TIM3-targeted delivery of venetoclax overcomes drug resistance and reinvigorates NK cell activity in acute myeloid leukemia
Abstract Acute myeloid leukemia (AML) remains challenging, especially for older or unfit individuals who develop resistance to venetoclax (VEN)-based regimens. Although TIM3 represents a promising AML target, clinical blockade has yielded suboptimal outcomes. Here, we develop VINCENT (VEN-integrated natural killer cell engager targeting TIM3), a therapeutic platform combining anti-CD16 and anti-TIM3 antibodies with nanoformulated VEN.

CircFN1 modulates IL-1β-induced osteoarthritis through the miR-19b-3p/PTEN signaling axis
Abstract Osteoarthritis (OA) is a degenerative joint disease marked by cartilage breakdown, chondrocyte apoptosis, and extracellular matrix (ECM) degradation. Non-coding RNAs, particularly circular RNAs (circRNAs), are increasingly recognized as key regulators of OA pathogenesis. This study reveals that circFN1 (hsa_circ_0058152) may act as a competing endogenous RNA, sponging miR-19b-3p and suppressing PTEN expression.

ACAT1 regulates tertiary lymphoid structures: A target for enhancing immunotherapy in non-small cell lung cancer
Abstract Non–small cell lung cancer (NSCLC), the most common type of lung cancer, remains a leading cause of cancer-related mortality worldwide. Immune checkpoint inhibitors (ICIs) have emerged as a promising therapy for NSCLC but only benefit a subset of patients. In this issue of the JCI, Jiao et al. revealed that acetyl-CoA acetyltransferase 1 (ACAT1) limited the efficacy of ICIs in NSCLC by impeding tertiary lymphoid structures (TLS) in the tumor microenvironment (TME).
