistic mapping of metabolite ensembles in mass spectrometry imaging - Nature Communications
Abstract Mass spectrometry imaging vows to enable simultaneous spatially resolved investigation of hundreds of metabolites in tissues, but it primarily relies on traditional ion images for non-data-driven metabolite visualization and analysis. The rendering and interpretation of ion images neither considers nonlinearities in the resolving power of mass spectrometers nor does it yet evaluate the statistical significance of differential spatial metabolite abundance.

Consistency of the “central vein sign” in chronic multiple sclerosis lesions
Introduction In multiple sclerosis (MS), there has been an increasing interest in the central vein sign (CVS) on advanced magnetic resonance imaging (MRI) techniques such as susceptibility-weighted imaging (SWI) (Sati et al., 2016).
Consistency of the “central vein sign” in chronic multiple sclerosis lesions
Highlights • We investigated the consistency of the central vein sign (CVS) in 183 chronic multiple sclerosis (MS) lesions • The CVS was consistent in the majority of chronic lesions (81%) • The CVS represents a stable biomarker that could increase the specificity of MRI in MS.
Impaired semantic memory during acute transient global amnesia
Abstract As a clinical model of hippocampal dysfunction, transient global amnesia (TGA) causes reversible memory disturbance. While episodic memory deficits in TGA patients have been extensively described, data regarding semantic memory involvement are sparse and contradictory. We report impaired semantic fluency performance in 16 patients with hippocampal lesions on MRI during acute TGA compared to their performance one day later and to that of 20 healthy subjects.
A vaccine targeting mutant IDH1 in newly diagnosed glioma
Abstract Mutated isocitrate dehydrogenase 1 (IDH1) defines a molecularly distinct subtype of diffuse glioma1,2,3. The most common IDH1 mutation in gliomas affects codon 132 and encodes IDH1(R132H), which harbours a shared clonal neoepitope that is presented on major histocompatibility complex (MHC) class II4,5.
The relevance of T helper cells that are induced by a mutant IDH1 vaccine in newly diagnosed astrocytoma
Published Mar 24, 2021 In the most recent edition of Nature, we are excited to report on the successful completion of our first-in-human phase I clinical trial (NOA16, NCT02454634) targeting a shared clonal driver mutation using a peptide vaccine in diffusely infiltrating low grade and anaplastic astrocytoma, a type of malignant brain tumor affecting remarkably young patients.
Tryptophan metabolism as a common therapeutic target in cancer, neurodegeneration and beyond
Abstract l-Tryptophan (Trp) metabolism through the kynurenine pathway (KP) is involved in the regulation of immunity, neuronal function and intestinal homeostasis. Imbalances in Trp metabolism in disorders ranging from cancer to neurodegenerative disease have stimulated interest in therapeutically targeting the KP, particularly the main rate-limiting enzymes indoleamine-2,3-dioxygenase 1 (IDO1), IDO2 and tryptophan-2,3-dioxygenase (TDO) as well as kynurenine monooxygenase (KMO).
T cells engineered to home in on brain cancer
Therapies that activate immune cells called T cells to target tumours are an efficient way to combat many types of cancer1. But an aggressive brain cancer called glioblastoma has proved a particular challenge for immunotherapies2. The blood–brain barrier protects the brain against immune-cell infiltration to prevent the potentially life-threatening effects of brain inflammation.
Novel, improved grading system(s) for IDH-mutant astrocytic gliomas
